Cytosolic Delivery of Multidomain Cargos by the N Terminus of Pasteurella multocida Toxin.

The zoonotic pathogen Pasteurella multocida produces a 146-kDa modular toxin (PMT) that enters host cells and manipulates intracellular signaling through action on its Gα protein targets. The N terminus of PMT (PMT-N) mediates cellular uptake through receptor-mediated endocytosis, followed by the delivery of the C-terminal catalytic domain from acidic endosomes into the cytosol. The putative native cargo of PMT consists of a 710-residue polypeptide with three distinct modular subdomains (C1-C2-C3), where C1 contains a membrane localization domain (MLD), C2 has an as-yet-undefined function, and

A systemic Pasteurella multocida toxin aggravates cardiac hypertrophy and fibrosis in mice.

Pasteurella multocida toxin (PMT) persistently activates heterotrimeric G proteins of the Gαq/11 , Gα12/13 and Gαi family without interaction with G protein-coupled receptors (GPCRs). We show that PMT acts on heart tissue in vivo and on cardiomyocytes and cardiac fibroblasts in vitro by deamidation of heterotrimeric G proteins. Increased normalized ventricle weights and fibrosis were detected after intraperitoneal administration of PMT in combination with the GPCR agonist phenylephrine. In neonatal rat cardiomyocytes, PMT stimulated the mitogen-activated protein kinase pathway, which is crucia

Pathology of experimental infection by Pasteurella multocida serotype A: 1 in buffalo calves.

Pasteurella multocida serotype A:3 has been mostly implicated in pneumonic pasteurellosis in ruminants. In contrast, our previous studies have reported that both serotypes A:1 and A:3 were responsible for respiratory diseases in cattle and buffaloes. However, the pathology and pathogenesis of P. multocida serotype A:1 (Pm A:1) infection have not been studied in ruminants. In the present study, 12- to 15-week-old buffalo calves (Bubalus bubalis) infected by Pm A:1 had fibrinous and suppurative bronchopneumonia with focal areas of coagulation necrosis typical of pneumonic pasteurellosis. For the

Pasteurella multocida bacteremia in asymptomatic plateletpheresis donors: a tale of two cats.

BACKGROUND: Bacterial contamination of platelet (PLT) concentrates occurs in 1 in 1000 to 1 in 3000 components and has been a leading cause of transfusion-associated morbidity and mortality. Two cases of Pasteurella multocida bacteremia in asymptomatic plateletpheresis donors are reported. Clinical outcomes were profoundly different, emphasizing the importance of robust methods to detect bacterial contamination. CASE REPORTS: The first case occurred before the implementation of bacterial testing of PLTs. A plateletpheresis component was collected from a 70-year-old man and transfused to an 88

Septicaemia and arthritis in pigs experimentally infected with Pasteurella multocida capsular serotype A.

Twenty-five caesarean-derived, colostrum-deprived (CDCD) pigs and 18 specific pathogen-free pigs, aged 8 to 14 weeks, were inoculated intranasally or intratracheally with Pasteurella multocida capsular serotype A, isolated from a severe pneumonic lesion in a growing pig. The pigs were killed for necropsy on day 6 or 14 post-inoculation (PI) or, in the case of the only fatally infected animal, examined at the time of death. One CDCD pig, inoculated intratracheally with 5 ml of a bacterial suspension containing 1.7x10(9) colony-forming-units/ml, died of septicaemia on day 1 PI. Histological lesi

[Experimental affecting of pulmonary clearance of Pasteurella multocida induced pneumonia in swine].

The pulmonary clearance of Pasteurella multocida in weaner pigs was not affected by Carrageenan and Silica, two substances which block the function of monocytes/macrophages. Neutropenia, caused by the application of hydroxyurea, however, inhibit the pulmonary clearance markedly and reduced the severity of simultaneously induced pneumonias considerably. This indicates the importance of polymorphonuclear neutrophils for early clearance mechanisms and their inflammation inducing and maintaining effects.

Serological types of Pasteurella multocida isolated from turkeys and chickens in Canada.

Outbreaks of fowl cholera continue to plague the Canadian poultry industry despite widespread immunization against the causative agent, Pasteurella multocida. Fowl cholera bacterins currently employed by domestic poultry growers contain three serological types, namely, serotypes 1, 3, and 4. In this study a total of 84 strains of P. multocida were isolated in Canada from outbreaks of fowl cholera in turkeys and chickens. Serotyping was accomplished using the gel diffusion precipitin test. Based on the gel diffusion precipitation patterns, 27 serotypes containing one to six antigenic determinan

Cepharanthine as a potential Fis inhibitor enhances Enrofloxacin efficacy against Pasteurella multocida.

BACKGROUND: Pasteurella multocida, a primary bacterial pathogen responsible for bovine respiratory diseases, has developed resistance to multiple antibiotics, including fluoroquinolones, and is also an important zoonotic pathogen. Previous studies have demonstrated that Fis protein plays a critical role in bacterial virulence and antibiotic resistance formation. In resistant strains, Fis expression is upregulated and deletion of this gene significantly delays the development of fluoroquinolone resistance. Thus, Fis represents a potential target protein of antibiotic resistance inhibitors. RESU

Epididymitis with Pasteurella multocida isolated from two male Japanese Black beef calves.

Two male Japanese Black calves developed an enlarged scrotum and testis. Orchiectomy was performed and pus was collected during surgery. After removal of the testis, bacteriological and histopathological examinations were conducted to investigate the cause and confirm the diagnosis. Based on the results obtained, both cases were diagnosed with epididymitis caused by an infection with Pasteurella multocida. This is the first study to show that P. multocida causes epididymitis in male calves. Further studies are required to clarify the details underlying the infection of calves with P. multocida

Pasteurella multocida activates Rassf1-Hippo-Yap pathway to induce pulmonary epithelial apoptosis.

Pasteurella multocida is an opportunistic zoonotic pathogen that primarily causes fatal respiratory diseases, such as pneumonia and respiratory syndromes. However, the precise mechanistic understanding of how P. multocida disrupts the epithelial barrier in mammalian lung remains largely unknown. In this study, using unbiased RNA-seq analysis, we found that the evolutionarily conserved Hippo-Yap pathway was dysregulated after P. multocida infection. Given the complexity of P. multocida infection associated with lung injury and systemic inflammatory processes, we employed a combination of cell c

IFN-γ promotes PANoptosis in Pasteurella multocida toxin-induced pneumonia in mice.

Interferon-γ (IFN-γ) is a pleiotropic cytokine that regulates diverse biological functions, including modulation of inflammatory response and innate and adaptive immunity. In our study, we found that IFN-γ plays an important role in the regulation of Pasteurella multocida toxin-associated pneumonia. In work described here, we demonstrated that rPMT induced a lethal pneumonia in WT mice and the severity of the pneumonia was substantially alleviated in IFN-γ-deficient mice, IFN-γ deficiency significantly elevated the survival rate and reduced the pathological lesions of the lungs after rPMT chal

Intranasal instillation of Pasteurella multocida lipopolysaccharide in rabbits causes interstitial lung damage.

In order to characterize the in vivo lesions in the nasal cavities and lungs, twenty-eight rabbits were intranasally instilled with lipopolysaccharide (LPS) from P. multocida and then divided into seven groups according to euthanasia time. The nasal cavities and the lungs were processed for light microscopy, lectin histochemistry and transmission electron microscopy. Increased goblet cell activation and neutrophil infiltration were relevant changes in the nasal cavity. A predominantly interstitial pattern of diffuse alveolar damage and bronchopneumonic foci were the main lesions found in the l

Therapeutic efficacy of doxycycline against experimental Pasteurella multocida infection in broiler chickens.

The efficacy of doxycycline was investigated in two sets of experiments. In the first experiment 40, in the second experiment 60, hence altogether 100 five-week-old Ross broilers of both sexes were used. The birds were randomly allocated into groups (A and B in experiment 1; A, B and C in experiment 2) of 20 birds in each. All birds were infected intramuscularly with approx. 2 x 10(3) colony forming units of Pasteurella multocida strain X-73 (serotype A:1). Birds in groups A were non-medicated controls. Chickens in groups B were given doxycycline via the drinking water at a dose of 10 mg/kg bo

Preliminary studies with a live streptomycin-dependent Pasteurella multocida and Pasteurella haemolytica vaccine for the prevention of bovine pneumonic pasteurellosis.

Twelve Pasteurella-free Holstein-Friesian calves were used in a study to test the efficacy of a live streptomycin-dependent Pasteurella multocida A:3 and streptomycin-dependent Pasteurella haemolytica A1 vaccine. The calves were inoculated intramuscularly twice at 14-day intervals with either the streptomycin-dependent vaccine, containing 1 X 10(6) colony forming units/mL P. multocida and 4 X 10(8) colony forming units/mL P. haemolytica, commercial bacterin, or phosphate buffered saline. Pathological and molecular evidence for the role of Pasteurella multocida in respiratory disease of lambs.

Pasteurella multocida is an opportunistic pathogen associated with respiratory disease in sheep, although its relevance has not yet been fully defined. The objective of the present study was to investigate the association between P. multocida and pulmonary lesions through an integrated pathological and molecular approach. A comparative cross-sectional study was conducted in southern Spain using 201 lung samples collected from apparently healthy fattening lambs (50-60 days old) during winter and summer. This study aimed to molecularly characterize pulmonary lesions in fattening lambs and assess

Severe epiglottitis caused by Pasteurella multocida: A case report and the literature review.

Pasteurella multocida is a small, facultatively anaerobic coccobacillus commonly found in the upper respiratory tract of many mammals, birds, and especially felines. It can cause a wide range of infections in humans, often resulting from bites or scratches from pet animals. The literature has previously reported 15 cases of epiglottitis caused by Pasteurella multocida. Here we describe a case of severe epiglottitis complicated with neck area cellulitis in a previously healthy middle-aged woman. Blood cultures were positive for Pasteurella multocida, likely originating from a scratch by the pat
