Screening and identification of a novel PGAM5-specific inhibitor for attenuating multi-organ injury.

INTRODUCTION: The necroptosis regulator PGAM5 drives a pathological cycle of mitochondrial dysfunction and necroptotic signaling, contributing to multi-organ injury and representing a potential therapeutic target. Despite its clinical relevance, few PGAM5-specific small-molecule inhibitors have been developed. OBJECTIVES: We aimed to identify a safe and effective natural small-molecule inhibitor targeting PGAM5 as a novel therapeutic strategy. METHODS: Global PGAM5 knockout mice and pancreas-specific PGAM5 knockdown mice were used to clarify the regulatory role of PGAM5 in pancreatic injury

Cellular source and circuit context organize functional specificity in the Drosophila NPF system.

Neuropeptides regulate diverse and sometimes opposing functions, yet how a single peptide achieves functional specificity remains unclear. Receptor diversity can provide specificity, but not in systems with only one receptor. Drosophila neuropeptide F (NPF) acts through a single receptor, providing a model to test whether specificity is organized by the peptide's cellular source. Using the whole-brain connectome, we defined four adult NPF cell types with no shared first-order synaptic partners and separate networks. These cell types made distinct and partly opposing contributions to reinforcem

Division-Specific Organization of a Shared Functional Scaffold in the Early-Life Human Brain.

Early cognition and behavior emerge from coordinated maturation of functional systems spanning the brain, making cross-division integration essential for defining whole-brain architecture and understanding its role in neurodevelopment. Yet early human functional development is still largely investigated through cortical networks or through isolated, coarsely resolved noncortical structures, leaving unclear how this integrated architecture is organized across the cerebral cortex, subcortex and cerebellum. Moving beyond structure-isolated and adult-derived maps, we create a reproducible, fine-gr

Acute Dehydration Drives Organ-Specific Modulation of Phosphorylated AQP4ex in Brain and Kidney.

Water deprivation triggers coordinated physiological responses to preserve body fluid balance, yet the molecular mechanisms that regulate aquaporin-mediated water transport under dehydration remain incompletely understood. Aquaporin-4 (AQP4), the main water channel in the brain and a basolateral water pathway in the kidney collecting duct, exists in multiple isoforms, including the translational readthrough variant AQP4ex, whose regulatory role is only beginning to be defined. Here, we investigated the effects of acute water deprivation (6-12 h) on AQP4 isoform expression and phosphorylation i

Distinct glycosyltransferase expression governs organ-specific survival in pancreatic cancer metastasis.

Pancreatic ductal adenocarcinoma (PDAC) is among the most lethal cancers, with over 83% of patients presenting metastases to vital organs such as the liver, lungs, and peritoneum. Although glycosylation has been established as a critical factor in PDAC development, little is known about the molecular underpinnings of organ-specific metastasis. In this study, we investigated the role of glycosyltransferases (GTs) in mediating PDAC metastatic organotropism. Through an unbiased transcriptomic screen, we identified distinct GT expression patterns in liver- and lung-tropic PDAC cells. Notably, GCNT

Mice Undergo Organ-Specific Transcriptional and Metabolic Remodeling During Late Adolescence.

Mouse models aimed at deciphering human aging have typically focused on early development or late-life stages. Much less is known about the transition from late adolescence to young adulthood, spanning 8 to 12 weeks of age. Here, we show that C57Bl6 mice undergo a comprehensive developmental transition characterized by significant transcriptional and metabolic reprogramming during this time. Transcriptional analysis revealed substantial, organ-specific changes: the heart shifts toward a maintenance-focused phenotype with down-regulated oxidative phosphorylation (OXPHOS) and increased expressio

Organ-specific transcriptional programs regulate cellular plasticity in lymphatic endothelial cells.

Lymphatic vessels play essential roles in maintaining fluid homeostasis and responding to inflammatory and tissue environmental changes. During these biological processes, lymphatic endothelial cells (LECs) lining lymphatic vessels adapt to external stimuli in various organs and alter their characteristics. While organ-specific molecular and functional heterogeneity has been well-characterized in blood vascular endothelial cells, the extent and functional significance of such heterogeneity in LECs remain poorly understood. In this study, we established LECs derived from the heart and lung tiss

Comprehensive metabolomics study identifies SN-38 organ specific toxicity in mice.

SN-38 (7-ethyl-10-hydroxycamptothecin), the active metabolite of irinotecan, is a crucial anticancer agent frequently studied in drug delivery systems. Irinotecan (CPT-11) is used to treat various solid tumors but is associated with adverse effects such as nausea, vomiting, diarrhea, and steatohepatitis. However, the precise biochemical pathways underlying these side effects remain unclear. To explore SN-38's toxic mechanisms and provide insights for clinical applications of SN-38 delivery systems, we performed untargeted metabolomics to assess metabolic changes in the lungs, heart, stomach, b

Patient-specific organ dose and in-vivo image quality assessment in clinical CT.

PURPOSE: To develop and characterize individualized dose and quality measures at organ level compared to their generic counterparts across a clinical CT dataset. MATERIALS AND METHODS: The study included 9801 chest-abdomen-pelvis and abdomen-pelvis CT exams (7,763 patients, mean age, 56 ± 17 years; 4113 women) representing 20 unique protocols. For each exam, patient-specific organ dose of all radiosensitive organs was estimated using a validated method by generating personalized computational phantoms and Monte Carlo simulations. Effective dose (EOD) was calculated by weighted sum of the orga

The Burden of Non-Infectious Organ-Specific Immunopathology in Pediatric Common Variable Immunodeficiency.

The pediatric common variable immunodeficiency (CVID) is the most frequent symptomatic antibody production defect characterized by infectious and non-infectious autoimmune, inflammatory, and lymphoproliferative complications. The background for CVID-related organ-specific immunopathology is associated with immune dysregulation and immunophenotypic biomarkers with expansion of CD21low B cells, and dysfunctional memory B cell, follicular T cell, and regulatory T cell compartments. The ever-increasing progress in immunogenetics shows the heterogeneity of genetic background for CVID related to the

A Comparison of a Solid Organ Transplant Population-Specific Antibiogram With the Hospital-Wide Antibiogram.

BACKGROUND: Solid organ transplant (SOT) recipients are vulnerable to infections with multidrug-resistant organisms, and they often do not receive adequate empiric antimicrobials for serious Gram-negative infections. Knowledge of differences in antimicrobial resistance rates in this specific population can help guide empiric antimicrobial choices. METHODS: We performed a retrospective cohort study comparing antimicrobial susceptibility patterns of Gram-negative organisms isolated from the adult SOT recipients with documented sepsis with the hospital-wide inpatient antibiogram. To evaluate emp

Organ-specific tumor dynamics predict survival of patients with metastatic colorectal cancer.

BACKGROUND: We aim to compare the prognostic value of organ-specific dynamics with the sum of the longest diameter (SLD) dynamics in patients with metastatic colorectal cancer (mCRC). METHODS: All datasets are accessible in Project Data Sphere, an open-access platform. The tumor growth inhibition models developed based on organ-level SLD and SLD were used to estimate the organ-specific tumor growth rates (KGs) and SLD KG. The early tumor shrinkage (ETS) from baseline to the first measurement after treatment was also evaluated. The relationship between organ-specific dynamics, SLD dynamics, an

Identification, Organic Synthesis, and Sensitive Analysis of a cis-Homosalate-Specific Exposure Biomarker.

Homosalate (HMS) is an organic UV filter used in sunscreens and personal care products. Despite its widespread use and detection in environmental matrices, little is known regarding its exposure in humans. HMS is used as a mixture of cis- and trans-isomers, and we recently revealed major differences in human toxicokinetics, indicating the need to consider these isomers separately in exposure and risk assessments. In the course of these previous investigations of human HMS toxicokinetics, we identified two trans-HMS-specific and one cis-HMS-specific biomarker candidates. However, the latter lac

Rapid estimation of patient-specific organ doses using a deep learning network.

BACKGROUND: Patient-specific organ-dose estimation in diagnostic CT examinations can provide useful insights on individualized secondary cancer risks, protocol optimization, and patient management. Current dose estimation techniques mainly rely on time-consuming Monte Carlo methods or/and generalized anthropomorphic phantoms. PURPOSE: We proposed a proof-of-concept rapid workflow based on deep learning networks to estimate organ doses for individuals following thorax Computed Tomography (CT) examinations. METHODS: CT scan data from 95 individuals undergoing thorax CT examinations were used.

Toward Patient-specific Pessary to Manage Pelvic Organ Prolapse: Design and Simulation.

This study proposed a novel design and personalized approach to developing an intra-vaginal device, also known as a pessary, for the treatment of Pelvic Organ Prolapse (POP). Although POP is likely to have a more diverse dynamic than other health conditions in women, it is currently treated as a "one-shape-fits-all" problem in all cases. Pessaries are conservative devices inserted into the vagina to support its internal structure and predominantly come in a ring shape design. Failure rates as high as 50% within the first year of use have been attributed to the poor design of these pessaries; w

Organ-specific or personalized treatment for COVID-19: rationale, evidence, and potential candidates.

Although extrapulmonary manifestations of coronavirus disease 2019 (COVID-19) are increasingly reported, no effective therapeutic strategy for these multisystemic complications is available due to a poor understanding of the pathophysiology of COVID-19 multiorgan involvement. In this study, differentially expressed genes (DEGs) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-infected extrapulmonary organs including human pluripotent stem cells (hPSCs)-derived liver organoids and choroid plexus organoids besides transformed lung alveolar (A549) cells were analyzed. Hyperoxia Reprogrammes Microvascular Endothelial Cell Response to Hypoxia in an Organ-Specific Manner.

Organ function relies on microvascular networks to maintain homeostatic equilibrium, which varies widely in different organs and during different physiological challenges. The endothelium role in this critical process can only be evaluated in physiologically relevant contexts. Comparing the responses to oxygen flux in primary murine microvascular EC (MVEC) obtained from brain and lung tissue reveals that supra-physiological oxygen tensions can compromise MVEC viability. Brain MVEC lose mitochondrial activity and undergo significant alterations in electron transport chain (ETC) composition when
