Evaluation of N-palmitoylethanolamine (PEA) binding to nuclear receptors through docking and molecular dynamics studies.

N-palmitoylethanolamine (PEA) is an endogenous bioactive compound recognized for its anti-inflammatory effects and its role in tissue protection and repair. Despite the proposal of peroxisome proliferator-activated receptor alpha (PPARα) as a potential receptor for PEA, direct evidence of binding remains insufficient. This study offers a comprehensive analysis of human nuclear receptors (NRs) through structural bioinformatics and molecular docking, evaluating a total of 367 unique NR structures across 47 subfamilies. To explore the stability and binding affinity of PEA with selected nuclear re

Multi-omics dissection of heat stress reveals CIITA as a central regulator of metabolic thermotolerance in cattle (Bos taurus).

Climate-driven heat stress disrupts metabolic homeostasis in livestock, yet the molecular mechanisms underlying adaptive responses remain poorly understood. Here, we integrated newly generated plasma metabolomic data from 111 heat-stressed cows with previously published whole-genome sequencing datasets from the same animals, identifying 30 metabolic markers and 27 copy number variations (CNVs) associated with 25 candidate genes involved in the regulation of these metabolites. Notably, a CNV hotspot encompassing CIITA emerged as a key pleiotropic locus strongly associated with acylcarnitine lev

[Increased efficacy of radiation protection against fission neutrons using unithiol].

It was found that the combination of unithiol (Sodium salt of 2,3-dimercapto-1-propansulfonic acid) with cystamine and AET diminished their toxicity. The optimum ratio for the antitoxic effect is 0.5 molar equivalent of unithiol per radioprotective 1.0 equivalent of thiol. Animals withstand big doses of protectors well, that gives an opportunity to use increased amounts of cystamine and AET. In the experiments with circular irradiation of male (CBA x C57B1)F1 mice weighing 18-22 g with fission neutrons (the neutron mean energy was 0.85 MeV, the contribution of gamma-quanta to the total was 25%

Non-cancer related chromosome translocation frequency measured from a leukemia patient's blood.

A new approach used to measure pre-cancer-induced genetic damage in a patient diagnosed as early B-precursor acute lymphoblastic leukemia, by totally separating the malignant B-lymphocytes and the normal T-lymphocytes is described. The lymphocytes separation by a rosetting method and chromosome translocation analysis using chromosome painting were employed. In the case presented here, the utility of this approach is illustrated using blood lymphocytes from a nuclear dockyard worker who claims that his leukemia was induced by work-related radiation exposures. Blood lymphocytes were obtained aft

Non-linear bonding trends in maleonitrile-1,2-dithiolate complexes of the transuranium actinides.

The trivalent actinides are produced in the nuclear fuel cycle during power production and provide the largest long-term radiation dose in used nuclear fuel. It is ideal for these elements to be removed from used nuclear fuel for disposal and a necessity for fuel recycling. A key challenge to this is the similarity of chemical behavior of the trivalent actinides to the lanthanides that are also present as fission products in used fuel. Thus far, some of the most effective separations of actinides from lanthanides utilise chelating agents containing sulfur moieties such as dithiophosphinates th

Mitochondrial dysfunction in bipolar disorder: Evidence, pathophysiology and translational implications.

Bipolar disorder (BD) is a chronic psychiatric illness characterized by severe and biphasic changes in mood. Several pathophysiological mechanisms have been hypothesized to underpin the neurobiology of BD, including the presence of mitochondrial dysfunction. A confluence of evidence points to an underlying dysfunction of mitochondria, including decreases in mitochondrial respiration, high-energy phosphates and pH; changes in mitochondrial morphology; increases in mitochondrial DNA polymorphisms; and downregulation of nuclear mRNA molecules and proteins involved in mitochondrial respiration. A comparative study of the responses of lithium borate and calcium sulphate phosphors in a TL personal dosemeter.

This work summarises the results of a series of experiences made on Panasonic UD-802AS, a multi-element dosemeter that is currently used in Caorso Nuclear Power Plant for personnel external monitoring. Two main topics have been considered: energy response and the influence of working place dose rate on relative readings of the elements at the end of the monitoring period. The presence of four elements of different materials and filtrations gives the possibility of evaluating radiation energy by a simple algorithm, so that an accurate estimate of the personal dose equivalent can be provided; th

Mutant huntingtin causes defective actin remodeling during stress: defining a new role for transglutaminase 2 in neurodegenerative disease.

Huntington's disease (HD) is caused by an expanded CAG tract in the Interesting transcript 15 (IT15) gene encoding the 350 kDa huntingtin protein. Cellular stresses can trigger the release of huntingtin from the endoplasmic reticulum, allowing huntingtin nuclear entry. Here, we show that endogenous, full-length huntingtin localizes to nuclear cofilin-actin rods during stress and is required for the proper stress response involving actin remodeling. Mutant huntingtin induces a dominant, persistent nuclear rod phenotype similar to that described in Alzheimer's disease for cytoplasmic cofilin-act

FLG249 Exhibits FXR Antagonist Activity by Inducing Dissociation of Both Corepressors and Coactivators from FXR.

The farnesoid X receptor (FXR), a nuclear receptor activated by endogenous bile acids, regulates not only bile acid synthesis but also lipid and carbohydrate metabolism. Therefore, FXR ligands, including FXR antagonists, show potential as therapeutic agents for various metabolic diseases. However, the mechanism by which FXR antagonists influence FXR activity is unclear. We previously synthesized an FXR antagonist, FLG249, which reduced the expression of several FXR target genes in the mouse ileum when orally administered and improved lipid metabolism in the liver and ileum of high-fat diet-ind

Methods of assessing body shape and composition in HIV-associated lipodystrophy.

Although HIV-associated lipodystrophy can be diagnosed just from clinical examination, criteria for diagnosis and judgement of severity are not clearly defined. Clinical assessment can be systematized with questionnaires for patients and physicians. Computed tomography, nuclear magnetic imaging and dual-energy X-ray absorptiometry are objective reference methods but are expensive for routine follow-up and diagnosis. Therefore, anthropometry and other non-invasive bedside methods deserve further validation studies.

Reversible compromise of physiological resilience by accumulation of heteroplasmic mtDNA mutations.

Somatically acquired mitochondrial DNA (mtDNA) mutations accumulate with age, but the mechanisms and consequences of this accumulation are poorly understood. Here we show that transient injuries induce a burst of persistent mtDNA mutations that impair resilience to future injuries. mtDNA mutations suppressed energy-intensive nucleotide metabolism. Repletion of adenosine, but not other nucleotides, restored adenosine triphosphate generation, which required a nuclear-encoded purine biosynthetic enzyme, adenylate kinase 4 (AK4). Analysis of 369,912 UK Biobank participants revealed a graded associ

Malate-aspartate shuttle inactivation drives pulmonary hypertension through glucose carbon flux rewiring and α-ketoglutarate-dependent epigenetic remodeling.

Pulmonary hypertension (PH) is characterized by progressive pulmonary vascular remodeling, primarily driven by the hyperproliferation of pulmonary artery smooth muscle cells (PASMCs) and their resistance to apoptosis. Metabolic reprogramming, particularly enhanced glycolysis, has been proposed to be involved in PASMCs activation. However, the implicated mechanisms remain incompletely understood. Here, using in-vitro cultured PASMCs and in-vivo PH models, we showed that malate-aspartate shuttle (MAS), a critical system connecting glycolysis to oxidative phosphorylation, was significantly inhibi

A conserved role for AMP-activated protein kinase in NGLY1 deficiency.

Mutations in human N-glycanase 1 (NGLY1) cause the first known congenital disorder of deglycosylation (CDDG). Patients with this rare disease, which is also known as NGLY1 deficiency, exhibit global developmental delay and other phenotypes including neuropathy, movement disorder, and constipation. NGLY1 is known to regulate proteasomal and mitophagy gene expression through activation of a transcription factor called "nuclear factor erythroid 2-like 1" (NFE2L1). Loss of NGLY1 has also been shown to impair energy metabolism, but the molecular basis for this phenotype and its in vivo consequences

Nuclear genes of human complex I of the mitochondrial electron transport chain: state of the art.

The mitochondrial electron transport chain (mtETC) consists of four multi-subunit enzyme complexes. Complex I or NADH:ubiquinone oxidoreductase, the largest mtETC multisubunit complex, consists of approximately 41 subunits. Seven of these subunits are encoded by the mitochondrial genome, the remainder by the nuclear genome. Among the mitochondriocytopathies, complex I deficiencies are encountered frequently. Although some complex I deficiencies have been associated with mitochondrial DNA mutations, the genetic defect has not been elucidated in the majority of complex I-deficient patients. It i

Excessive intracellular acidosis of skeletal muscle on exercise in a patient with a post-viral exhaustion/fatigue syndrome. A 31P nuclear magnetic resonance study.

A patient with prolonged post-viral exhaustion and excessive fatigue was examined by 31P nuclear magnetic resonance. During exercise, muscles of the forearm demonstrated abnormally early intracellular acidosis for the exercise performed. This was out of proportion to the associated changes in high-energy phosphates. This may represent excessive lactic acid formation resulting from a disorder of metabolic regulation. The metabolic abnormality in this patient could not have been demonstrated by traditional diagnostic techniques.

Early response of plant cell to carbon deprivation: in vivo 31P-NMR spectroscopy shows a quasi-instantaneous disruption on cytosolic sugars, phosphorylated intermediates of energy metabolism, phosphate partitioning, and intracellular pHs.

• In plant cells, sugar starvation triggers a cascade of effects at the scale of 1-2 days. However, very early metabolic response has not yet been investigated. • Soluble phosphorus (P) compounds and intracellular pHs were analysed each 2.5 min intervals in heterotrophic sycamore (Acer pseudoplatanus) cells using in vivo phosphorus nuclear magnetic resonance ((31)P-NMR). • Upon external-sugar withdrawal, the glucose 6-P concentration dropped in the cytosol, but not in plastids. The released inorganic phosphate (Pi) accumulated transiently in the cytosol before influx into the vacuole; nucleoti

PCNA and XPF cooperate to distort DNA substrates.

XPF is a structure-specific endonuclease that preferentially cleaves 3' DNA flaps during a variety of repair processes. The crystal structure of a crenarchaeal XPF protein bound to a DNA duplex yielded insights into how XPF might recognise branched DNA structures, and recent kinetic data have demonstrated that the sliding clamp PCNA acts as an essential cofactor, possibly by allowing XPF to distort the DNA structure into a proper conformation for efficient cleavage to occur. Here, we investigate the solution structure of the 3'-flap substrate bound to XPF in the presence and absence of PCNA us
