Efficacy of amine-based disinfectant KENO™COX on the infectivity of Cryptosporidium parvum oocysts.

Cryptosporidium parvum is a zoonotic protozoan parasite that may cause severe neonatal diarrhoea or even mortality in newborn ruminants: its oocysts are extremely resistant to normal environmental conditions and to most common disinfectants. KENO™COX, a patent pending amine-based formula, was tested for its ability to inactivate C. parvum oocysts. The Daugschies assay (2002), a standardized assay for chemical disinfection initially described for Eimeria spp., was adapted for C. parvum oocysts. KENO™COX diluted in water at 2% and 3% concentration and incubated with oocyst suspensions for 2h, al…

Detection of Cryptosporidium oocysts in green mussels (Perna viridis) from shell-fish markets of Thailand.

Mussels filter large volumes of water and can concentrate pathogenic organisms, which may act as potential vehicles of transmission to the c … significant economical impact if the mussels' cultures are found contaminated. Cryptosporidium spp. oocysts were detected by the immunofluorescence antibody method (IFA) in 12.5% of the samples examined. The detection of Cryptosporidium oocysts in green mussels' population of Samut Prakan was higher (15.6%) than in Bangkok market (8.3%). These differences …

Effect of tillage and rainfall on transport of manure-applied Cryptosporidium parvum oocysts through soil.

Most waterborne outbreaks of cryptosporidiosis have been attributed to agricultural sources due to the high prevalence of Cryptosporidium oocysts in animal wastes and manure spreading on farmlands. No-till, an effective conservation practice, often results in soil having higher water infiltration and percolation rates than conventional tillage. We treated six undisturbed no-till and six tilled soil blocks (30 by 30 by 30 cm) with 1 L liquid dairy manure containing 10(5) C. parvum oocysts per milliliter to test the effect of tillage and rainfall on oocyst transport. The blocks were subjected to…

Genotypic characterization of cryptosporidium oocysts isolated from healthy people in three different counties of Korea.

To investigate Cryptosporidium infection among healthy people, we collected stool samples from 150 healthy individuals in Gokseong, Muan, and Imshil Counties, southwest Korea, where neighbors on both an animal farm and a river respectively. In 12 of 150 samples, Cryptosporidium oocysts were detected by means of modified acid-fast staining. The bovine genotype, Cryptosporidium parvum, was identified by PCR/RFLP and 18S rRNA sequencing. C. parvum existed endemically in these areas, and the residents showed a relatively higher infection rate for C. parvum than that for C. hominis. Our results ind…

Optimization of DNA extraction and molecular detection of Cryptosporidium oocysts in natural mineral water sources.

The numerous published methods for extracting DNA from Cryptosporidium oocysts for PCR identify the lack of an optimized standard method for clinical, environmental, and public health investigations of cryptosporidiosis. A method that maximizes DNA extraction reliably, particularly from small numbers of partially purified or purified oocysts present in mineral waters and environmental samples, is required. We describe a maximized method for liberating DNA from Cryptosporidium parvum oocysts by 15 cycles of freezing (liquid nitrogen) and thawing (65 degrees C) in lysis buffer containing sodium …

Recovery of waterborne oocysts of Cryptosporidium from water samples by the membrane-filter dissolution method.

The cellulose-acetate membrane (CAM)-filter dissolution method implemented into a Millipore Glass Microanalysis system was used for recovery of Cryptosporidium parvum oocysts seeded into 25 l of drinking water in polyethylene carboy aspirator bottles. CAM-entrapped oocysts were detected by immunofluorescence microscopy. From 65 to 94 oocysts/l (mean 75 oocysts/l), 34.7% overall of the inoculated oocysts, were unrecovered as determined after the water had been drained from the bottle, rinsed with 1 l of eluting fluid (EF), and CAM-filtered. Efficiency rates of oocyst recovery ranged from 24.0% …

Assessment of the conventional detection of fecal Cryptosporidium serpentis oocysts in subclinically infected captive snakes.

Fecal specimens of seven captive snakes naturally subclinically infected with Cryptosporidium serpentis were monitored for 131 days for the presence and concentration of oocysts. Thirty-three of 81 (41%) of the monoclonal antibody positive stools were read as negative (sensitivity 59%) by the acid-fast-stained (AFS) fecal smears. Oocyst concentrations in the false-negative stools ranged between 6.0 x 10(3)/g to 2.5 x 10(4)/g. The experimentally determined concentration-threshold of oocyst detection by AFS fecal smear was 3.0 x 10(4)/g. The stool weights did not conform to a normal distribution…

Simplified methods for obtaining purified oocysts from mice and for growing Cryptosporidium parvum in vitro.

Seven- to 8-day-old Arc/Swiss mice were infected with 100,000-120,000 Cryptosporidium parvum oocysts. At 8 days postinfection (PI) the jejunum, ileum, cecum, colon, and rectum were removed. Using a simple extraction procedure and purification by Ficoll gradient centrifugation, we rountinely obtained between 3-6 million and up to 15 million purified oocysts per mouse. For in vitro cultivation, purified oocysts were pretreated in a low pH (2.5-3) 0.5% trypsin solution for 20 min, resuspended in supplemented RPMI-1640 containing glucose 0.1 g (5.55 mM), sodium bicarbonate 0.3 g, bovine bile 0.02 …

Cryptosporidium oocysts in immunocompetent children: epidemiologic investigations in the day-care centers of Poitiers, France.

The recent discovery of self-resolving Cryptosporidium infection in immunocompetent patients has aroused growing interest in this parasite, which has thus far been known to affect mainly immunodeficient individuals. Following the hospitalization of two children attending the same day-care center in February 1986 Cryptosporidium oocysts were found in the stools of one of them. This led to an epidemiological investigation to assess the frequency of Cryptosporidium oocysts in the stools of children attending the 7 day-care centers in the city of Poitiers, France. Testing for Cryptosporidium oocys…

Oocyst wall protein EtOWP6 is critical for sporocyst formation and developmental progression in Eimeria tenella.

Transmission of apicomplexan parasites-including Eimeria, Toxoplasma, and Cryptosporidium-relies on the environmentally resilient oocyst wall, a bilayered structure whose integrity is maintained by cysteine-rich oocyst wall proteins (OWPs). Yet the functional roles of these conserved proteins remain poorly defined. Here, we characterize EtOWP6, a conserved OWP in Eimeria tenella, the causative agent of avian coccidiosis. Immunofluorescence reveals that EtOWP6 localizes specifically to wall-forming bodies and oocyst/sporocyst walls during sexual development. CRISPR/Cas9-mediated knockout demons

Coccidiosis in chickens: From temporal patterns of Lesion development and Oocyst shedding to predictive modeling.

Coccidiosis continues to be an impactful parasitic disease in the poultry industry, and the increasing resistance to conventional anticoccidials such as ionophores and chemicals underscores the necessity of a deeper understanding in the behavior of this parasite. These experiments aimed to evaluate the distribution of lesion scores, oocysts shedding, growth performance, and to develop coccidiosis prediction models in broiler chickens undergoing a coccidiosis challenge and supplemented or not with ionophores or chemical/ionophore blend. Three experiments were conducted with 1,200 (Exp. 1) and 8…

Eimeria acervulina is a promising surrogate for Toxoplasma gondii oocysts exposed to chemical and physical treatments.

Oocysts of the coccidian parasite Toxoplasma gondii withstand a wide range of chemical and physical factors, contributing to their food- and water-borne transmission to humans worldwide. Assessing the efficacy of processes to remove or inactivate oocysts at a pilot or industrial scale encounters major ethical, economic, and methodological constraints. The coccidian parasite Eimeria acervulina has been proposed as a non-human pathogenic alternative of T. gondii to assess food decontamination, however it is not known whether the two parasites exposed to chemical and thermal treatments parallel i…

Quantitative Risk Assessment of Oocyst Versus Bradyzoite Foodborne Transmission of Toxoplasma gondii in Brazil.

Toxoplasma gondii is a globally distributed zoonotic protozoan parasite. Infection with T. gondii can cause congenital toxoplasmosis in developing fetuses and acute outbreaks in the general population, and the disease burden is especially high in South America. Prior studies found that the environmental stage of T. gondii, oocysts, is an important source of infection in Brazil; however, no studies have quantified this risk relative to other parasite stages. We developed a Bayesian quantitative risk assessment (QRA) to estimate the relative attribution of the two primary parasite stages (bradyz…

Development of a Coccidiosis Disease Challenge Model Using a Commercially Available Live Oocyst Vaccine.

With growing cross-disciplinary collaboration among researchers, it is increasingly important to record detailed methodology to prevent the repetition of preliminary experiments. The purpose of this paper is to explain the development of a coccidiosis challenge model for the investigation of dietary interventions to coccidiosis in broiler chickens. The objectives are to select a dose of mixed species coccidial vaccine and evaluate the suitability (ability to produce a consistent, marked change) of selected response variables important to nutritional studies at different times postinfection (PI

Evaluation of quantitative polymerase chain reaction for the detection of Toxoplasma gondii oocysts shed by cats.

Felines are definitive hosts of Toxoplasma gondii and can shed oocysts in their feces, contaminating the environment. Sporulated oocysts are highly resistant to the environment and have higher infectivity, which are attributed to many toxoplasmosis outbreaks. The aim of the present study was to evaluate a quantitative polymerase chain reaction (qPCR) technique for the detection of T. gondii oocysts shed by cats. Twelve cats from a previous vaccine experiment were challenged orally with 600 cysts of the TgDoveBr8 strain on day 72. Fecal samples were collected daily using the centrifugal flotati…

Detection and removal of Giardia spp. cysts and Cryptosporidium spp. oocysts by anaerobic reactors in Brazil.

The discharge of raw wastewater into the environment can be a contamination source of Giardia spp. cysts and Cryptosporidium spp. oocysts. The UASB (Upflow Anaerobic Sludge Blanket) reactor is the most popular technology applied for wastewater treatment in Brazil, nevertheless there is little information concerning its capacity for (oo)cyst removal. In this context, this study investigated the occurrence and removal of Giardia spp. cysts and Cryptosporidium spp. oocysts by three different UASB reactors (i.e. Reactor A, B, and C) treating different wastewater types. In the wastewater influent, …

Identification of a Cyclospora cayetanensis Oocyst Antigens and Their Validity in the Detection of Immunogenic Patterns of Cyclosporiasis Patients.

INTRODUCTION: The diagnosis of cyclosporiasis is currently based on the microscopic detection of oocysts, which may provide invalid results. The availability of simple, objective immunological screening tests would facilitate epidemiological studies of cyclosporiasis. Therefore, the present study aimed to identify the antigens of Cyclospora cayetanensis oocysts and their validity in serodiagnosis. METHODS: According to parasitological and molecular diagnoses, three study groups were specified. Group (G) I included 30 patients with cyclosporiasis, GII included 12 patients with other parasitic …

Evaluation of Different Oocyst DNA Extraction Methods for Cryptosporidium spp. Research in Environmental Samples.

PURPOSE: The major problem of the PCR method for the search of protozoan cysts/oocysts in environmental samples is the presence of inhibitors. DNA extraction methods capable of removing inhibitory substances of environmental origin and recovering the DNA are decisive for the efficiency of PCR. This study aimed to compare the efficiency of different DNA extraction methods for the search by Cryptosporidium oocysts in water samples by molecular methods. METHODS: DNA extraction from water samples was performed using four different methods. Two methods use a chaotropic buffer to extract DNA and pr…

Comparison of Cryptosporidium oocyst recovery methods for their applicability for monitoring of consumer-ready fresh shellfish.

Growing demand for fresh, unprocessed food favours the emergence of Cryptosporidium infections in humans. Mainly it is food of plant origin or unpasteurized milk which have been involved in food-borne outbreaks of cryptosporidiosis. So far consumption of shellfish contaminated with Cryptosporidium were not associated with human infections although such as possibility exists. In this study an attempt was undertaken to evaluate the analytical performance of three commonly used methods for recovery of Cryptosporidium oocysts from shellfish: i) pepsin digestion of shellfish in conjunction with imm…
