Mycotoxin toxicosis has been implicated in the etiopathogenesis of Keshan disease (KD), an endemic cardiomyopathy prevailing in some regions of China. Butenolide (4-acetamido-4-hydroxy-2-butenoic acid gamma-lactone, CAS No. 16275-44-8), a mycotoxin produced by several Fusarium species such as Fusarium tricinctum and Fusarium graminearum, is frequently detected from the cereals in the endemic areas of KD. The present study is undertaken to investigate whether this mycotoxin can induce myocardial damage. Exposure of primary culture of cardiac myocytes to butenolide resulted in significant cytoto

Ten samples of stored wheat grain and 10 samples of settled grain dust released during machine threshing of wheat grain were collected on 10 farms located in Lublin province (eastern Poland). The samples were examined for the concentration of total microfungi, Fusarium species, fusariotoxins (moniliformin, deoxynivalenol, nivalenol), and ochratoxin. Microfungi able to grow on malt agar were present in 30% of grain samples (median for all examined samples = 0, range 0-227.5 x 10(3) cfu/g) and in all samples of grain dust (median = 977.5 x 10(3) cfu/g, range 115.0-16,700.0 x 10(3) cfu/g). The fumonisins, mycotoxins produced by Fusarium moniliforme, are known to occur worldwide as natural contaminants of corn. They are associated with several animal diseases and are a potential threat to human health. A total of 127 samples of corn-based foods (gluten-free diet) in the Czech Republic were analysed by Ridascreen Fumonisin Fast ELISA methods in years 1995-1996. Eighty eight % of the corn-based foods were found to be positive for fumonisins (FB1, FB2, FB3) and 12% of the examined corn-based foods laid below of a determination limit which was about 9 ng fumonisins/g corn-based foods

Sweetpotato black rot, caused by Ceratocystis fimbriata, is a major postharvest disease that leads to substantial storage losses worldwide. In this study, a salt-tolerant rhizobacterial strain, Bacillus albus SSR3, was isolated from the rhizosphere of sweetpotato grown in saline-alkali soil, with broad-spectrum antagonistic activity against postharvest fungal pathogens. LC-MS/MS analysis revealed diverse bioactive metabolites associated with its antifungal activity. Integrated transcriptomic and metabolomic analyses showed that SSR3 bioactive metabolites extensively reprogrammed fungal metabol

Tenuazonic acid (TeA) is one of the most frequently detected Alternaria mycotoxins in contaminated food. Despite its frequent occurrence, its immunomodulatory effects remain insufficiently characterized. Therefore, the present study investigated the impact of TeA on inflammatory signaling and cytokine regulation in monocytes and intestinal epithelial cell (IEC) models. NF-κB activity was assessed using a reporter gene assay in THP1-Lucia™ monocytes, while cytokine mRNA expression and protein secretion were quantified in Caco-2 and HCEC-1CT cells by qRT-PCR and ELISA, respectively. In THP-1 mon

Alternariol (AOH) is a low-molecular-weight mycotoxin produced by Alternaria species, frequently found in cereal grains, tomatoes, and processed foods. While its genotoxic and endocrine-disrupting effects are well established, emerging studies suggest a neurotoxic potential that remains poorly understood. Astrocytes-key regulators of neuronal support and calcium (Ca2+) balance in the central nervous system (CNS)-are an underexplored target of AOH toxicity. Here, we investigated the effects of AOH on Ca2+ signaling and viability in human astrocytes (Gibco Human Astrocyte, GHA cells). AOH exposu

Mycotoxins are the most frequently occurring natural contaminant in food and feed products. Through the deployment of diverse agricultural strategies or biological, chemical, or physical treatments of crop products, mycotoxin contamination remains a persistent issue for the agricultural sector and food/feed industry. We previously suggested that halophytes, thanks to their high antioxidant activity, could protect animal cell lines from mycotoxin contamination. Here, a hydroalcoholic extract of Calystegia soldanella L. leaves was evaluated for in vitro total antioxidant capacity (TAC) and 2,2-d

Fusarium proliferatum causes rice spikelet rot and contaminates grains with fumonisins. Here, we report that the methyltransferase FpLaeA is a global regulator essential for its pathogenicity. Deletion of FplaeA impaired conidiation and led to conspicuous accumulation of β-1,3-glucan. The ΔFplaeA mutant failed to produce fumonisin B1 (FB1), accompanied by the downregulation of the biosynthetic gene cluster, depletion of the precursor alanine, and disruption of sphingolipid homeostasis. It also showed defective invasive hyphal growth and attenuated secretion of pectate lyase (PL) and polygalact

Mycotoxins are toxic secondary metabolites produced by various fungi that can contaminate food crops, which, in turn, may lead to human exposure. Chronic exposure to mycotoxins can cause adverse health effects including reproductive and developmental toxicity. Pregnant women and their foetuses present a vulnerable group for exposure to mycotoxins that can cross the placenta. Human biomonitoring of mycotoxins provides a real-life approach to estimate internal exposure. In this pilot study, 24-h urine samples from 36 pregnant Dutch women were analysed for aflatoxin M1 (AFM1), total deoxynivaleno

INTRODUCTION: Fusarium graminearum is a most destructive fungal pathogen that causes Fusarium head blight (FHB) disease in cereal crops, resulting in severe yield loss and mycotoxin contamination in food and feed. Silver nanoparticles (AgNPs) are extensively applied in multiple fields due to their strong antimicrobial activity and are considered alternatives to fungicides. However, the antifungal mechanisms and the effects of AgNPs on mycotoxin production have not been well characterized.
OBJECTIVES: This study aimed to investigate the antifungal activity and mechanisms of AgNPs against both

Mycotoxicosis is the second most important problem faced by the Pakistan poultry industry, after high feed prices. The present experimental study was designed to investigate the toxicopathological effects of aflatoxin B1 (AFB1) in commercial broiler chicks and its amelioration with locally produced mycotoxin binder. Total of 125 broiler chicks was divided into five equal groups (A-E). Group A served as negative control, group B (300 μg AFB1/kg feed) as positive control, group C (300 μg AFB1/kg + Local Mycotoxin Binder (LMB), 1 g/kg feed), group D (300 μg AFB1/kg + 2 g LMB/kg feed), and group E

Novel strategies that address vitamin A deficiency have been developed, such as high-carotenoid maize, a biofortified transgenic maize line rich in carotenoids generated by genetic transformation. The South African white maize inbred (M37W), which is devoid of carotenoids, was engineered to accumulate high levels of β-carotene (provitamin A), lutein, and zeaxanthin. Maize seeds contaminated with fumonisins and other mycotoxins pose a serious threat to both humans and livestock. During three consecutive harvests, the fungal incidence and the fumonisin and aflatoxin content of maize seeds grown

Enniatins (ENs) are secondary metabolites produced by several Fusarium strains, chemically characterized as N-methylated cyclohexadepsipeptides. These compounds are known to act as antifungal and antibacterial agents, but they also possess anti-insect and phytotoxic properties. In this study, the antimicrobial effect of pure fractions of the bioactive compounds ENs A, A₁, A₂, B, B₁, and B₄ was tested towards nine probiotic microrganisms, twenty-two Saccharomyces cerevisiae strains and nine Bacillus subtilis strains. Antimicrobial analyses were carried out the disc-diffusion method using ENs co

INTRODUCTION: Infant and young child (IYC) growth impairment remains a public health problem in Africa partly because infants are exposed to staple foods (contaminated with mycotoxins) at an early age. Understanding the role of mycotoxins in IYC growth is vital, and this paper systematically reviews the available knowledge.
METHODS: Studies were searched and included if they provided information on African IYC mycotoxin exposure rates and/or growth. Studies were excluded if subjects were older than 15 years, if they were animal studies or focusing on other mycotoxins. Relevant search words we

1. The objective of this study was to investigate in vitro and in vivo (in broiler chickens) ochratoxin A (OTA) adsorption efficiency of three different adsorbents: inorganic (modified zeolite); organic (esterified glucomannans) and mixed (inorganic and organic components plus enzymes). 2. The aim of the study was to investigate which of these adsorbents provided the best protection against the presence of residues of OTA in the pectoral muscle and liver of broilers given an OTA-contaminated diet. In addition, it was important to test and compare the results of adsorbent efficiency using two d

Butenolide, a mycotoxin elaborated by several toxigenic Fusarium species, has been implicated as an etiological factor of Kashin-Beck disease and it is always detected in food from endemic Kashin-Beck disease areas. Although butenolide is considered as a potential health risk to humans and animals, its toxicity targets and mechanism of action have not been fully understood and the knowledge of its developmental toxicity is absent. The present study investigated butenolide embryotoxicity via an in vitro whole embryo culture system using rat embryos. Embryos exposed to butenolide at a concentrat

One hundred and fifty soil samples collected from different crop fields in southern Ontario, Canada were screened to obtain microorganisms capable of transforming deoxynivalenol (DON) to de-epoxy DON (dE-DON). Microbial DON to dE-DON transformation (i.e. de-epoxydation) was monitored by using liquid chromatography-ultraviolet-mass spectrometry (LC-UV-MS). The effects of growth substrates, temperature, pH, incubation time and aerobic versus anaerobic conditions on the ability of the microbes to de-epoxydize DON were evaluated. A mixed microbial culture from one composite soil sample showed 100%

The aim of the current study was to evaluate and compare two representative samples of different classes of adsorbents intended for use as feed additives in the prevention or reduction of the adverse effects exerted by mycotoxins, specifically ochratoxin A (OTA) and zearalenone (ZEN). The adsorbents, an organically activated bentonite (OAB) and a humic acid polymer (HAP), were tested in a common in vitro model with a pH course comparing the maximum pH changes that can be expected in the digestive system of a monogastric animal, i.e. pH 7.4 for the oral cavity, pH 3.0 for the stomach, and pH 8.

Fumonisin B1 (FB1) is a mycotoxin produced by Fusarium moniliforme, a common fungus which occurs naturally on corn, and other Fusarium species. FB1 and other fumonisins are now recognized as having potentially important animal and human health implications. However, few toxicological data are currently available. Male and female B6C3F1 mice and Fischer 344 rats were fed diets containing 0, 1, 3, 9, 27, or 81 ppm FB1 (> or = 98% purity) for 13 weeks. No differences in behavior or appearance, body weight or food consumption between control and FB1-fed groups were found. In mice, hepatopathy and
