Distribution of ciprofloxacin in the dog prostate and various tissues.

The distribution in the dog prostate and other tissues of ciprofloxacin, a quinoline carboxylic acid derivative, was investigated in an experimental model. The concentrations in prostatic tissue, prostatic interstitial fluid (PIF), and prostatic secretion (PS) were lower than the corresponding plasma (P) concentrations, as would be expected for an acidic compound. The experiments were carried out under steady state conditions during intravenous infusion in one group of dogs and following gastric administration in another group. During steady state the ciprofloxacin concentrations were signific

Astaxanthin alleviates Parkinson's disease via dual regulation of tryptophan metabolism: central STAT3/IDO1 inhibition and gut microbiota remodeling.

Parkinson's disease (PD) is a progressive neurodegenerative disorder with limited disease-modifying treatments. Dysregulation of tryptophan … and downregulated IDO1 expression in the brain, reducing the conversion of tryptophan to neurotoxic quinolinic acid (QUIN). Concurrently, AX reshaped the gut microbiota by enriching Lactobacillus, which redirected tryptophan metabolism toward the production of neuroprotective indole derivatives, including indole-3-propionic acid (IPA) and indole-3-acetic acid (IA …

Improved synthesis and anticancer activity of a potent neuronal nitric oxide synthase inhibitor.

An improved synthesis of 4-methyl-7-(3-((methylamino)methyl)phenethyl)quinolin-2-amine (1) is reported. A scalable, rapid, and efficient methodology was developed to access this compound with an overall yield of 35%, which is 5.9-fold higher than the previous report. The key differences in the improved synthesis are a high yielding quinoline synthesis by a Knorr reaction, a copper-mediated Sonogashira coupling to the internal alkyne in excellent yield, and a crucial deprotection of the N-acetyl and N-Boc groups achieved under acidic conditions in a single step rather than a poor yielding quino

Effective mineralization of quinoline and bio-treated coking wastewater by catalytic ozonation using CuFe2O4/Sepiolite catalyst: Efficiency and mechanism.

In this study, CuFe2O4 nanocomposite loaded on natural sepiolite (CuFe2O4/SEP) was prepared by the citrate sol-gel method. CuFe2O4/SEP was characterized by X-ray diffraction, Brunauer-Emmett-Teller adsorption analysis, scanning electron microscopy, and energy dispersive spectroscopy. The CuFe2O4/SEP composite was stable and showed an excellent catalytic activity for ozonation. The efficiency of quinoline mineralization in the catalytic ozonation with CuFe2O4/SEP was 90.3%, and this value was 5.4 times higher than that of the uncatalyzed ozonation (16.8%). The quinoline mineralization followed

Proton Relaying in Carbenium-Based Oxygen Reduction.

The catalytic reduction of oxygen is at the heart of many energy-related and biological processes. Traditionally mediated by transition metals, metal-free systems have emerged as a powerful alternative. In this study, we describe newly prepared quinoline-carbenium species as catalysts for the two-electron reduction of oxygen and show that the quinoline nitrogen atom, depending on its position, can act as a proton relay, thereby boosting the H2O2 production rate. Kinetic studies, H/D kinetic isotope effect measurements, and cyclic voltammetric investigations support a mechanism that involves in

The clinical spectrum and pathogenesis of HIV encephalopathy, myelopathy, and peripheral neuropathy.

Human immunodeficiency virus infection commonly affects the nervous system, with the most important features being a dementing illness, a myelopathy, and a neuropathy. In the past year, important advances have been made in the epidemiology and treatment of these conditions. Moreover, a better understanding of the pathogenetic mechanisms has emerged, with particular emphasis being placed upon gp120, tumor necrosis factor, and quinolinic acid and other partly characterized neurotoxins. This review highlights these advances in the context of the clinical syndromes.

BDNF regulation under GFAP promoter provides engineered astrocytes as a new approach for long-term protection in Huntington's disease.

Brain-derived neurotrophic factor (BDNF) is the main candidate for neuroprotective therapeutic strategies for Huntington's disease. However, the administration system and the control over the dosage are still important problems to be solved. Here we generated transgenic mice overexpressing BDNF under the promoter of the glial fibrillary acidic protein (GFAP) (pGFAP-BDNF mice). These mice are viable and have a normal phenotype. However, intrastriatal administration of quinolinate increased the number of reactive astrocytes and enhanced the release of BDNF in pGFAP-BDNF mice compared with wild-t

Novel tricyclic pyrrolo-quinolines as pharmacological correctors of the mutant CFTR chloride channel.

F508del, the most frequent mutation in cystic fibrosis (CF), impairs the stability and folding of the CFTR chloride channel, thus resulting in intracellular retention and CFTR degradation. The F508del defect can be targeted with pharmacological correctors, such as VX-809 and VX-445, that stabilize CFTR and improve its trafficking to plasma membrane. Using a functional test to evaluate a panel of chemical compounds, we have identified tricyclic pyrrolo-quinolines as novel F508del correctors with high efficacy on primary airway epithelial cells from CF patients. The most effective compound, PP02

Long-Term Neuropsychiatric Sequelae of COVID-19 in an Open Population: A Prospective Pilot Study.

OBJECTIVE: COVID-19 has been associated with a wide range of systemic and neurological complications, known as long COVID or postacute sequelae of COVID-19 (PASC). Such sequelae can be observed among all infected individuals, even among those with a mild disease course. Dysbiosis, a common condition associated with low-grade inflammation, has been proposed as a potential mechanism of PASC by altering levels of circulating lipopolysaccharide (LPS) and the tryptophan pathway metabolites kynurenine and quinolinic acid, known to affect neurocognitive function. The authors evaluated the evolution o…

Neurobehavioral manifestations of symptomatic HIV-1 disease in children: can nutritional factors play a role?

Central nervous system (CNS) abnormalities are significant and frequent complications of human immunodeficiency virus (HIV-1) infection in infants and children. Although the predominant cause of neurological and neuropsychological abnormalities appears to be related to HIV infection of the CNS, other factors including malnutrition may also play a role. We retrospectively evaluated the association of change in body weight with changes in neurocognitive function, ventricular brain ratio, and cerebrospinal quinolinic acid levels in a small cohort of children (n=15; mean age 6.3 years) with sympto…
