Monosynaptic Tracing using Modified Rabies Virus Reveals Early and Extensive Circuit Integration of Human Embryonic Stem Cell-Derived Neurons.

Human embryonic stem cell (hESC)-derived dopamine neurons are currently moving toward clinical use for Parkinson's disease (PD). However, the timing and extent at which stem cell-derived neurons functionally integrate into existing host neural circuitry after transplantation remain largely unknown. In this study, we use modified rabies virus to trace afferent and efferent connectivity of transplanted hESC-derived neurons in a rat model of PD and report that grafted human neurons integrate into the host neural circuitry in an unexpectedly rapid and extensive manner. The pattern of connectivity

Advancing glaucoma detection with convolutional neural networks: a paradigm shift in ophthalmology.

A leading cause of irreversible vision loss, glaucoma needs early detection for effective management. Intraocular Pressure (IOP) is a significant risk factor for glaucoma. Convolutional Neural Networks (CNN) demonstrate exceptional capabilities in analyzing retinal fundus images, a non-invasive and cost-effective imaging technique widely used in glaucoma diagnosis. By learning from large datasets of annotated images, CNN can identify subtle changes in the optic nerve head and retinal structures indicative of glaucoma. This enables early and precise glaucoma diagnosis, empowering clinicians to

Measuring instability in chronic human intracortical neural recordings towards stable, long-term brain-computer interfaces.

Intracortical brain-computer interfaces (iBCIs) enable people with tetraplegia to gain intuitive cursor control from movement intentions. To translate to practical use, iBCIs should provide reliable performance for extended periods of time. However, performance begins to degrade as the relationship between kinematic intention and recorded neural activity shifts compared to when the decoder was initially trained. In addition to developing decoders to better handle long-term instability, identifying when to recalibrate will also optimize performance. We propose a method, "MINDFUL", to measure in

Computer-assisted prediction of atherosclerotic intimal thickness based on weight of adrenal gland, interleukin-6 concentration, and neural networks.

OBJECTIVE: Atherosclerosis (AS) is the main pathological basis of ischemic cardio-cerebrovascular diseases, and the intimal thickness (IT) of large arteries is regarded as a powerful evaluation indicator for AS. We established an effective neural network model for automatic prediction of the IT and analyzed the high-risk warning indicators of IT. METHODS: The weight of the left adrenal (WLA) was evaluated. The serum interleukin-6 (IL-6) concentration was measured by enzyme-linked immunosorbent assay. The statistical methods included neural network modeling, a cubic spline interpolation algori

Dynamic acetylation profile during mammalian neurulation.

BACKGROUND: Neural tube defects (NTDs) result from failure of neural tube closure during embryogenesis. These severe birth defects of the central nervous system include anencephaly and spina bifida, and affect 0.5-2 per 1,000 pregnancies worldwide in humans. It has been demonstrated that acetylation plays a pivotal role during neural tube closure, as animal models for defective histone acetyltransferase proteins display NTDs. Acetylation represents an important component of the complex network of posttranslational regulatory interactions, suggesting a possible fundamental role during primary n

Aggregation of scaffolding protein DISC1 dysregulates phosphodiesterase 4 in Huntington's disease.

Using a spatially filtered magnetoencephalography analysis (synthetic aperture magnetometry), we estimated neural activations in the Stroop task in nearly real time for schizophrenic patients with/without auditory hallucinations and for normal control subjects. In addition, auditory hallucinations were examined through the information processing flow of the brain neural network, including the frontal regions. One hundred unaveraged magnetoencephalography signals during the incongruent stimulus responses were analyzed with a time window of 200 ms in steps of 50 ms. In the 25-60-Hz band, cortica

[Prevalence and spatial distribution of neural tube defects in São Paulo State, Brazil, before and after folic acid flour fortification].

This cross-sectional study analyzed the prevalence and spatial distribution of neural tube defects before and after folic acid flour fortification. The study used the Information System on Live Births (SINASC) and presented prevalence rates according to maternal characteristics with odds ratios (OR) and 95% confidence intervals (95%CI). Polynomial regression was used in time trend analysis and empirical Bayesian smoothed maps for spatial analysis. Total prevalence of neural tube defects decreased by 35%, from 0.57/1,000 to 0.37/1,000 live births after fortification (OR = 0.65; 95%CI: 0.59-0.72

Hearing Aid Noise Reduction Algorithms Support Neural Speech Tracking by Enhancing Spectrotemporal Consistency of Phase-Locked Brain Responses to Acoustic Edges.

Previous works that have analyzed electroencephalographic (EEG) speech envelope encoding in hearing-aid users using stimulus reconstruction (SR) approaches have shown that noise reduction (NR) algorithms can improve the neural tracking of speech in noise. Beyond enhancing speech envelope encoding, NR may also restore the perceptibility of acoustic edges that trigger auditory late responses (ALRs), likely promoting more consistent ALR elicitation and thus enhancing brain-to-speech synchronization. To investigate this hypothesis, 26 hearing-impaired listeners were fitted with bilateral hearing a

Electrocochleography During Translabyrinthine Approach for Vestibular Schwannoma Removal.

HYPOTHESIS: Intraoperative electrocochleography (ECochG) can differentiate hair cell and neural dysfunction caused by a vestibular schwannoma (VS) and help define the site of lesion as peripheral or central to the spiral ganglion. BACKGROUND: Hearing loss in patients with a VS can be caused by both sensory (cochlear) and neural dysfunction. METHODS: Round-window ECochG using low and high-frequency tone bursts was performed in 49 subjects with a VS. Responses were analyzed for magnitude, spectrum, and shape of the time waveform. Components examined included the cochlear microphonic, auditory

Cardiac outflow tract development relies on the complex function of Sox4 and Sox11 in multiple cell types.

Congenital heart defects represent the most common human birth defects and are often life-threatening. Frequently, they are caused by abnormalities of the outflow tract whose formation results from coordinated development of cells from mesodermal and neural crest origin and depends on the activity of many different transcription factors. However, place, time, and mode of action have only been analyzed for a few of them. Here we assess the contribution of the closely related high-mobility-group transcription factors Sox4 and Sox11 to outflow tract development and determine their function. Using

Alterations of plasma neural-derived extracellular vesicles microRNAs in patients with bipolar disorder.

BACKGROUND: MicroRNAs (miRNAs) alterations in patients with bipolar disorder (BD) are pivotal to the disease's pathogenesis. Since obtaining brain tissue is challenging, most research has shifted to analyzing miRNAs in peripheral blood. One innovative solution is sequencing miRNAs in plasma extracellular vesicles (EVs), particularly those neural-derived EVs emanating from the brain. METHODS: We isolated plasma neural-derived EVs from 85 patients with BD and 39 healthy controls (HC) using biotinylated antibodies targeting a neural tissue marker, followed by miRNA sequencing and expression anal

Consistent spectro-spatial features of human ECoG successfully decode naturalistic behavioral states.

OBJECTIVE: Understanding the neural correlates of naturalistic behavior is critical for extending and confirming the results obtained from trial-based experiments and designing generalizable brain-computer interfaces that can operate outside laboratory environments. In this study, we aimed to pinpoint consistent spectro-spatial features of neural activity in humans that can discriminate between naturalistic behavioral states. APPROACH: We analyzed data from five participants using electrocorticography (ECoG) with broad spatial coverage. Spontaneous and naturalistic behaviors such as "Talking"

[Artificial intelligence in predicting the risk of facial bone osteoporosis: clinical significance and prospects.].

Osteoporosis of the jawbones is a significant concern in dental practice, particularly for implant treatment planning. This review summarizes current diagnostic approaches with a focus on the use of artificial intelligence (AI) algorithms, including convolutional neural networks, for analyzing panoramic radiographs and cone-beam computed tomography. The findings demonstrate that AI models achieve high diagnostic accuracy in the automated classification of radiographic images, comparable to dual-energy X-ray absorptiometry. AI reduces subjectivity in image interpretation, although further stand

The endocannabinoid system and neurogenesis in health and disease.

The endocannabinoid system exerts an important neuromodulatory function in different brain areas and is also known to be involved in the regulation of neural cell fate. Thus, CB(1) cannabinoid receptors are neuroprotective in different models of brain injury, and their expression is altered in various neurodegenerative diseases. Recent findings have demonstrated the presence of a functional endocannabinoid system in neural progenitor cells that participates in the regulation of cell proliferation and differentiation. In this Research Update, the authors address the experimental evidence regard

Rapid and non-destructive estimation of chemical properties of date syrup using visible-near infrared hyperspectral imaging combined with machine learning predictive models.

In this study, an innovative hybrid approach combining hyperspectral imaging and artificial intelligence was developed to replace costly and time-consuming laboratory methods of determining chemical properties of date syrup. Hyperspectral data related to acidity, Brix, fructose, HMF, pH, and viscosity were analyzed using three machine learning models: Artificial neural network, partial least squares regression, and support vector regression. These models were evaluated under two strategies, with and without feature selection step. Among the models, the Artificial Neural Network demonstrated su

Putative cell-type-specific multiregional mode in posterior parietal cortex during coordinated visual behavior.

In the reach and saccade regions of the posterior parietal cortex (PPC), multiregional communication depends on the timing of neuronal activity with respect to beta-frequency (10-30 Hz) local field potential (LFP) activity, termed dual coherence. Neural coherence is believed to reflect neural excitability, whereby spiking tends to occur at a particular phase of LFP activity, but the mechanisms of multiregional dual coherence remain unknown. Here, we investigate dual coherence in the PPC of non-human primates performing eye-hand movements. We computationally model dual coherence in terms of mul

Large-scale electrophysiology and deep learning reveal distorted neural signal dynamics after hearing loss.

Listeners with hearing loss often struggle to understand speech in noise, even with a hearing aid. To better understand the auditory processing deficits that underlie this problem, we made large-scale brain recordings from gerbils, a common animal model for human hearing, while presenting a large database of speech and noise sounds. We first used manifold learning to identify the neural subspace in which speech is encoded and found that it is low-dimensional and that the dynamics within it are profoundly distorted by hearing loss. We then trained a deep neural network (DNN) to replicate the ne
