Significance of maternal uterine proteins in the establishment of pregnancy.

Mammalian embryonic development depends on extrinsic support by the maternal organism. During the preimplantation period the growing blastocyst needs favourable environmental conditions, which are provided by endometrial transformations and secretion. The macromolecular composition of uterine secretion at different times before implantation is characterized by a spectrum of protein patterns which change daily. In an attempt to shed light on the significance of the uterine secretion proteins, particularly uteroglobin in the rabbit, in the establishment of early pregnancy, analyses are reported

Regulation of embryonic implantation.

The preimplantation embryo produces several factors during its development to signal its presence to the maternal organism. This paper will focus on the role of two distinctive cytokine and growth factor systems (interleukin-1 (IL-1) system and the vascular endothelial growth factor (VEGF) system) during early embryonic development and implantation. IL-1 receptor is expressed in the endometrium of various species and antagonising the biological effects of IL-1 leads to implantation failure in mice. We could show that this is due to an endometrial, not an embryonic effect. Furthermore, we could

Three-dimensional reconstruction of the embryonic pancreas in the grass snake Natrix natrix L. (Lepidosauria, Serpentes) based on histological studies.

The aim of this study was to evaluate two research hypotheses: H0-the embryonic pancreas in grass snakes develops in the same manner as in all previously investigated amniotes (from three buds) and its topographical localization within the adult body has no relation to its development; H1-the pancreas develops in a different manner and is related to the different topography of internal organs in snakes. For the evaluation of these hypotheses we used histological methods and three-dimensional (3D) reconstructions of the position of the pancreatic buds and surrounding organs at particular develo

Embryonic stem cell therapy of heart failure in genetic cardiomyopathy.

Pathogenic causes underlying nonischemic cardiomyopathies are increasingly being resolved, yet repair therapies for these commonly heritable forms of heart failure are lacking. A case in point is human dilated cardiomyopathy 10 (CMD10; Online Mendelian Inheritance in Man #608569), a progressive organ dysfunction syndrome refractory to conventional therapies and linked to mutations in cardiac ATP-sensitive K(+) (K(ATP)) channel subunits. Embryonic stem cell therapy demonstrates benefit in ischemic heart disease, but the reparative capacity of this allogeneic regenerative cell source has not bee

Retroperitoneal tumors and lymphadenopathy.

The retroperitoneum is the space between the parietal peritoneum anteriorly and the transversalis fascia posteriorly. Most retroperitoneal tumors arise from mesenchyme, neurogenic tissue, or embryonic rests. By this definition and by including lymph nodes as a retroperitoneal organ, lymphoma is not considered a primary retroperitoneal tumor.

Molecular and developmental consequences of heat stress on the bovine oocyte and embryo competence.

Seasonal heat stress (HS) is a pervasive environmental challenge with profound consequences for female reproductive physiology, affecting ovarian function, oocyte maturation, and early embryonic development. At the ovarian level, HS disrupts follicular growth, impairs steroidogenesis, and compromises granulosa cell function, thereby creating a suboptimal microenvironment that reduces oocyte competence. In oocytes, HS induces oxidative stress, mitochondrial dysfunction, endoplasmic reticulum (ER) stress, spindle abnormalities, chromosomal missegregation, and persistent epigenetic alterations. T

From Static to Dynamic: Fluorescence Imaging Technology Advances Precise Embryo Evaluation.

Live-cell imaging technology has revolutionized our understanding of preimplantation embryonic development, shifting the field from static morphological descriptions to dynamic functional analyses. This has tremendously advanced the fields of in vitro fertilization (IVF) and embryonic development. At the heart of this transition lies the strategic application of fluorescent probes, which provide the requisite sensitivity and specificity to resolve complex biological events. This review provides a comprehensive overview of fluorescent probe-based strategies designed to address four cardinal que

3-Hydroxyacyl CoA Dehydratase 2 Is Essential for Embryonic Development and Hepatic Metabolic Function Under a Low-Fat, High-Carbohydrate Diet.

The conversion of carbohydrates into fatty acids is central for energy storage and the development and functioning of organs. Our previous study revealed that Hacd2 deficiency alleviates the fatty liver and diabetes induced by HFD. This study aimed to explore the roles of Hacd2 in organ development and metabolic homeostasis under an LFHCD, which still need to be more deeply explored. We found that the germline deletion of Hacd2 impairs long-chain fatty acid synthesis, which caused embryonic abnormalities after 7.5 days and led to embryonic lethality, as confirmed via photograph and hematoxylin

Multiplication of influenza A viruses with cleavable and non-cleavable haemagglutinin in chicken embryo membranes or organs, and cell cultures derived therefrom.

Pathogenic properties of influenza A viruses introduced into embryonated chicken eggs via the allantoic cavity, the amniotic cavity or the yolk sac were studied using viruses with cleavable or non-cleavable haemagglutinin (HA), or reassortants derived from the highly pathogenic fowl plague virus (FPV) which has a cleavable HA. The various organs, membranes and fluids were analysed for virus yields, and by immunohistochemistry for production of viral nucleoprotein. Virus replication in primary tissue cultures derived from various embryonic organs was also studied. Some of the reassortants, whic

Lymphatics-epicardial cross talk via Reelin controls cardiac growth.

How mammalian organ size is precisely regulated remains incompletely understood. This process depends on the coordinated balance between cell proliferation, differentiation, and apoptosis. In the heart, the epicardium plays a central role in embryonic cardiac growth as a source of multiple cardiac progenitor lineages. The transcription factor Wt1 is essential for epicardial development and maturation, and its loss disrupts epicardial maintenance and the generation of epicardial-derived cells (EPDCs). In addition, the epicardium produces mitogenic factors required for cardiomyocyte (CM) prolife

The mechanism and effects of remdesivir-induced developmental toxicity in zebrafish: Blood flow dysfunction and behavioral alterations.

The antiviral drug remdesivir has been used to treat the growing number of coronavirus disease 2019 (COVID-19) patients. However, the drug is mainly excreted through urine and feces and introduced into the environment to affect non-target organisms, including fish, which has raised concerns about potential ecotoxicological effects on aquatic organisms. Moreover, studies on the ecological impacts of remdesivir on aquatic environments have not been reported. Here, we aimed to explore the toxicological impacts of microinjection of remdesivir on zebrafish early embryonic development and larvae and

Mechanisms underlying pre- and postnatal development of the vomeronasal organ.

The vomeronasal organ (VNO) is sensory organ located in the ventral region of the nasal cavity in rodents. The VNO develops from the olfactory placode during the secondary invagination of olfactory pit. The embryonic vomeronasal structure appears as a neurogenic area where migratory neuronal populations like endocrine gonadotropin-releasing hormone-1 (GnRH-1) neurons form. Even though embryonic vomeronasal structures are conserved across most vertebrate species, many species including humans do not have a functional VNO after birth. The vomeronasal epithelium (VNE) of rodents is composed of tw

p53, stem cell biology and childhood blastomas.

PURPOSE OF REVIEW: Childhood blastomas, unlike adult cancers, originate from developing organs in which molecular and cellular features exhibit differentiation arrest and embryonic characteristics. Conventional cancer therapies, which rely on the generalized cytotoxic effect on rapidly dividing cells, may damage delicate organs in young children, leading to multiple late effects. Deep understanding of the biology of embryonal cancers is crucial in reshaping the cancer treatment paradigm for children. RECENT FINDINGS: p53 plays a major physiological role in embryonic development, by controllin

Immunolocalization of glutaryl-CoA dehydrogenase (GCDH) in adult and embryonic rat brain and peripheral tissues.

Glutaryl-CoA dehydrogenase (GCDH) is a mitochondrial enzyme that is involved in the degradation of tryptophan, lysine and hydroxylysine. Deficient enzyme activity leads to glutaric aciduria type-I (GA-I). This neurometabolic disease usually manifests with acute encephalopathic crises and striatal neuronal death in early childhood leading to an irreversible dystonic-dyskinetic movement disorder. Fronto-temporal atrophy and white matter changes are already present in the pre-symptomatic period. No detailed information on GCDH expression during embryonic development and in adulthood was available

The isolation of enteroviruses from cases of acute conjunctivitis.

In two epidemics of acute enterovirus conjunctivitis the diagnosis could only be established by the use of human embryonic organ cultures. In one instance cultures of ciliated respiratory epithelium proved sensitive to the virus but similar culture failed to support growth of the virus from the second outbreak which was isolated in organ cultures of conjunctiva. The virus associated with a third outbreak had previously been isolated from the original material by culture in human embryo kidney cells, but it required passage in organ culture for its detection after the specimens had been diluted

Donor MHC-specific thymus vaccination allows for immunocompatible allotransplantation.

Organ transplantation is the last-resort option to treat organ failure. However, less than 10% of patients benefit from this only option due to lack of major histocompatibility complex (MHC)-matched donor organs and 25%-80% of donated organs could not find MHC-matched recipients. T cell allorecognition is the principal mechanism for allogeneic graft rejection. We herein present a "donor MHC-specific thymus vaccination" (DMTV) strategy to induce T cell tolerance to both autologous and allogeneic donor MHC. Allogeneic MHC molecules were expressed in the recipient thymus through adeno-associated

Engineered Development: Directed Morphogenesis of an Embryonic Heart Tube.

Morphogenesis is a major developmental process that shapes organs in utero through coordinated structural deformations. Outside of simple cellular or animal models, limited models of morphogenesis exist due to the complexity of hierarchical structural change. In development, the embryonic heart undergoes a complex morphogenesis process involving looping, chamber growth, and septation, transforming the primordial tube to an adult four-chambered organ. An in vitro model of looping could provide unique insights into this phase of development and improve understanding of congenital defect generati

Zebrafish ventral gastrula fate map reveals neural crest progenitor domain.

BACKGROUND: Fate maps relate progenitor cell positions to later fates and locations of their progeny, revealing early embryonic organization. Previous zebrafish fate maps identified the origin of germ layers and derivative cell fates, but ventral gastrula progenitor domains were not fully resolved. In particular, the neural crest, a multipotent cell type central to vertebrate development and evolution, was not characterized in the zebrafish fate map. RESULTS: We generated a region-based fate map to resolve the origin of the neural crest and overall organization of the ventral gastrula. This f

The role of FGF15/FGF19 in the development of the central nervous system, eyes and inner ears in vertebrates.

Fibroblast growth factor 19 (FGF19), and its rodent ortholog FGF15, is a member of a FGF subfamily directly involved in metabolism, acting in an endocrine way. During embryonic development, FGF15/FGF19 also functions as a paracrine or autocrine factor, regulating key events in a large number of organs. In this sense, the Fgf15/Fgf19 genes control the correct development of the brain, eye, inner ear, heart, pharyngeal pouches, tail bud and limbs, among other organs, as well as muscle growth in adulthood. These growth factors show relevant differences according to molecular structures, signallin

Molecular staging of marine medaka: a model organism for marine ecotoxicity study.

Oryzias melastigma, also called O. dancena, is becoming a very useful model for estuarine and marine ecotoxicity studies. With O. melastigma being adopted by ILSI Health and Environmental Science Institute (HESI) for embryo toxicity testing, improved knowledge of biomarker based embryonic development becomes especially important for mechanism-based toxicity evaluations. Using whole mount in situ hybridization and immunostaining techniques together with widely used molecular markers, this study describes the molecular development of marine medaka embryos, focusing on the brain, eye, heart, pect
