Glomeruli are signal-processing units of the olfactory bulb (OB) that play a key role in many OB computations, including contrast enhancement, gain control, and odorant-selective habituation. In awake mice, we unveil an extremely stable, inhomogeneous map of basal glomerulus-specific activity that serves as the background against which olfactory signal processing occurs. This activity is strongly driven by centrifugal cholinergic inputs; endogenous and airflow-evoked spiking of olfactory sensory neurons; and, to a minor extent, by the odor environment. Moreover, it is brain-state dependent and

The majority of patients with Parkinson disease (PD) experience a loss in their sense of smell and accumulate insoluble α-synuclein aggregates in their olfactory bulbs (OB). Subjects affected by a SARS-CoV-2-linked illness (COVID-19) also frequently experience hyposmia. We previously postulated that microglial activation as well as α-synuclein and tau misprocessing can occur during host responses following microbial encounters. Using semiquantitative measurements of immunohistochemical signals, we examined OB and olfactory tract specimens collected serially at autopsies between 2020 and 2023.

BACKGROUND: Anosmia has an estimated prevalence of 5% of the general population. Outside of inflammatory causes, therapeutic options are limited despite research advances. Bypassing peripheral neuronal damage through central stimulation is a potential therapeutic option that has shown success in other sensory systems, most notably with hearing. We performed a pilot study to determine the feasibility of inducing smell through artificial electrical stimulation of the olfactory bulbs in humans.
METHODS: Subjects with a history of sinus surgery, including total ethmoidectomy, with intact ability

Olfactory sensory neurons (OSNs) can sense both odorants and airflows. In the olfactory bulb (OB), the coding of odor information has been well studied, but the coding of mechanical stimulation is rarely investigated. Unlike odor-sensing functions of OSNs, the airflow-sensing functions of OSNs are also largely unknown. Here, the activity patterns elicited by mechanical airflow in male rat OBs were mapped using fMRI and correlated with local field potential recordings. In an attempt to reveal possible functions of airflow sensing, the relationship between airflow patterns and physiological para

BACKGROUND: Olfactory disorder is an early manifestation of Parkinson's disease (PD), likely to be associated with abnormalities of the dopaminergic neurons in the olfactory bulb (OB); however, the causes of olfactory disorder in PD are not entirely clear. Some studies showed that melatonin (MT) and androgens (mainly testosterone, T) might participate in the pathogenesis of PD. The research aimed to investigate effects of MT or T deficiency on OB dopaminergic neurons in rats.
METHODS: One hundred and twenty normal male Wistar rats were randomly divided into the control, sham operation pineale

The olfactory system of mammals comprises a main olfactory system that detects hundreds of odorants and a vomeronasal system that detects specific chemicals such as pheromones. The main (MOB) and accessory (AOB) olfactory bulbs are the respective primary centers of the main olfactory and vomeronasal systems. Most mammals including artiodactyls possess a large MOB and a comparatively small AOB, whereas most cetaceans lack olfactory bulbs. The common hippopotamus (Hippopotamus amphibius) is semiaquatic and belongs to the order Cetartiodactyla, family Hippopotamidae, which seems to be the closest

We recently described the boundary-like expression pattern of the extracellular matrix molecule tenascin-C (Tnc) in the developing mouse olfactory bulb (OB) (Shay et al., 2008). In the present study, we test the hypothesis that Tnc inhibits olfactory sensory neuron (OSN) axon growth in the developing OB before glomerulogenesis. The period of time before glomerular formation begins, when axons remain restricted to the developing olfactory nerve layer (ONL), is crucial for axon sorting. Here, we show with in vitro analyses that OSN neurite outgrowth is inhibited by Tnc in a dose-dependent manner

The main olfactory bulb (MOB) is the first site of synaptic processing in the central nervous system for odor information that is relayed from olfactory receptor neurons in the nasal cavity via the olfactory nerve (ON). Glutamate and ionotropic glutamate receptors (iGluRs) play a dominant role at ON synapses. Similarly, glutamate and iGluRs mediate dendrodendritic transmission between several populations of neurons within the MOB network. Neuroanatomical studies demonstrate that metabotropic glutamate receptors (mGluRs) are densely expressed through the MOB network, and they are particularly a

Cell death is widespread in the developing nervous system and is under complex regulation by numerous intra- and intercellular mechanisms. Blockade of the N-methyl-D-aspartate (NMDA) subtype of glutamate receptor has been shown to promote cell death in the developing brain (Ikonomidou et al., 1999), suggesting that afferent functional activation is an important regulator of cell survival. The olfactory bulb, the first central relay for olfactory information from the nose, is well suited for examining the role of afferent activity in neuronal development. Functional deprivation is easily perfor

In the present study, we describe the structural and cytological changes observed in staggerer mutant olfactory bulbs, as compared to normal mice. On the basis of photonic and ultrastructural observations we tried to define the alterations induced by the mutation: i.e. a reduction of bulb size, a reduction in the volume of three out of the six architectonic layers (glomerular, external and internal plexiform), a reduction of glomeruli size, a loss of half the mitral cells and a slight decrease in juxtaglomerular interneuron number. In staggerer, an hypertrophy of glial ensheathing cell process

The capacity of periglomerular cells (PGc) to give fast, Na-dependent action potentials is a crucial and debated issue for the comprehension of how sensory information is processed in the olfactory bulb (OB). Using patchclamp whole cell recording in thin slices of rat OB (P8-P20) we showed that fast sodium conductance is present in all the PGc studied, that this current is sufficiently large to generate action potentials and that action potentials can be evoked in these cells by direct stimulation of the olfactory nerve. A comprehensive kinetic characterization of INa is also presented.

We have derived highly enriched populations of ensheathing cells (ECs) from the olfactory nerve layer of neonatal rat olfactory bulbs. Contaminating cells, such as fibroblasts, were eliminated from EC cultures by cytosine arabinoside and immunoadsorption with antiserum to Thy-1.1. At the same time, ECs were stimulated to divide by the addition of bovine pituitary extract into the culture media. Confluent cultures containing 96-99% ECs, were comprised of either spindly bipolar cells or cells bearing multiple processes oriented on opposite poles. The ECs immunostained positively for GFAP, S-100

The olfactory bulb of many elasmobranch fishes is morphologically subdivided into distinct units or sub-bulbs immediately adjacent to the olfactory epithelium. We investigated this morphological feature in two species of shark and one species of ray in order to understand its impact on the arrangement of the primary olfactory projections onto the bulb. Using anterograde tracing methods in vitro (biocytin) as well as in fixed tissue (DiI), we observed a direct segregated projection of the olfactory afferents onto the bulb. Application of tracers to the lateral part of the olfactory epithelium r

Axons immunoreactive for calcitonin gene-related peptide (CGRP) and substance P are present in the olfactory nerve, although few, if any, olfactory receptor cells contain immunocytochemically detectable levels of these peptides. The possible trigeminal origin of these fibers was tested by performing unilateral stereotaxic lesions of the ophthalmic division of the trigeminal nerve, followed 2-25 days later by immunocytochemistry for CGRP and substance P. As reported previously, free nerve endings immunoreactive for both peptides were found transversing the nasal epithelium on the unlesioned sid

Intrinsic neurons provide inhibitory synaptic input to mitral (and tufted) output cells within several laminae of the olfactory bulb. In rodents, the two main types of intrinsic neurons are granule and periglomerular cells, both of which contain gamma-aminobutyric acid (GABA). In the present study, immunocytochemical techniques were used to determine whether intrinsic neurons in the salamander olfactory bulb might also contain GABA. With the aid of two antisera to different GABA-conjugates, immunoreactivity for GABA was localized within the olfactory bulb laminae. In the glomerular layer, peri

The presence and localization of dopamine D2 receptors was studied by means of in vitro autoradiography with [3H]N-n-propylnorapomorphine in olfactory bulbs obtained postmortem from Alzheimer patients and age-matched controls. It appeared that, in 5 age-matched controls, the greatest density of dopamine D2 receptors was found in the glomerular layer of the bulb. In 6 of 7 Alzheimer patients, the labeling of the glomerular layer was decreased so that glomerular and granular layer did not differ in labeling density. Tangles, as revealed by Thioflavin S staining, were present in all different lay

1. Intracellular and extracellular recordings were made from rat olfactory bulb mitral and tufted cells during odor stimulation and during electrical stimulation of the olfactory nerve. Neurons were identified by horseradish peroxidase injections and/or antidromic activation. The presentation of multiple concentrations of at least one odorant in a cyclic artificial sniff paradigm, as reported previously (10), allowed the study of odor responses. This approach was extended to multiple odorants to compare their concentration-response profiles. This procedure avoids the problems of interpretation
