T-cell lines have been derived from aeroallergen-induced eczematous patch test sites of patients with atopic dermatitis (AD). A primary culture of human antral somatostatin cells has been developed and used in release studies. A rapid method for quantitating individual free fatty acids (FFA) in plasma has been developed. An internal standard is added to plasma, the FFA are extracted into an organic solvent, concentrated and injected into a gas chromatograph. For routine estimations, extraction and assay of a single sample takes about 30 min and up to 30 samples can be assayed in a day. Seven FFA (myristic, palmitic, palmitoleic, stearic, oleic, linoleic and linolenic acids), are routinely quantitated. Sinomenine, an epimorphinan alkaloid, was tested for protecting hepatitis induced by lipopolysaccharide (LPS) in galactosamine (GalN)-sensitized mice. Sinomenine protected against the hepatic injuries in the dose range of 10-100 mg/kg in a dose-dependent manner and suppressed the production of tumor necrosis factor (TNF), which appeared in serum earlier than aminotransferases in GalN/LPS-treated mice. BACKGROUND: Islet amyloid polypeptide (IAPP) is deposited as amyloid in the islets of Langerhans in type 2 diabetes. The mechanism behind the formation of the cytotoxic fibrils is unknown.
