Expression and regulation of the 67-kda laminin-binding protein and its precursor gene in lymphoid-cells.

The 67-kDa laminin-binding protein is a non-integrin laminin-binding protein that mediates cancer cell adhesion and migration. The expression of the 67-kDa laminin-binding protein and of its putative precursor, a 37-kDa polypeptide, was studied in peripheral T-cells and T-lymphoma cell lines. Immunofluorescence experiments detected antigen in both the cytosol and on the cell membrane. On immunoblots of T-cell protein extracts, both the 37-kDa precursor and the mature 67-kDa protein were present. The mRNA for the precursor was expressed in both immature and mature thymocytes. In three independe

Regulation and expression of the Alzheimer's beta/A4 amyloid protein precursor in health, disease, and Down's syndrome.

A four- to fivefold overexpression of the gene for the Alzheimer beta/A4 amyloid precursor protein (APP) in individuals with Down's Syndrome (DS) appears to be responsible for the fifty year earlier onset of Alzheimer's disease (AD) pathology in DS compared to the normal population. It is therefore likely that a deregulated overexpression of the APP gene is a risk factor for the beta/A4 amyloid formation. To test this hypothesis and to get a better understanding of how APP expression is regulated, we studied the 5' control region of the human APP gene, alternative splicing of the 19 APP exons,

Evidence for excitoprotective and intraneuronal calcium-regulating roles for secreted forms of the beta-amyloid precursor protein.

The beta-amyloid precursor protein (beta APP) is a membrane-spanning glycoprotein that is the source of the beta-amyloid peptide (beta AP) which accumulates as senile plaques in the brains of patients with Alzheimer's disease. beta APP is normally processed such that a cleavage occurs within the beta AP, liberating secreted forms of beta APP (APPss) from the cell. The neuronal functions of these forms are unknown. We now report that APPss have a potent neuroprotective action in cultured rat hippocampal and septal neurons and in human cortical neurons. APPs695 and APPs751 protected neurons agai

Acute effect of protein or carbohydrate breakfasts on human cerebrospinal fluid monoamine precursor and metabolite levels.

Patients with normal pressure hydrocephalus who had three lumbar punctures during 1 week ingested either water, a protein breakfast, or a carbohydrate breakfast 2.5 h before each of the lumbar punctures. The CSF was analyzed for biogenic amine precursors and metabolites. The protein meal raised CSF tyrosine levels, a finding consistent with animal data, but did not alter those of tryptophan or any of the biogenic amine metabolites. The carbohydrate meal increased CSF 3-methoxy-4-hydroxyphenylethylene glycol, an unexplained finding. The carbohydrate meal did not affect CSF tryptophan, tyrosine,

High level of accumulation of a mRNA coding for a precursor-like protein in the submaxillary gland of male rats.

NaDodSO4/PAGE analysis of in vitro translation products of rat submaxillary gland (SMG) mRNAs has revealed an important sexual dimorphism. Moreover, most of the rat male-specific major translation products differ in size from those translated from male mouse SMG mRNAs. To characterize proteins accumulated in the rat SMG under androgen control, a cDNA library was constructed. Here we report the nucleotide sequence of a 0.7-kilobase mRNA that is 1000-3000 times more abundant in male rats than in female rats. The predicted corresponding protein, SMR1, has a molecular weight of 16,000 and contains

[Effect of the quality and quantity of dietary proteins on vitamin A depletion rate, and biological availability of vitamin A precursors].

The effect of quantity and quality of protein from cereal and legume sources on the rate of vitamin A depletion was evaluated through changes in retinol serum levels and liver stores in male and female Wistar strain rats. A total of 64 animals were distributed into four groups and fed ad libitum with diets adequate in all nutrients, except vitamin A. The protein in the diets was derived from 91% common maize (Diet A); 91% Opaque-2 maize (Diet B), 64% common maize plus 27% precooked common black bean flour (Diet C), and 64% Opaque-2 maize with 27% bean flour (Diet D). The total depletion period

The microcephaly protein Abnormal Spindle has an essential role in symmetrically dividing neural precursors to promote brain growth and development.

Neurogenesis must be coordinated in time and space to ensure proper neural development. Defects in this process can lead to a variety of brain malformations that affect tissue size and architecture. Here we examine the spatiotemporal requirements of the Abnormal Spindle (Asp) protein and its human ortholog Abnormal Spindle-Like, Microcephaly Associated (ASPM) in a fly model of human microcephaly (MCPH). By utilizing the fly optic lobe, whose neurogenic program parallels that of mammals, and the genetic tools available in this organism, we uncover the progenitor cell types and critical neurogen

Global O-GlcNAcome Identifies O-GlcNAcylated Proteins Regulating Oligodendrocyte Precursor Cells Differentiation.

Oligodendrocyte precursor cells (OPCs) differentiate into myelinating oligodendrocytes to form the myelin sheaths essential for neural function, a process frequently compromised during aging and in diseases like multiple sclerosis. However, the regulatory landscape of myelination remains incompletely understood. Here, we perform a comprehensive proteomic analysis of the O-GlcNAcome in postnatal day 0 mouse OPCs, identifying 165 O-GlcNAcylation sites across 118 proteins. These proteins are mostly nuclear and characterized by single-site modifications. Comparative analysis identifies 74 novel si

Impaired Cognitive Flexibility With Preserved Learning in an Amyloid Precursor Protein Knock-In Mouse Model of Amyloidopathy.

Alzheimer's disease is a debilitating neurodegenerative condition characterized by amyloid beta plaques and tau neurofibrillary tangles, which leads to progressive cognitive decline. Several new mouse models of fast amyloid deposition have been generated with compound mutations, but how these affect high-level cognitive function is still not fully understood. Four cohorts of a second-generation amyloid precursor protein knock-in mouse model, AppNL-G-F/NL-G-F, which develops aggressive amyloidopathy, were compared with two different control groups that do not produce plaques (AppNL/NL and wildt

Identification of fibrinogen Aα-chain as an amyloid precursor protein in a case of canine coronary artery amyloidosis.

Spontaneous amyloid deposition occurs in the coronary arteries in older dogs, but the pathogenesis, including the precursor protein, is not known. We conducted proteome analysis using liquid chromatography-tandem mass spectrometry to identify the amyloid precursor protein in a 13-y-old male mixed-breed dog with amyloid deposition in intramural coronary arteries, and identified fibrinogen Aα-chain. This form of amyloidosis is characterized by the deposition of amyloid derived from fibrinogen Aα-chains, which has been reported previously only in humans and Japanese squirrels. Within amyloid depo

Precursor A-Kinase Anchor Protein 4 as a Predictive Biomarker of Post-Thaw Semen Quality in Goats.

The evaluation of sperm proteins has emerged as a promising approach to predicting semen quality across animal species. This study investigated the relationship between post-thaw concentrations of precursor A-kinase anchor protein 4 (proAKAP4) and objective sperm quality parameters in goats. Semen was collected from 16 adult goats (Boer, n = 8; Anglo-Nubian, n = 8) and frozen using a standardized protocol with OptiXcell (IMV Technologies, l'Aigle, France) extender (n = 5). After thawing, proAKAP4 concentrations were measured with an enzyme-linked immunosorbent assay (ELISA), while sperm motili

Expression and function of β-site amyloid precursor protein-cleaving enzyme 2 in vascular endothelium.

The physiological function of β-site amyloid precursor protein-cleaving enzyme 2 (BACE2) in vascular endothelium of systemic arteries is unknown. In the present study, we generated conditional tamoxifen-inducible endothelial BACE2-deficient mice (eBACE2-/- mice). Electron microscopic and Western blot analyses revealed that BACE2 protein is mainly present in endothelial cells of aorta. Genetic deletion of BACE2 in endothelial cells significantly impaired endothelium-dependent relaxations to Ca2+-ionophore A23187 in eBACE2-/- aortas as compared with tamoxifen-treated control mice, irrespective o

Translational potential of synaptic alterations in Alzheimer's disease patients and amyloid precursor protein knock-in mice.

Synaptic dysfunction is an early event in Alzheimer's disease. Post-mortem studies suggest that alterations in synaptic proteins are associated with cognitive decline in Alzheimer's disease. We measured the concentration of three synaptic proteins, zinc transporter protein 3, dynamin1 and AMPA glutamate receptor 3 in cerebrospinal fluid of subjects with mild cognitive impairment (n = 18) and Alzheimer's disease (n = 18) and compared the levels to cognitively and neurologically healthy controls (n = 18) by using ELISA assay. In addition, we aimed to assess the translational potential of these s

Effects of Overexpression of Neurosecretory Protein GL-Precursor Gene on Glucose Homeostasis and Insulin Sensitivity in Mice.

A high-fat diet (HFD) quickly induces obesity with insulin resistance and hyperglycemia. We previously reported that a novel hypothalamic small protein, named neurosecretory protein GL (NPGL), stimulates feeding and fat accumulation in mice. However, the effects of NPGL on insulin sensitivity and glucose homeostasis remain unknown. Hence, we subjected NPGL-precursor gene (Npgl)-overexpressing mice to the oral glucose tolerance test (OGTT) and intraperitoneal insulin tolerance test (IPITT) under normal chow (NC) and HFD conditions. Npgl overexpression promoted body mass gain and tended to incre

Structure of a Cell Entry Defective Human Adenovirus Provides Insights into Precursor Proteins and Capsid Maturation.

Maturation of adenoviruses is distinguished by proteolytic processing of several interior minor capsid proteins and core proteins by the adenoviral protease and subsequent reorganization of adenovirus core. We report the results derived from the icosahedrally averaged cryo-EM structure of a cell entry defective form of adenovirus, designated ts1, at a resolution of 3.7 Å as well as of the localized reconstructions of unique hexons and penton base. The virion structure revealed the structures and organization of precursors of minor capsid proteins, pIIIa, pVI and pVIII, which are closely associ

Pathogenesis of Alzheimer's disease: Multiple interacting causes against which amyloid precursor protein protects.

Alzheimer's disease is the commonest form of senile dementia. It is characterised by neuronal cell death and amyloid deposition. Amyloid precursor protein (APP), which is highly conserved in evolution, is expressed in neurones in response to a wide range of damaging agents. The hypothesis proposed is that APP has a protective function to counter damage but if it fails and the neurone dies then breakdown products of APP miss-fold and lead to amyloid deposition. This fits with the evidence that amyloid deposition is a consequence rather than a cause of cell death. Germ line mutations in APP impa

Amyloid precursor protein intracellular domain-dependent regulation of FOXO3a inhibits adult hippocampal neurogenesis.

The amyloid precursor protein (APP) intracellular domain (AICD) is a metabolic by-product of APP produced through sequential proteolytic cleavage by α-, β-, and γ-secretases. The interaction between AICD and Fe65 has been reported to impair adult neurogenesis in vivo. However, the exact role of AICD in mediating neural stem cell fate remains unclear. To identify the role of AICD in neuronal proliferation and differentiation, as well as to clarify the molecular mechanisms underlying the role of AICD in neurogenesis, we first generated a mouse model expressing the Rosa26-based AICD transgene. Amyloid precursor protein is a restriction factor that protects against Zika virus infection in mammalian brains.

Zika virus (ZIKV) is a neurotropic flavivirus that causes several diseases including birth defects such as microcephaly. Intrinsic immunity is known to be a frontline defense against viruses through host anti-viral restriction factors. Limited knowledge is available on intrinsic immunity against ZIKV in brains. Amyloid precursor protein (APP) is predominantly expressed in brains and implicated in the pathogenesis of Alzheimer's diseases. We have found that ZIKV interacts with APP, and viral infection increases APP expression via enhancing protein stability. Moreover, we identified the viral pe

Amyloid Precursor Protein Overexpression in Down Syndrome Trophoblast Reduces Cell Invasiveness and Interferes with Syncytialization.

The placentas of Down syndrome (DS) pregnancies exhibit morphologic and functional abnormalities. Although the increase in dosage of certain genes on chromosome 21 has been associated with the DS phenotype, the effects on placenta have seldom been studied. Herein, we examine the expression of four dosage-sensitive genes (APP, ETS2, SOD1, and HMGN1) in normal and DS placentas. We demonstrated significant overexpression of amyloid precursor protein (APP) in DS placentas at RNA and protein levels by real-time quantitative PCR, Western blot analysis, and immunohistochemistry.
