Stimulation of human prolactin secretion by mescaline.

Prolactin (PRL) and Growth Hormone (GH) secretions were studied in human serum after the oral administration of 5 mg/kg mescaline (3,4,5-trimethoxy-beta-phenylethylamine) or 2,3,4-trimethoxy-beta-phenylethylamine (2,3,4-TMPEA) respectively. Mescaline stimulated the secretion of PRL more than four-fold above base-line levels. Peak concentrations were found 90--120 min after drug intake. Five hours later serum PRL was still markedly increased. Mescaline also triggered GH secretion. There was no alteration of serum PRL and GH concentrations after intake of the non-hallucinogenic 2,3,4-TMPEA.

[The pathological secretion of prolactin (author's transl)].

The problem of hyperprolactinaemia is dominated by the search for an active pituitary adenoma, which, if it is at all large, requires neuro-surgical operation. In cases where a microadenoma is responsible or where the pathological secretion of prolactin is istrogenic, the most serious consequence is sterility. Hyperprolactinaemia in fact works at two levels: on cyclical gonadotrophic activity and on luteal function. Whatever the cause is, bromocriptine allows hyperprolactinaemia to be corrected together with the disturbances of ovulation resulting from it.

Evaluation of research on control of prolactin secretion.

At least three substances have been reported to be present in the hypothalamus that can inhibit prolactin release, namely a PIF, catecholamines and acetylcholine. At least four substances have been reported to be present in the hypothalamus that can stimulate prolactin release, namely PRF, TRH, serotonin and prostaglandins. Neither the existence of a distinctive PIF or PRF in the hypothalamus can be considered as definitely established. The predominant action of the mammalian hypothalamus on prolactin release is inhibitory under most conditions, and is stimulatory in avian species. In addition…

Prolactin concentrations in patients with breast cancer.

Basal prolactin levels were measured before operation in 113 patients undergoing surgery for a lump in the breast. Prolactin concentrations were not significantly higher in those patients with breast cancer than in those with benign lumps. Prolactin concentrations were, however, found to be higher in premenopausal women than in postmenopausal ones, regardless of tumour histology.

From the perspective of prolactin: a view on obesity.

The physiological roles of prolactin extend beyond its classical functions in reproductive regulation. Emerging evidence indicates that prolactin is involved in energy homeostasis and may interact pathophysiologically with obesity; this has attracted increasing attention in endocrinology and metabolic research. Hyperprolactinemia (HPRL) is frequently observed in obese individuals. Observational studies have reported that weight-loss interventions are associated with reduced circulating prolactin levels, whereas dopamine agonists, which suppress prolactin secretion, improve metabolic and endocr…

BREAST CANCER AND PROLACTIN: A LIFETIME RELATIONSHIP.

Prolactin is a multifunctional hormone produced by the pituitary gland, essential for more than just the growth of mammary tissue. It plays a crucial role in lobuloalveolar development and lactation. Understanding multifunctional hormone, prolactin, and its role in breast tissue is a promising avenue in our fight against breast cancer. This paper aims to shed light on the physiological mechanisms of prolactin in breast cancer, its prognostic value, and potential therapeutic approaches, offering a significant step forward in our battle against this disease.

Prolactin effects on the pathogenesis of diabetes mellitus.

BACKGROUND: Prolactin (PRL) is a pituitary hormone promoting lactation in response to the suckling reflex. Beyond its well-known effects, novel tissue-specific and metabolic functions of PRL are emerging. AIMS: To dissect PRL as a critical mediator of whole-body gluco-insulinemic sensitivity. METHODS: PubMed-based search with the following terms 'prolactin', 'glucose metabolism', 'type 2 diabetes mellitus', 'type 1 diabetes mellitus', 'gestational diabetes mellitus' was performed. DISCUSSION: The identification of the PRL-glucose metabolism network poses the basis for unprecedented avenues …

The Involvement of Prolactin in Stress-Related Disorders.

The most important and widely studied role of prolactin (PRL) is its modulation of stress responses during pregnancy and lactation. PRL acts as a neuropeptide to support physiological reproductive responses. The effects of PRL on the nervous system contribute to a wide range of changes in the female brain during pregnancy and the inhibition of the hypothalamic-pituitary axis. All these changes contribute to the behavioral and physiological adaptations of a young mother to enable reproductive success. PRL-driven brain adaptations are also crucial for regulating maternal emotionality and well-be…

The brain as a source and a target of prolactin in mammals.

Prolactin is a polypeptide hormone associated with an extensive variety of biological functions. Among the roles of prolactin in vertebrates, some were preserved throughout evolution. This is the case of its function in the brain, where prolactin receptors, are expressed in different structures of the central nervous system. In the brain, prolactin actions are principally associated with reproduction and parental behavior, and involves the modulation of adult neurogenesis, neuroprotection, and neuroplasticity, especially during pregnancy, thereby preparing the brain to parenthood. Prolactin is…

Variant Prolactin Receptor in Agalactia and Hyperprolactinemia.

A loss-of-function variant in the gene encoding the prolactin receptor ( PRLR) was reported previously in a woman with persistent postpartum galactorrhea; however, this paradoxical phenotype is not completely understood. Here we describe a 35-year-old woman who presented with idiopathic hyperprolactinemia that was associated with a complete lack of lactation after each of her two deliveries. She is a compound heterozygote for loss-of-function variants of PRLR. Her unaffected parents are heterozygotes. These findings are consistent with previous work showing that mice deficient in functional Pr…

Prolactin function and putative expression in the brain.

INTRODUCTION: Prolactin is a peptide hormone mainly synthetized and secreted by the anterior pituitary gland, but also by extrapituitary tissues, such as mammary gland, decidua, prostate, skin, and possibly the brain. Similarly, prolactin receptor is expressed in the pituitary gland, many peripheral tissues, and in contrast to prolactin, its receptor has been consistently detected in several brain regions, such as cerebral cortex, olfactory bulb, hypothalamus, hippocampus, amygdala, among others. Classically, prolactin function has been related to the stimulation of lactogenesis and galactopoi…

Sellar gangliocytoma with adrenocorticotropic and prolactin adenoma.

We report a case of a 60-year-old man who presented with weight gain, headaches, dizziness, erectile dysfunction and decreased libido. He was found to have elevated adrenocorticotropic hormone (ACTH) and prolactin serum levels. The imaging studies revealed a 1.4 cm sella/suprasellar mass which was compressing the optic chiasm. Histologic slides of the lesion showed a pituitary adenoma, marked by a proliferation of biphenotypic appearing cells, associated with a gangliocytoma, and marked by a proliferation of atypical appearing neuronal cells arranged against a glial-appearing background. Pitui…

Prolactin and aggression in women with fertility problems.

This study tested the hypothesis that women with higher prolactin feel more hostility, anger and aggression. A total of 66 women with moderate fertility problems were grouped into the 50% who had the highest and the 50% who had the lowest levels of prolactin. Levels of hostility, aggression and anger were compared. Women with higher prolactin levels did not report significantly increased hostility. After Bonferroni correction, women with lower prolactin showed non-significantly increased scores on two measures of state anger, and on a measure of trait temper. When comparing those with the high…

[Prolactin as an immunomodulatory factor in psoriatic arthritis].

Prolactin (PRL) is a hormone synthesized and secreted by lactotroph cells in the anterior pituitary gland. There is also extrapituitary hormone secretion by many cells, including cells of the immune system. In physiological conditions PRL is responsible for lactogenesis and other processes associated with it. PRL plays a significant role during the immune response as a cytokine, affecting proliferation and differentiation of many immune system cells. The biological effect of the hormone depends on binding with the specific prolactin receptor PRL-R, and activation of the transcription factors o…

Is prolactin involved in the evolution of atherothrombotic disease?

Cardiovascular diseases (CVDs) account for approximately 30% of all deaths globally. The most important cause of CVD is atherothrombosis, in other words, narrowing of the arteries as a result of the deposition of cholesterol and other lipoid substances within the arterial wall. Several endocrine disorders have been linked to this pathological state. Recent clinical and experimental studies have suggested that prolactin, a pleiotropic pituitary hormone, may potentially contribute to CVD, either through direct modulation of local cellular processes within atherosclerotic plaques/thrombi and/or t…
