OTUD6A drives dopaminergic neuronal degeneration of Parkinson's disease through deubiquitinating ACTG1 in neuronal cells.

Parkinson's disease (PD) is a severe neurodegenerative disorder characterized by the progressive loss of dopaminergic neurons. Emerging evid … UD6A specifically interacts with the 8-181 aa domain of ACTG1 and preferentially cleaves K48-linked polyubiquitin chains, thereby enhancing ACTG1 protein stability in neuronal cells. The stabilized ACTG1 subsequently binds to p53 and facilitates its nuclear translocation, leading to the transcriptional activation of pro-apoptotic genes and promoting neuronal ap …

Ubiquitin-activating enzyme1 (TgUAE1) acts as a key regulator of Toxoplasma gondii lytic cycle and homeostasis.

Ubiquitylation, regulated by the ubiquitin-proteasome system (UPS), is crucial for cell division and cycle transitions in Toxoplasma gondii. However, the primary E1 ubiquitin-activating enzyme (UAE1) in this process has been unclear. This study identified and characterized TGGT1_290290 (TgUAE1) as the canonical E1 enzyme in T. gondii. Through a combination of bioinformatics, biochemical, pharmacological, and genetic approaches, TgUAE1 was shown to exhibit typical E1 activity, particularly in forming K48- and K63-linked polyubiquitin chains. TAK-243, a UAE1 inhibitor, can effectively inhibit th…

Moyamoya disease patient mutations in the RING domain of RNF213 reduce its ubiquitin ligase activity and enhance NFκB activation and apoptosis in an AAA+ domain-dependent manner.

Moyamoya disease (MMD) is a cerebrovascular disease characterized by progressive occlusion of the internal carotid arteries. Genetic studies … ain, identified Ubc13/Uev1A as a key ubiquitin conjugating enzyme that together generate K63-linked polyubiquitin chains. However, nearly all MMD patient mutations in the RING domain greatly reduced this activity. When full-length proteins were overexpressed in HEK293T cells, patient mutations that abolished the ubiquitin ligase activities conversely enhanced n …

Nucleocytoplasmic O-glycosylation in protists.

O-Glycosylation is an increasingly recognized modification of intracellular proteins in all kingdoms of life, and its occurrence in protists has been investigated to understand its evolution and its roles in the virulence of unicellular pathogens. We focus here on two kinds of glycoregulation found in unicellular eukaryotes: one is a simple O-fucose modification of dozens if not hundreds of Ser/Thr-rich proteins, and the other a complex pentasaccharide devoted to a single protein associated with oxygen sensing and the assembly of polyubiquitin chains. These modifications are not required for l…

CYLD mediates ciliogenesis in multiple organs by deubiquitinating Cep70 and inactivating HDAC6.

Cilia are hair-like organelles extending from the cell surface with important sensory and motility functions. Ciliary defects can result in … dies and axenomes. The ciliary function of CYLD is partially attributed to its deconjugation of the polyubiquitin chain from centrosomal protein of 70 kDa (Cep70), a requirement for Cep70 to interact with γ-tubulin and localize at the centrosome. In addition, CYLD-mediated inhibition of histone deacetylase 6 (HDAC6), which promotes tubulin acetylation, constitu …

Multiple independent regulatory pathways control UBI4 expression after heat shock in Saccharomyces cerevisiae.

Transcription of the polyubiquitin gene UBI4 of Saccharomyces cerevisiae is strongly induced by a variety of environmental stresses, such as heat shock, nutrient depletion and exposure to DNA-damaging agents. This transcriptional response of UBI4 is likely to be the primary mechanism for increasing the pool of ubiquitin for degradation of stress-damaged proteins. Deletion and promoter fusion studies of the 5' regulatory sequences indicated that two different elements, heat shock elements (HSEs) and stress response element (STREs), contributed independently to heat shock regulation of the UBI4 …

MAVS undergoes aggregation under hypoxia to enhance the hypoxia signaling pathway by promoting the generation of mitochondrial ROS.

The mitochondrial antiviral signaling protein (MAVS) is a key component of the innate immune response against RNA viruses. Its involvement in other cellular stresses is largely unknown. Here, we found that hypoxia induces MAVS aggregation, leading to the stabilization of HIF1/2α proteins and the enhancement of hypoxia signaling. Mechanistic studies revealed that MAVS enhances TRAF6 binding to ECSIT, which catalyzes ECSIT polyubiquitination. This promotes mitochondrial ROS generation and likely inhibits PHD2 enzymatic activity, thereby reducing HIF1/2α proteasomal degradation. Disrupting MAVS i…

WWP1 mediates the ubiquitination and degradation of HIPK3 in bladder cancer cells.

Protein homeostasis is primarily regulated by post-translational modifications (PTMs). HIPK3 has been recognized as a tumor suppressor across various cancers. However, the impact of PTMs on HIPK3 remains insufficiently explored. This study identified WWP1 as an E3 ubiquitin ligase targeting HIPK3, demonstrating that WWP1 downregulates HIPK3 protein levels by facilitating its ubiquitination. Mechanistically, WWP1 directly interacts with HIPK3, promoting K48-linked polyubiquitination at the K1187 site. The WWP1/HIPK3 axis modulates cancer cell chemosensitivity through the regulation of the JNK s…

Role of the proteasome in Alzheimer's disease.

The proteasome is a multicatalytic complex involved in the degradation of polyubiquitinated proteins. Here we review the clues of a possible involvement of the proteasome in Alzheimer's disease neuropathology. Thus, we discuss the fact that the proteasome modulates the intracellular concentrations of presenilins 1 and 2. These two proteins, when mutated, appear responsible for most of early onset forms of Alzheimer's disease and this is thought to be due to the exacerbation of the pathogenic pathway of the maturation of the beta-amyloid precursor protein. Controlling presenilins concentrations…

Artemisinin Bioactivity and Resistance in Malaria Parasites.

Artemisinin is the most widely-used compound against malaria and plays a critical role in the treatment of malaria worldwide. Resistance to artemisinin emerged about a decade ago in Southeast Asia and it is paramount to prevent its spread or emergence in Africa. Artemisinin has a complex mode of action and can cause widespread injury to many components of the parasite. In this review, we outline the different metabolic pathways affected by artemisinin, including the unfolded protein response, protein polyubiquitination, proteasome, phosphatidylinositol-3-kinase, and the eukaryotic translation …

A BIN2-SOG1 molecular module regulates replication stress response independently on ATR.

DNA replication stress poses a severe threat to genome stability. It is well-known that SOG1 plays a crucial role in DNA replication stress response in plants. Although ATM-mediated phosphorylation and CRL4PRL1-mediated polyubiquitination of SOG1 play important roles in regulating DNA damage response (DDR), the posttranslational modifications of SOG1 during the replication stress response are little known. Here we show that, during replication stress, the activity of the GSK3-like kinase BIN2 is activated by an unknown mechanism independent of ATR. Subsequently, BIN2 phosphorylates SOG1 at T24…

F-box protein FBXL19-mediated ubiquitination and degradation of the receptor for IL-33 limits pulmonary inflammation.

The ST2L receptor for interleukin 33 (IL-33) mediates pulmonary inflammation and immune system-related disorders, such as asthma and rheumatoid arthritis. At present, very little is known about the molecular regulation of ST2L expression. Here we found that FBXL19, an 'orphan' member of the Skp1-Cullin-F-box family of E3 ubiquitin ligases, selectively bound to ST2L to mediate its polyubiquitination and elimination in the proteasome. Degradation of ST2L involved phosphorylation of ST2L at Ser442 catalyzed by the kinase GSK3β. Overexpression of FBXL19 abrogated the proapoptotic and inflammatory …

PSMD14 promotes hepatocellular carcinoma cell proliferation through E2F4-dependent PHF5A transcription and alternative splicing.

Dysregulation of the ubiquitin-proteasome system contributes to hepatocellular carcinoma (HCC), but how deubiquitination is linked to transcriptional regulation and alternative splicing remains unclear. In this study, proteomic analysis identified E2F4 as a PSMD14-interacting protein. Co-immunoprecipitation and ubiquitination assays showed that PSMD14 removed K48-linked polyubiquitin chains from E2F4, thereby reducing its degradation and increasing its protein stability. Stabilized E2F4 directly bound to the PHF5A promoter and promoted PHF5A transcription. As a component of the spliceosome, PH…
