Semiquantitative cytochemical estimation of glucose-6-phosphate dehydrogenase activity in benign diseases and carcinoma of the breast.

The level of glucose-6-phosphate dehydrogenase (G6PDH) activity was semiquantitatively evaluated in fresh imprints of infiltrative ductal carcinoma, fibrocystic disease and fibroadenoma of the breast. A significantly higher level of G6PDH activity was found in the carcinomas. The results suggest that the estimation of G6PDH activity could be a valuable method for evaluating the cells in benign and malignant breast lesions. It is possible that the intensification of G6PDH activity in carcinomas is a sign of the shift of the carbohydrate metabolism from an aerobic path or that the activity of th

Mellituria screening in some metabolic diseases.

Mellituria was studied in 83 subjects: 25 normal adults and children and 58 patients with several metabolic diseases. In comparison to the controls, no significant differences were found in 9 patients with cystinuria and in 2 patients with Apert's syndrome. The large excretion of glucose was the only important pattern of the 11 patients with insulin-dependent diabetes. In 23 patients with porphyria cutanea tarda a statistically significant increase in the excretion of pentose was observed. In 16 children with the classical form of phenylketonuria, a significant hypoexcretion of glucose was fou

Electrophoresis of glucose-6-phosphate and 6-phosphogluconate dehydrogenases in erythrocytes from malaria-infected animals.

Electrophoresis of red-cell extracts from control and malaria-infected animals on 'Cellogel' demonstrated the absence of G-6-PD activity of 'parasite' origin, although 6-PGD activity was present. An identical 6-PGD isoenzyme was found in mice, rats and hamsters infected with the same strain of Plasmodium berghei indicating the parasite as the source of the enzyme. A similar 6-PGD isoenzyme was also found in a few preliminary experiments with P. knowlesi-infected monkey erythrocytes. The implications of these findings are discussed in relation to previous studies and also to the status of the p

Catalytic Properties of NADP-Reducing Enzymes from Streptococcus cristatus ATCC 51100.

Streptococcus cristatus (S. cristatus) belongs to the viridans group of streptococci and is a commensal of the human upper respiratory tract. With the non-phosphorylating glyceraldehyde-3-phosphate dehydrogenase, GapN, and the oxidative part of the pentose phosphate pathway (oxPPP), S. cristatus can use two different metabolic pathways to provide reduced nicotinamide adenine dinucleotide phosphate (NADPH), an essential cofactor of anabolic reactions such as fatty acid and amino acid biosynthesis. Regarding their NADP-reducing capacity, streptococci can be categorized into three groups: those t

Reduced cyclin D3 expression in erythroid cells protects against malaria.

The severity of malaria varies substantially between individuals, but the mechanisms that underlie these differences remain unclear. Because erythrocytes have a key role in malaria biology, genetic variants associated with the development of these cells could inform the mechanisms that determine disease severity. Here we investigate the mechanistic basis of the association of the variant rs112233623-T with erythrocyte properties, and examine its role in modulating malaria severity. This variant is associated with increased levels of haemoglobin A2, increased erythrocyte size and reduced erythr

Metatranscriptomic analysis reveals gut microbiome bacterial genes in pyruvate and amino acid metabolism associated with hyperuricemia and gout in humans.

Several pathologies with metabolic origin, such as hyperuricemia and gout, have been associated with the gut microbiota taxonomic profile. However, there is no evidence of which bacterial genes are being expressed in the gut microbiome, and of their potential effects on hyperuricemia and gout. We sequenced the RNA of 26 fecal samples from 10 healthy normouricemic controls, 10 with asymptomatic hyperuricemia (AH), and six gout patients. The coding sequences were mapped to KEGG orthologues (KO). We compared the expression levels using generalized linear models and validated the expression of fou

Exploring the impact of age, and body condition score on erythrocytic B1-Dependent transketolase activity in cats: A comprehensive analysis of thiamine status.

One of the key factors influencing aging and morbidity is the overall antioxidant status and regenerative capacity. In examining contributors to the antioxidant status, we analyzed the thiamine status in felines and the influence of age, gender, and body condition score. We measured erythrocytic B1-dependent specific transketolase (STKT) activity, an enzyme in the pentose phosphate pathway, in a group of 60 sexually intact, healthy, and specific pathogen-free felines (44 females, 16 males, aged 1-17 years) with thiamine diphosphate (TDP; 0.3 and 3 mM) and without it. Only two parameters (STKT

Fibrosis Development Linked to Alterations in Glucose and Energy Metabolism and Prooxidant-Antioxidant Balance in Experimental Models of Liver Injury.

The development of liver fibrosis is one of the most severe and life-threatening outcomes of chronic liver disease (CLD). For targeted therapy of CLD, it is highly needed to reveal molecular targets for normalizing metabolic processes impaired in damaged liver and associated with fibrosis. In this study, we investigated the morphological and biochemical changes in rat liver models of fibrosis induced by chronic administration of thioacetamide, carbon tetrachloride, bile duct ligation (BDL), and ischemia/reperfusion (I/R), with a specific focus on carbohydrate and energy metabolism. Changes in

Integrative Metabolomics, Proteomics and Transcriptomics Analysis Reveals Liver Toxicity of Mesoporous Silica Nanoparticles.

As pharmaceutical excipients, mesoporous silica nanoparticles (MSNs) have attracted considerable concern based on potential risks to the public. The impact of MSNs on biochemical metabolism is poorly understood, and few studies have compared the effects of MSNs administered via different routes. To evaluate the hepatotoxicity of MSNs, metabolomics, proteomics and transcriptomic analyses were performed in mice after intravenous (20 mg/kg/d) or oral ad-ministration (200 mg/kg/d) of MSNs for 10 days. Intravenous injection induced significant hepatic injury based on pathological inspection and inc

Qualitative and quantitative profiles of jejunal oligosaccharides and cecal short-chain fatty acids in broiler chickens receiving different dietary levels of fiber, protein and exogenous enzymes.

BACKGROUND: Dietary supplemental carbohydrases are able to degrade non-starch polysaccharides and generate oligosaccharides in the gastrointestinal tract. This study was conducted to investigate the influence of dietary fiber and protein levels on growth performance, nutrient utilization, digesta oligosaccharides profile and cecal short-chain fatty acid (SCFA) profile in broilers receiving diets supplemented with xylanase or protease individually or in combination. RESULTS: Enzyme supplementation had no effect on growth performance. There was significant (P < 0.05) fiber × protein × xylanase

Glycolytic activity of the tissue stem cells in the macula flava of the human vocal fold.

OBJECTIVES: In our previous studies, the features of the mitochondria of tissue stem cells in the maculae flavae of the human vocal fold suggested that their metabolic activity and oxidative phosphorylation was low. This study investigated the metabolic activity, especially glycolysis of the tissue stem cells in the maculae flavae of the human adult vocal fold. STUDY DESIGN: Histologic analysis of the human vocal folds. METHODS: Three normal human adult vocal folds obtained from autopsy cases were investigated using immunohistochemistry. RESULTS: Among the three phenotypes of cells in the h

Myeloid Slc2a1-Deficient Murine Model Revealed Macrophage Activation and Metabolic Phenotype Are Fueled by GLUT1.

Macrophages (MΦs) are heterogeneous and metabolically flexible, with metabolism strongly affecting immune activation. A classic response to proinflammatory activation is increased flux through glycolysis with a downregulation of oxidative metabolism, whereas alternative activation is primarily oxidative, which begs the question of whether targeting glucose metabolism is a viable approach to control MΦ activation. We created a murine model of myeloid-specific glucose transporter GLUT1 (Slc2a1) deletion. Bone marrow-derived MΦs (BMDM) from Slc2a1M-/- mice failed to uptake glucose and demonstrate

Stimulation by iodide of H(2)O(2) generation in thyroid slices from several species.

The regulation of thyroid metabolism by iodide involves numerous inhibitory effects. However, in unstimulated dog thyroid slices, a small inconstant stimulatory effect of iodide on H(2)O(2) generation is observed. The only other stimulatory effect reported with iodide is on [1-(14)C]glucose oxidation, i.e., on the pentose phosphate pathway. Because we have recently demonstrated that the pentose phosphate pathway is controlled by H(2)O(2) generation, we study here the effect of iodide on basal H(2)O(2) generation in thyroid slices from several species. Our data show that in sheep, pig, bovine,

Effect of acetic acid on xylose conversion to ethanol by genetically engineered E. coli.

Efficient utilization of the pentosan fraction of hemicellulose from lignocellulosic feedstocks offers an opportunity to increase the yield and to reduce the cost of producing fuel ethanol. During prehydrolysis (acid hydrolysis or autohydrolysis of hemicellulose), acetic acid is formed as a consequence of the deacetylation of the acetylated moiety of hemicellulose. Recombinant Escherichia coli B (ATCC 11303), carrying the plasmid pLO1297 with pyruvate decarboxylase and alcohol dehydrogenase II genes from Zymomonas mobilis (CP4), converts xylose to ethanol with a product yield that approaches t

Membrane characteristics and metabolic properties of glucose-6-phosphate dehydrogenase deficient red cells.

Two Finnish variants of reduced erythrocyte glucose-6-phosphate dehydrogenase (G-6-PD) activity were studied. The G-6-PD Espoo variant is characterized by severe enzyme deficiency which is normally non-haemolytic although primaquine sensitive. The other variant, G-6-PD Helsinki, in which the enzyme activity is moderately reduced, is associated with chronic haemolytic anaemia. The activity of the pentose phosphate pathway was not stimulated by methylene blue in G-6-PD Espoo cells, whereas in normal and G-6-PD Helsinki cells there were increases in shunt activity of 64.5- and 5.3-fold, respectiv

Protocol for metabolic profiling of antigen-specific CD8+ T cells using spectral flow cytometry.

CD8+ T cells rely on tightly regulated metabolic remodeling to support effector function. Here, we present a protocol for single-cell metabolic profiling of rare antigen-specific CD8+ T cells in unstimulated and antigen-stimulated conditions using spectral flow cytometry. The workflow enables detailed assessment of key metabolic pathways, including glycolysis, fatty acid oxidation, amino acid metabolism, the pentose phosphate pathway, and mitochondrial oxidative phosphorylation. For complete details on the use and execution of this protocol, please refer to Mülling et al.1.

Enhanced stability of erythrocyte-entrapped glucocerebrosidase activity.

The stability of free glucocerebrosidase activity has been compared with the stability of glucocerebrosidase entrapped in hemolyzed, resealed erythrocytes. Encapsulation markedly stabilizes the enzyme, partly by providing an environment with a high protein concentration. In addition, in the presence of glucose, enzyme activity and GSH content remain constant, but in its absence, enzyme activity and GSH levels fall, indicating the existance of an additional, metabolically linked protective mechanism. Since the free enzyme is inactivated by p-CMB and is protected by DTT, it is likely that GSH ge

[Thiamine deficit in acute alcoholic psychoses].

In 35 females and 22 males with acute alcoholic disorders and in 44 individuals of a control group the author studied the indices of thiamine balance--the content of thiamine and pyruvic acid in the blood and urine, concentration of citric acid in the blood, activity of transketolase of erythrocytes and TDP-effect. The results demonstrated that a thiamide deficiency in excretion and insufficiency in coenzyme functions of oxidative decarboxylation of pyruvic acid and in the pentose cycle is seen not only in alcoholic psychoses, but in the withdrawal syndrome. Sex differences of these indices an

Identification of shared gene signature between lung cancer and postoperative delirium by transcriptome data analysis.

Postoperative delirium (POD) is clinically associated with a variety of adverse outcomes. The incidence of POD in patients undergoing radical resection of lung cancer is about 5% to 16%, higher than that in patients undergoing other surgeries. However, the molecular mechanisms that specifically lead to POD after radical resection of lung cancer have not been fully elucidated. Gene expression profile datasets of lung cancer (GSE31210) and POD (GSE163943) were downloaded from the Gene Expression Omnibus database. The weighted gene co-expression network analysis (WGCNA) was used to screen key mod

Xy-31 and Lat-1 regulate the induction of polysaccharide disintegrating genes by arabinose in Neurospora crassa OR74A.

BACKGROUND: Arabinose has been identified as a potent inducer of the cellulase regulon in filamentous fungi. However, the precise molecular mechanisms that drive this induction remain largely unknown. This study aimed to elucidate the mechanisms underlying the arabinose-induced activation of cellulase and hemicellulase enzyme clusters in the model filamentous fungus Neurospora crassa OR74A. RESULTS: Arabinose stimulates the secretion of cellulases and hemicellulases in N. crassa. While genome-wide scrutiny predicted several sugar transporters in N. crassa, expression studies under avicel and
