AKI and the Neuroimmune Axis.

Neuroimmune interaction is an emerging concept, wherein the nervous system modulates the immune system and vice versa. This concept is gaining attention as a novel therapeutic target in various inflammatory diseases including acute kidney injury (AKI). Vagus nerve stimulation or treatment with pulsed ultrasound activates the cholinergic anti-inflammatory pathway to prevent AKI in mice. The kidneys are innervated by sympathetic efferent and sensory afferent neurons, and these neurons also may play a role in the modulation of inflammation in AKI. In this review, we discuss several neural circuit

A direct projection from the subthalamic nucleus to the ventral thalamus in monkeys.

The current basal ganglia model considers the internal division of the globus pallidus and the substantia nigra pars reticulata as the sole sources of basal ganglia output to the thalamus. However, following the delivery of retrograde tracers into the ventral anterior/ventral lateral thalamic nuclei, a moderate number of labeled neurons were found within the subthalamic nucleus (STN) in control cases, MPTP-treated monkeys and animals with levodopa-induced dyskinesias. Furthermore, dual tracing experiments showed that subthalamo-thalamic and subthalamo-pallidal projections arise from different

Behavior-reactive neuron populations in the monkey neostriatum.

Comparative analysis of neuron activity in the monkey putamen during multistep behavior showed that putamen neurons are active during all the animal's behavioral actions. The difference between the number of active neurons at a given step of the behavior as compared with the preceding step was found to be significantly smaller than the number of neurons reorganizing their activity at this step. Reorganization of neuron activity in the putamen is regarded as a reflection of the efferent code controlling the behavior, while the extent of reorganization is regarded as a measure of the change in t

Nonaminergic striatal neurons convert exogenous L-dopa to dopamine in parkinsonism.

In intact striatum, the enzyme dopa decarboxylase is localized predominantly in dopaminergic nerve terminals. In Parkinson disease, loss of dopaminergic neurons is associated with massive depletion of striatal decarboxylase activity. Nevertheless, efficacy of exogenous L-dopa in parkinsonism is generally believed to result from its enzymatic decarboxylation to dopamine in the corpus striatum. It has previously been suggested that, after degeneration of nigrostriatal pathways, decarboxylation of administered L-dopa may occur mainly at such striatal sites as surviving dopaminergic terminals, ser

Nerve pathways in celiac plexus of the guinea pig.

In vitro preparations consisted of the right and left celiac, superior mesenteric and inferior mesenteric ganglia with attached extrinsic nerves, vasculature, mesentery, and colon. There were no systematic differences in membrane electrical properties (recorded intracellularly) between neurons in the different ganglia. Stimulation of associated nerve trunks produced graded synaptic responses in plexus neurons. Presynaptic fibers were found in splanchnic and mesenteric nerves. Input from celiac nerves dominated in the celiac galglia; input from the intermesenteric fibers dominated in the superi

Electrophysiological study of vagal afferent and efferent units in conscious sheep.

Crossed-nerve anastomoses were performed to record activity of vagal efferents and afferents during regurgitation and eructation in conscious sheep. Following anastomoses of the central end of a vagus nerve to the peripheral end of a phrenic nerve, discharges in units of the diaphragm reinnervated by vagal efferent neurones were associated with contractions of the caudal oesophagus and of the rumen. During regurgitation in the course of rumination, discharges in oesophageal efferents consisted of a characteristic pattern of two bursts in a constant association with the additional phase of the

Innervation of the carotid body of the adult rat. A serial ultrathin section analysis.

The innervation of the carotid body of adult rats was studied by means of serial ultrathin sections. A single branching nerve fiber innervates 12 chief cells through several kinds of terminals (vesicle-containing, mitochondrial sack, and calyx-type) in en-passant and bouton forms. Two types of synaptic contacts between nerve terminals and chief cells are found; type 1 in which chief cells are postsynaptic, and type 2 in which chief cells are presynaptic. Since a single nerve fiber (possibly from the glossopharyngeal nerve) forms both types of synapses with type 2 predominating, the nerve fiber

Nerve endings in rat carotid body.

The carotid body of the rat consists of "glomera" interspersed by blood vessels and nerve bundles. Each "glomus" consists of 2-12 Type I cells, incompletely invested by 1-3 Type II cells. Type I cells are characterised by the presence of numerous dense cored vesicles in their cytoplasm and may exhibit "synaptic" -like contacts with each other. Small efferent nerve endings make synaptic contact with one or more Type I cells. Large cup-shaped afferent nerve endings make multiple synaptic contacts of two kinds with one or more Type I cells. A second kind of efferent nerve ending is occasionally s

Cranial sensory ganglia neurons require intrinsic N-cadherin function for guidance of afferent fibers to their final targets.

Cell adhesion molecules, such as N-cadherin (cdh2), are essential for normal neuronal development, and as such have been implicated in an array of processes including neuronal differentiation and migration, and axon growth and fasciculation. cdh2 is expressed in neurons of the peripheral nervous system during development, but its role in these cells during this time is poorly understood. Using the transgenic zebrafish line, tg(p2xr3.2:eGFP(sl1)), we have examined the involvement of cdh2 in the formation of sensory circuits by the peripheral nervous system. The tg(p2xr3.2:eGFP(sl1)) fish allows

[Influence of GABAergic neurons in the brain on central effects of kainate on the cardiovascular system].

Microinjection of kainate (KA 0.125, 0.25, or 0.5 microgram), a special agonist for KA receptor, into rat lateral cerebral ventricle produced dose-dependent increases in blood pressure and heart rate. Pretreatment with semicarbazide (SCZ 140 mg/kg, ip), an inhibitor of GABA biosynthesis, or picrotoxin (PIC 1 mg/kg, iv), an antagonist for GABA, significantly enhanced the effects of KA on the cardiovascular system. These effects of KA were reduced by aminoxyacetic acid (AOAA 25 micrograms/rat, icv), an inhibitor of GABA aminotransferase. All these results described above imply that the function

Auditory efferent feedback system deficits precede age-related hearing loss: contralateral suppression of otoacoustic emissions in mice.

The C57BL/6J mouse has been a useful model of presbycusis, as it displays an accelerated age-related peripheral hearing loss. The medial olivocochlear efferent feedback (MOC) system plays a role in suppressing cochlear outer hair cell (OHC) responses, particularly for background noise. Neurons of the MOC system are located in the superior olivary complex, particularly in the dorsomedial periolivary nucleus (DMPO) and in the ventral nucleus of the trapezoid body (VNTB). We previously discovered that the function of the MOC system declines with age prior to OHC degeneration, as measured by contr

Vagal stimulation targets select populations of intrinsic cardiac neurons to control neurally induced atrial fibrillation.

Mediastinal nerve stimulation (MNS) reproducibly evokes atrial fibrillation (AF) by excessive and heterogeneous activation of intrinsic cardiac (IC) neurons. This study evaluated whether preemptive vagus nerve stimulation (VNS) impacts MNS-induced evoked changes in IC neural network activity to thereby alter susceptibility to AF. IC neuronal activity in the right atrial ganglionated plexus was directly recorded in anesthetized canines (n = 8) using a linear microelectrode array concomitant with right atrial electrical activity in response to: 1) epicardial touch or great vessel occlusion vs. 2

Localization of kainate receptors in inner and outer hair cell synapses.

Glutamate plays a role in hair cell afferent transmission, but the receptors that mediate neurotransmission between outer hair cells (OHCs) and type II ganglion neurons are not well defined. A previous study using in situ hybridization showed that several kainate-type glutamate receptor (KAR) subunits are expressed in cochlear ganglion neurons. To determine whether KARs are expressed in hair cell synapses, we performed X-gal staining on mice expressing lacZ driven by the GluK5 promoter, and immunolabeling of glutamate receptors in whole-mount mammalian cochleae. X-gal staining revealed GluK5 e

Physiological significance of 2-buten-4-olide (2-B4O), an endogenous satiety substance increased in the fasted state.

A sugar acid, 2-B4O, has been found to increase from 3.5 to 13 microM in rat serum at 36 h after food deprivation. Injections of 2-B4O (2.5 microM) into the rat III cerebral ventricle (III ICV) suppress food intake and single neuronal activity in the lateral hypothalamic area (LHA). 2-B4O is effective even in 72 h food-deprived rats. 2-B4O hyperpolarizes glucose-sensitive neurons in the LHA via Na+-K+ pump activation, but depolarizes glucoreceptor neurons in the ventromedial nucleus (VMH) via closure of ATP-sensitive K channels. The plasma levels of glucose, corticosterone, and catecholamines,

The spatial patterns of pathological brain lesions in 12 patients with corticobasal degeneration.

Corticobasal degeneration (CBD) is a rare and progressive neurological disorder characterised by the presence of ballooned neurons (BN) and tau positive inclusions in neurons and glial cells. We studied the spatial patterns of the BN, tau positive neurons with inclusions (tau+ neurons), and tau positive plaques in the neocortex and hippocampus in 12 cases of CBD. All lesions were aggregated into clusters and in many brain areas, the clusters were distributed in a regular pattern parallel to the tissue boundary. In the majority of cortical areas, the clusters of BN were larger in the lower comp

Neural mechanisms in disorders of movement.

1. Experimental models of ballism, chorea and Parkinson's disease have been developed in the primate, and the underlying neural mechanisms which mediate these disorders of movement have been investigated using the 2-deoxyglucose uptake technique. 2. In ballism, the subthalamic nucleus is either lesioned or underactive. Because of the excitatory nature of subthalamic efferent fibres, this leads to abnormal underactivity of neurons in the medical segment of the globus pallidus which project to the ventral anterior and ventral lateral nuclei of the thalamus, and to the pedunculopontine nucleus of

Botulinum neurotoxin serotype A suppresses neurotransmitter release from afferent as well as efferent nerves in the urinary bladder.

BACKGROUND: Botulinum neurotoxin A (BoNTA), which alleviates overactive bladder symptoms, is thought to act predominantly via the inhibition of transmitter release from parasympathetic nerves. However, actions at other sites such as afferent nerve terminals are possible. OBJECTIVE: To evaluate the effects of BoNTA on bladder afferent neuropeptide release and firing. DESIGN, SETTING, AND PARTICIPANTS: One side of the bladder of control and chronic (1-2 wk) spinal cord transected (SCT; T(8)-T(9)) adult female mice was injected with BoNTA (0.5 U/5 μl saline). After 48 h, bladders with L(6)-S(2)

The Aubert-Fleischl phenomenon: a temporal frequency effect on perceived velocity in afferent motion perception.

Apparent velocities of moving visual stimuli are known to be different depending on whether the subject pursues the stimulus (efferently controlled motion perception) or whether the eye is stationary and the image moves across the retina (afferent motion perception). Afferent motion perception of a periodic pattern or a moving single object causes overestimation of velocity (magnitude estimations) as compared to smooth pursuit. This socalled Aubert-Fleischl phenomenon is shown to depend on local temporal frequency stimulation on the retina caused by the repetitive passage of contrast borders o

A study on the pattern of alkaline phosphatase activity correlated with observations on silver-impregnated structures in the developing mouse brain.

Alkaline phosphatase activity was localised in specific groups of cellular processes, nuclei and fibre tracts in fetal mouse brain at 9.5-17.5 days post coitum. A radial band of intense enzymatic activity extended longitudinally within the rhombencephalon at 10.5 to 15.5 days. This enzymatic band was topographically related to the efferent nuclei of the cranial nerves, except those nuclei which had migrated laterally to their final positions. The enzymatic activity was seen in some but not all silver-impregnated fibre tracts that appeared early in development, but such association disappeared
