To determine the role of oxygen radicals in the killing of Mycobacterium tuberculosis by neutrophils, the effects of free-radical inhibitors and enzymes, catalase, superoxide dismutase, taurine, deferoxamine, and histidine were evaluated. Changes in the viability of M. tuberculosis were determined by agar plate colony counts and a radiometric assay. No impairment in killing was seen with any of the inhibitors or enzymes. Patients with chronic granulomatous disease (CGD) have a defect in the NADPH oxidase pathway, causing their neutrophils to be unable to generate oxygen radicals. If these radi…

A 5-year-old neutered male Siamese cat was examined by a veterinarian because of a recent decrease in appetite and a large lymph node in the left mandibular area. Clinical findings included fever, icterus, leukopenia, and progressive anemia. Despite various treatments, the cat died approximately 3 weeks after initial examination. The main necropsy findings included necrotizing and granulomatous lymphadenitis of the left mandibular lymph node, multifocal necrotizing hepatitis, and interstitial pneumonia. Acid-fast bacilli were detected in lesions of the mandibular lymph node, liver, lung, splee

Tuberculosis is a respiratory infectious disease that persists worldwide, largely due to the robustness and ease of dissemination of its causative agent, Mycobacterium tuberculosis. To mitigate the spread of infection, various measures have been developed to inactivate the bacteria in its aerosol form. Advanced photohydrolysis technology (APHT) was previously reported to inactivate both respiratory viral and bacterial pathogens; however, its efficacy against M. tuberculosis has yet to be evaluated. In this study, we assessed the ability of APHT to inactivate aerosolized M. tuberculosis. The ba…

Mycobacterium tuberculosis is tolerant to many antibiotics, leading to impaired antibiotic killing. Using CRISPR interference (CRISPRi) transcriptional knockdowns, we generated a panel of metabolically compromised strains to identify tolerance pathways for pursuing in therapeutic development. Disrupting the regulation of intracellular iron storage, amino acid biosynthesis and redox defence mechanisms potentiated the lethality of multiple drugs and translated to infected THP-1 macrophages. This work reinforces the role of metabolism as a major contributor to drug tolerance in M. tuberculosis.

Tuberculosis remains one of the leading major global health challenges, driven by the emergence of multidrug-resistant and extensively drug-resistant bacterial strains. Resistant strains complicate treatment, which often requires prolonged use of toxic second- and third-line drugs. Protein phosphorylation plays critical roles in Mycobacterium tuberculosis, with serine/threonine kinases PknA, PknB, and PknG being essential for survival, virulence, and persistence. In this study, we screened an in-house kinase inhibitor library to identify compounds targeting these kinases. Four structurally div…

Mycobacterium tuberculosis (Mtb) causes more deaths annually than any other pathogen, yet an effective vaccine remains elusive. IFN-γ-producing CD4+ T cells are necessary but insufficient for protection against infection. In humans, the development of IL-17A producing Th17 T cells correlates with protection, however not all individuals develop a Th17 response. In mice, experimental vaccines can elicit protective Th17 cells, yet Th17s are rare in primary infection. Why Mtb fails to consistently elicit Th17s is unknown. Here, we identify factors suppressing Th17 responses during primary infectio…

The identification of novel agents with mechanisms of action distinct from those currently utilized in tuberculosis treatment remains a significant challenge. The mycobacterial protein MmpL3 has emerged as a promising drug target due to its essential role in the synthesis of the cell wall of Mycobacterium tuberculosis. We previously identified novel thienopyrimidine amides(TPAs) with good anti-tubercular activity. We profiled a subset of TPAs, determining activity against intracellular bacteria and bactericidal activity against replicating bacteria. We ran assays to determine the mode of actio…

Mycobacterium tuberculosis (Mtb) is among the most successful bacterial pathogens, with multidrug-resistant strains posing significant challenges to global tuberculosis control. Traditional single-genome analyses, while essential for identifying strain-specific mutations, are limited in capturing the full spectrum of genetic diversity related to virulence, drug susceptibility, and transmission dynamics. Pangenomics examines the complete gene repertoire across all sequenced representatives of a species and addresses these limitations by enabling comprehensive, species-wide assessments of geneti…

Heterogeneity during Mycobacterium tuberculosis (Mtb) infection greatly impacts disease outcome and complicates treatment. This heterogeneity encompasses many facets, spanning local differences in the host immune response to Mtb and the environment experienced by the bacterium, to nonuniformity in Mtb replication state. All of these facets are interlinked and each can affect Mtb susceptibility to antibiotic treatment. In-depth spatiotemporal understanding of Mtb-host interactions is thus critical to both fundamental comprehension of Mtb infection biology and for the development of effective th…

MmpL3 is a promising new target for antitubercular drugs, but the microbiological properties of MmpL3 inhibitors are not fully understood. We compared the activity and mode of action of 11 structurally diverse compound series that target MmpL3. We confirmed the activity was via MmpL3 using strains with differential expression of MmpL3. MmpL3 inhibitors had potent activity against replicating M. tuberculosis, with increased activity against intramacrophage bacilli and were rapidly bactericidal. MmpL3 inhibition induced cell wall stress concomitantly with a boost in the ATP levels in M. tubercul

Tuberculosis of the cerebral nervous system is a diagnostic and therapeutic emergency that can lead to death and sometimes disabling sequelae. It can be complicated by stroke in the context of vasculitis. We report a pediatric case. We report the case of a 13-year-old child who presented with a sudden onset of altered consciousness in the context of fever. He had been experiencing unexamined cervical lymphadenopathy and a cough for a year. On day 4 of hospitalisation, he developed a right pyramidal syndrome. Cerebral magnetic resonance imaging revealed a deep middle cerebral artery infarction.

In Mycobacterium tuberculosis (Mtb), persisters are genotypically drug-sensitive bacteria that nonetheless survive antibiotic treatment. Persisters represent a significant challenge to shortening TB treatment and preventing relapse, underscoring the need for new therapeutic strategies. In this study, we screened 2,336 FDA-approved compounds to identify agents that enhance the sterilizing activity of standard anti-TB drugs and prevent the regrowth of persisters. Netupitant (NTP), an FDA-approved antiemetic, emerged as a promising candidate with bacteriostatic activity on its own. However, in co…
