Chemosensory cilia of olfactory receptor neurons contain an adenylate cyclase which is stimulated by high concentrations of odorants. Cyclic AMP produced by this enzyme has been proposed to act as second messenger in olfactory transduction. Here we report that olfactory cilia contain calmodulin and that calmodulin potently activates olfactory adenylate cyclase by a mechanism additive to and independent from direct stimulation by odorants. Activation by calmodulin is calcium dependent and enhanced by GTP. Thus, olfactory transduction may involve a second-messenger cascade in which an odorant-in…

Electro-olfactograms (EOGs) were recorded from both sensory and non-sensory epithelia on the olfactory lamellae and from other areas within the rosette of rainbow trout (Salmo gairdneri). The trout EOG induced by amino acids was a monophasic negative voltage composed of a phasic component which declined to a steady level (tonic component) that was maintained throughout stimulus duration. The time period (2 min) for complete recovery of the EOG was obtained by applying two identical stimuli (10(-5) mol/liter L-serine for 5 s) successively at increasing intervals. EOG response to amino acids inc

Olfactory sensory axons navigate from the nasal cavity to the olfactory bulb and sort from among 1,000 different odorant receptor-expressing types to converge upon the same two or three glomeruli. To achieve this task during development, it is likely that multiple classes of regulatory molecules, including cell adhesion molecules, are involved. Cell adhesion molecules have been shown to be important in controlling axon guidance, fasciculation, and synapse formation. To gain further understanding of the involvement of adhesion molecules in olfactory circuitry development, we examined the dynami

The spiny lobster, Panulirus argus, has two classes of chemosensilla representing "olfaction" and "distributed chemoreception," as is typical for decapod crustaceans. Olfactory sensilla are found exclusively on antennular lateral flagella and are innervated only by olfactory receptor neurons (ORNs) that project into olfactory lobes organized into glomeruli in the brain. Distributed chemoreceptor sensilla are found on all body surfaces including the antennular lateral flagella (LF) and walking leg dactyls (dactyls), and are innervated by both chemoreceptor neurons (CRNs) and mechanoreceptor neu…

The number of vomeronasal receptor cells and olfactory receptor cells were determined in several turtles and tortoises. Using semi-aquatic turtles with developed vomeronasal organs, neural responses to various odor stimuli were recorded as extracellular unitary spike responses of vomeronasal receptor cells, compound action potentials from vomeronasal nerve twigs, induced wave responses on the accessory olfactory bulb (AOB) surface and unitary responses of AOB neurons. The vomeronasal system in turtles responded to various volatile and non-volatile chemicals. These results yield insights concer

Insect repellents are widely used to protect against insect bites and thus prevent allergic reaction and the spread of disease. To gain insight into the mosquito's response to chemicals repellents, we investigated the interaction between the olfactory system of the mosquito Culex quinquefasciatus Say and chemical repellents using single sensillum recording. The interactions of 50 repellent chemicals with olfactory receptor neurons were measured in six different types of mosquito sensilla: long sharp trichoid (LST), short sharp trichoid (SST), short blunt trichoid I (SBT-I), short blunt trichoi…

Anosmia, the loss of the sense of smell, is one of the main neurological manifestations of COVID-19. Although the SARS-CoV-2 virus targets the nasal olfactory epithelium, current evidence suggests that neuronal infection is extremely rare in both the olfactory periphery and the brain, prompting the need for mechanistic models that can explain the widespread anosmia in COVID-19 patients. Starting from work identifying the non-neuronal cell types that are infected by SARS-CoV-2 in the olfactory system, we review the effects of infection of these supportive cells in the olfactory epithelium and i

Primary olfactory neurons that express the same odorant receptor are distributed mosaically throughout the olfactory neuroepithelium lining the nasal cavity, yet their axons converge and form discrete glomeruli in the olfactory bulb. We previously proposed that cell surface carbohydrates mediate the sorting out and selective fasciculation of primary olfactory axons en route to glomeruli. If this were the case, then axons that terminate in the same glomerulus would express the same complement of cell surface carbohydrates. In this study, we examined the expression of a novel carbohydrate (NOC-3

The olfactory system is a highly plastic region of the nervous system. Continuous remodeling of neuronal circuits in the olfactory bulb takes place throughout life as a result of constant turnover of primary sensory olfactory neurons in the periphery. Glycoconjugates are very important in olfactory development, regeneration and function. This article deals with different aspects of glycobiology relevant for the olfactory system. Various anatomical, developmental and functional subdivisions of the olfactory system have been labeled with exogenous lectins. The application of reverse lectin histo

Continuous replacement of sensory neurons is a normal process in the olfactory system of adult vertebrates. The capacity for replacement following experimentally induced degeneration of sensory neurons makes this system ideal for the study of the dynamics of neural populations. A quantitative analysis was made to determine the time course and degree of cell replacement in the olfactory epithelium following unilateral bulbectomy in adult hamsters. Histological measurements of number of cells and epithelial thickness were made for up to 194 days postoperatively. Results for each experimental ani

When inhaled, environmental toxins enter not only the respiratory system but also the olfactory system. Metallic compounds in the environment form various free radicals through Fenton reactions and Fenton-like reactions. These free radicals damage the olfactory system in various ways. Reactive oxygen species generated during Fenton and Fenton-like reactions damage olfactory G protein-coupled receptors, disrupting the receptor's coupling to GOₗf and cAMP- and IP3/Ca2+-dependent signaling. The cascade of reactions involves ion channels, mitochondria, enzymes, lipids, proteins, DNA, and antioxida

Five patients suffering post-traumatic anosmia were studied at the University of Colorado Health Sciences Center, Denver. Each patient underwent psychophysical testing, clinical evaluation, and olfactory biopsy. The biopsy specimens were examined ultrastructurally and were found to vary from normal tissues. The overall appearance of the olfactory epithelium in the post-traumatic patient is disrupted and the receptor cells are distorted. Large numbers of axons are located near the basement membrane and can often be found in bundles throughout the epithelium, extending even to the mucosal surfac

BACKGROUND: Understanding viral infection of the olfactory epithelium is essential because the olfactory nerve is an important route of entry for viruses to the central nervous system. Specialized chemosensory epithelial cells that express the transient receptor potential cation channel subfamily M member 5 (TRPM5) are found throughout the airways and intestinal epithelium and are involved in responses to viral infection.
RESULTS: Herein we performed deep transcriptional profiling of olfactory epithelial cells sorted by flow cytometry based on the expression of mCherry as a marker for olfacto

This review highlights the crucial role of olfaction in regulating energy homeostasis. Neurons in the olfactory system stimulate the limbic and hedonic pathways involved in food-related behaviors. These neurons are modulated by the body's energy metabolism status, which they, in turn, help to regulate. Olfactory sensitivity is reduced in obesity, while bariatric surgery restores olfactory function. In type 2 diabetes, the onset of olfactory dysfunction is predictive of cognitive decline. Finally, in a mouse model of obesity, local activation of glucagon-like peptide 1 (GLP-1) and its receptors

The main olfactory bulb (MOB) is the first site of synaptic processing in the central nervous system for odor information that is relayed from olfactory receptor neurons in the nasal cavity via the olfactory nerve (ON). Glutamate and ionotropic glutamate receptors (iGluRs) play a dominant role at ON synapses. Similarly, glutamate and iGluRs mediate dendrodendritic transmission between several populations of neurons within the MOB network. Neuroanatomical studies demonstrate that metabotropic glutamate receptors (mGluRs) are densely expressed through the MOB network, and they are particularly a…

Chemosensory neuron diversity in C. elegans arises from the action of transcription factors that specify different aspects of sensory neuron fate. In the AWB and AWA olfactory neurons, the LIM homeobox gene lim-4 and the nuclear hormone receptor gene odr-7 are required to confer AWB and AWA-specific characteristics respectively, and to repress an AWC olfactory neuron-like default fate. Here, we show that AWA neuron fate is also regulated by a member of the LIM homeobox gene family, lin-11. lin-11 regulates AWA olfactory neuron differentiation by initiating expression of odr-7, which then autor

Mosquito behavior is heavily dependent on olfactory and mechanosensory cues, which are detected by receptor neurons on the antenna and on the palps. Recent progress in mosquito sensory genomics highlights the need for an up-to-date understanding of the neural architecture of the mosquito brain. Here we present a detailed description of the neural structure of the primary target of the majority of these neurons, the deutocerebrum, in the African malaria (Anopheles gambiae) and yellow fever (Aedes aegypti) mosquitoes. Special focus is made on the olfactory system, the antennal lobe (AL), where w
