The orchid Gastrodia-derived extracellular vesicles deliver miR535 to suppress a fungal effector and facilitate symbiosis with Armillaria.

Plant-derived extracellular vesicles (PDEVs) are nanoscale lipid bilayer structures that facilitate cross-kingdom communication by transporting bioactive molecules, including proteins and small RNAs. While their roles in plant immunity against fungal pathogens are well-established, their functions in beneficial or symbiotic interactions, particularly with mycorrhizal fungi, remain largely unexplored. Here, we investigated the function of PDEVs in the symbiosis between the fully mycoheterotrophic orchid Gastrodia elata and its fungal partner Armillaria gallica. We isolated and characterized G.

Transcriptome datasets of salicylic acid-treated tomato plant cultures under normal and stress conditions.

Extreme weather events pose a significant challenge to agricultural systems, as they induce various abiotic stress responses in plants. Preventive crop protection strategies that aim to enhance plant resistance in advance are gaining increasing importance to mitigate these stress effects. Priming treatments based on the activation of the immune response allow plants to be prepared for stress by triggering some of the defense mechanisms before the actual stress occurs. In the study, healthy, young tomato plants were pre-treated with a foliar spray containing salicylic acid and then subjected to

A P-type pentatricopeptide repeat-containing protein interacts with the Japanese soil-borne wheat mosaic virus movement protein and modulates susceptibility to infection.

As obligate intracellular pathogens, plant viruses interact with host factors to achieve efficient replication and spread. Host factors also play important roles in virus detection and antiviral defence. Furoviruses cause substantial losses in cereals and efficient control measures are lacking. We therefore used Japanese soil-borne wheat mosaic furovirus movement protein (MPJSBWMV) in immunoprecipitation experiments to identify host interaction partners in the model host plant Nicotiana benthamiana. We identified a P-type pentatricopeptide repeat-containing protein (NbPPR) as the interaction p

Molecular translocation between parasitic plants and their hosts.

Parasitic plants are a special group deriving their nutrients from another plant, some of which such as witchweeds (Striga spp.) and broomrapes (Orobanche and Phelipanche spp.) are referred as weeds responsible for severe crop losses in agriculture. The parasite attaches to and feeds off its host using a haustorium, which also facilitates the transport of various molecules between the parasite and its host. These translocation molecules have received extensive attention from researchers. In this review, we summarize the existing knowledge on the transfer of molecules such as pathogens, herbici

Isolation of high quality RNA from bilberry (Vaccinium myrtillus L.) fruit.

A simple and efficient method is described for isolating high quality RNA from bilberry fruit. The procedure is based on the use of hexadecyltrimethyl ammonium bromide (CTAB), polyvinylpyrrolidone (PVP), and beta-mercaptoethanol in an extraction buffer in order to eliminate the polysaccharides and prevent the oxidation of phenolic compounds. This method is a modification of the one described for pine trees, and yields high-quality RNA suitable for cDNA based methodologies. This method is applicable for a variety of plant tissues.

Thapsigargin: key to new host-directed coronavirus antivirals?

Despite the great success of vaccines that protect against RNA virus infections, and the development and clinical use of a limited number of RNA virus-specific drugs, there is still an urgent need for new classes of antiviral drugs against circulating or emerging RNA viruses. To date, it has proved difficult to efficiently suppress RNA virus replication by targeting host cell functions, and there are no approved drugs of this type. This opinion article discusses the recent discovery of a pronounced and sustained antiviral activity of the plant-derived natural compound thapsigargin against enve

Comparative Transcriptomic, Anatomical and Phytohormone Analyses Provide New Insights Into Hormone-Mediated Tetraploid Dwarfing in Hybrid Sweetgum (Liquidambar styraciflua × L. formosana).

Polyploid breeding is an effective approach to improve plant biomass and quality. Both fast growth and dwarf types of in vitro or ex vitro plants are produced after polyploidization. However, little is known regarding the dwarf type mechanism in polyploids grown in vitro. In this study, the morphological and cytological characteristics were measured in tetraploid and diploid hybrid sweetgum (Liquidambar styraciflua × L. formosana) with the same genetic background. RNA sequencing (RNA-Seq) was used to analyse shoot and root variations between tetraploid and diploid plants; important metabolites

The Nucleocapsid Protein of Coronaviruses Acts as a Viral Suppressor of RNA Silencing in Mammalian Cells.

RNA interference (RNAi) is a process of eukaryotic posttranscriptional gene silencing that functions in antiviral immunity in plants, nematodes, and insects. However, recent studies provided strong supports that RNAi also plays a role in antiviral mechanism in mammalian cells. To combat RNAi-mediated antiviral responses, many viruses encode viral suppressors of RNA silencing (VSR) to facilitate their replication. VSRs have been widely studied for plant and insect viruses, but only a few have been defined for mammalian viruses currently. We identified a novel VSR from coronaviruses, a group of

ZmDLR9 is required for lateral root development via pre-rRNA processing in maize.

Pumilio (PUM) proteins comprise an evolutionarily conserved family of sequence-specific RNA-binding proteins that act as key post-transcriptional regulators in eukaryotes. In this study, we identified a maize PUM protein, named ZmDLR9 (Defective in Lateral Root 9), as a positive factor involved in lateral root development. Loss-of-function mutation in the ZmDLR9 (dlr9-1) severely impaired lateral root formation. Functional analysis revealed that ZmDLR9 localizes in the nucleolus and interacts with pre-rRNA processing-associated proteins, including BRX1-1 (BIOGENESIS OF RIBOSOMES IN XENOPUS 1-1

The nucleotidyl transferase URT1-directed mRNA surveillance is critical for embryo and seedling development in Arabidopsis.

In Arabidopsis, the nucleotidyl transferase UTP: RNA URIDYLYLTRANSFERASE (URT1) has been identified as a key mediator of mRNA uridylation and metabolic regulation. While its biochemical function in RNA modification has been established, the physiological relevance of URT1 across different developmental stages remains poorly understood. This study revealed that URT1 exhibits genetic interactions similar to those of EXORIBONUCLEASE 4 (XRN4) during developmental transitions, particularly at the globular embryo stage and during seedling establishment. Simultaneous dysfunction of URT1 and the SUPER

Dynamics and inheritance of the paternal epigenome.

During plant sexual reproduction, the paternal epigenome is remodelled as it progresses from meiocyte to sperm cell and, after fertilization, to the zygote. In flowering plants, sperm cells retain a condensed chromatin state, specialized histone variants and a relatively stable DNA methylation landscape, whereas the companion vegetative cell undergoes chromatin decondensation, DNA demethylation and transposon reactivation. These contrasting epigenetic states are associated with small RNA production and reinforce genome integrity in the male germline. Following fertilization, sperm-specific his

The Regulatory Army of Plant Defense: Transcription Factors in the War for Plant Immunity.

Plant diseases impose major constraints on global crop productivity and pose a major threat to food security. Here, we review transcription factors (TFs) as central orchestrators of plant defense, consolidating recent advances in how these regulators connect pathogen perception to immune signaling, transcriptional reprogramming, and durable defense responses. Initially, we combined a literature-based synthesis with a natural language processing (NLP) analysis of 1647 PubMed abstracts published between 2021 and 2026 to map dominant and underexplored TF families associated with plant immunity. Comparative physiological and transcriptomics analysis revealed crucial mechanisms of silicon-mediated tolerance to iron deficiency in tomato.

Iron (Fe) deficiency is a common abiotic stress in plants grown in alkaline soil that causes leaf chlorosis and affects root development due to low plant-available Fe concentration. Silicon (Si) is a beneficial element for plant growth and can also improve plant tolerance to abiotic stress. However, the effect of Si and regulatory mechanisms on tomato plant growth under Fe deficiency remain largely unclear. Here, we examined the effect of Si application on the photosynthetic capacity, antioxidant defense, sugar metabolism, and organic acid contents under Fe deficiency in tomato plants. The res

Morphological, Transcriptome, and Hormone Analysis of Dwarfism in Tetraploids of Populus alba × P. glandulosa.

Breeding for dwarfism is an important approach to improve lodging resistance. Here, we performed comparative analysis of the phenotype, transcriptome, and hormone contents between diploids and tetraploids of poplar 84K (Populus alba × P. glandulosa). Compared with diploids, the indole-3-acetic acid (IAA) and gibberellin (GA3) contents were increased, whereas the jasmonic acid (JA) and abscisic acid (ABA) contents were decreased in tetraploids. RNA-sequencing revealed that differentially expressed genes (DEGs) in leaves of tetraploids were mainly involved in plant hormone pathways. Most DEGs as

Differential expression of cucumber RNA-dependent RNA polymerase 1 genes during antiviral defence and resistance.

RNA-dependent RNA polymerase 1 (RDR1) plays a crucial role in plant defence against viruses. In this study, it was observed that cucumber, Cucumis sativus, uniquely encodes a small gene family of four RDR1 genes. The cucumber RDR1 genes (CsRDR1a, CsRDR1b and duplicated CsRDR1c1/c2) shared 55%-60% homology in their encoded amino acid sequences. In healthy cucumber plants, RDR1a and RDR1b transcripts were expressed at higher levels than transcripts of RDR1c1/c2, which were barely detectable. The expression of all four CsRDR1 genes was induced by virus infection, after which the expression level

Identification and characterization of an endogenous pararetrovirus in Gentiana triflora plants.

Deep sequencing and de novo assembly of small RNAs (sRNA analysis) provide a powerful approach to detect plant RNA and DNA viruses. Here, using sRNA analysis of Gentiana triflora (gentian) plants affected with kobu-sho disease, a serious disease of gentian, we found caulimovirus-like sequences, provisionally designated as gentian caulimovirus-like sequence (GCVS). Analysis of the complete nucleotide sequence indicated that GCVS is a novel virus with a genomic structure and organization typical of members of genus Caulimovirus (family Caulimoviridae). Based on Southern blot hybridization and ro

Advances in single-cell and spatial multi-omics for plant development and stress responses.

Recent advances in single-cell and spatial multi-omics have enabled high-resolution profiling of plant cellular heterogeneity and tissue organization. Single-cell RNA sequencing (scRNA-seq) captures transcriptional variation across cell types and states, while single-nucleus ATAC sequencing (snATAC-seq) provides information on chromatin accessibility and regulatory elements. Spatial transcriptomics (ST) and spatial metabolomics preserve spatial context, enabling in situ mapping of molecular and metabolic features in plant tissues. This review summarizes experimental and computational approache
