In vitro radiosensitization by pentoxifylline does not depend on p53 status.

PURPOSE: The mode by which the xanthine derivative, pentoxifylline, induces a radiosensitizing effect in cell cultures is a key and controversial radiobiological issue and requires further elucidation. MATERIALS AND METHODS: Six human glioblastoma cell lines were tested for the effect of pentoxifylline treatment at maximum G2/M block on the basis of cell survival, mitotic activity, and micronucleus formation after exposure to gamma radiation. Cell survival was measured by the colony-forming assay. Micronucleus formation (an indicator of DNA damage) and the proportion of binucleated cells (a r

Deinococcus radiodurans - the consummate survivor.

Relatively little is known about the biochemical basis of the capacity of Deinococcus radiodurans to endure the genetic insult that results from exposure to ionizing radiation and can include hundreds of DNA double-strand breaks. However, recent reports indicate that this species compensates for extensive DNA damage through adaptations that allow cells to avoid the potentially detrimental effects of DNA strand breaks. It seems that D. radiodurans uses mechanisms that limit DNA degradation and that restrict the diffusion of DNA fragments that are produced following irradiation, to preserve gene

Hypersensitivity of ataxia telangiectasia fibroblasts to ionizing radiation is associated with a repair deficiency of DNA double-strand breaks.

We have studied the intrinsic radiosensitivity, repair of potentially lethal damage (PLD) and the repair rate of radiation-induced DNA double-strand breaks (DSB) in 11 non-transformed human fibroblast cell lines, four of which were homozygous for the A-T mutation and two that were heterozygous (A-TH). All the experiments were done on cells in plateau phase of growth (97-99% of cells in G0/G1). With a dose of 30 Gy delivered at 4 degrees C, the A-T cell lines had faster repair rates of up to 6 h, after which the repair curve crossed that of the control so that the residual damage at 24 h was hi

Radiation sensitivity of visual and oculomotor pathways.

Adverse effects of stereotactic radiosurgery on cranial nerves, especially the optic nerve and its pathways, are not yet sufficiently understood. 29 patients who underwent Gamma Knife radiosurgery for benign skull base tumors were reviewed. In all of them, parts of the visual pathways and/or other cranial nerves in the middle cranial fossa received significant doses of radiation. The dose given was correlated with neuro-ophthalmological findings during a follow-up period of 6-24 months. Cranial nerves III, IV and VI within the cavernous sinus, exposed to a dose from 4.5 to 30 Gy, did not devel

[Light protection and increase of UV tolerance].

Topical sun-ray filter creams protect the skin from sunburn and from long-term damages caused by UV-radiation. The choice of the sun protecting factor depends on the radiation dose as well as on the individual sensitivity to UV-radiation. For the prevention of photodermatoses, the industry offers broad spectrum absorbents. For internal prophylaxis, oral ingestion of a new preparation containing 25 mg beta-carotene and 35 mg canthaxanthin per dragee, has proved to restore the tolerance of sunlight with polymorphic light eruption, light urticaria, protoporphyria, and other photodermatoses to a c

The Birth of the Illegitimate Linear No-Threshold Model: An Invalid Paradigm for Estimating Risk Following Low-dose Radiation Exposure.

This paper examines the birthing process of the linear no-threshold model with respect to genetic effects and carcinogenesis. This model was conceived >70 years ago but still remains a foundational element within much of the scientific thought regarding exposure to low-dose ionizing radiation. This model is used today to provide risk estimates for cancer resulting from any exposure to ionizing radiation down to zero dose, risk estimates that are only theoretical and, as yet, have never been conclusively demonstrated by empirical evidence. We are literally bathed every second of every day in lo

Optically guided stereotactic radiotherapy for lacrimal sac tumors: a report on two cases.

Adjuvant radiation treatment is often offered for lacrimal sac tumors. However, due to the adjacent critical structures, conventional radiation technique may result in severe side effects. We have treated two patients with lacrimal sac tumors using optically guided stereotactic radiotherapy. One patient with lacrimal sac melanoma was treated with optical-guidance intensity-modulated radiotherapy (IMRT). The other with mixed transitional and squamous cell carcinoma was treated with optical-guidance 3-D conformal radiation. Dose volume analysis revealed excellent target coverage and sparing of c

A Microenvironment-Adaptive Nanoadjuvant Resolves the Radioprotection and Radiosensitization Paradox in Cancer Therapy.

The intrinsic spatiotemporal discord between augmenting tumor radiosensitivity and safeguarding normal tissue has persistently circumscribed the therapeutic horizon of radiotherapy. Here, we introduce a microenvironment-adaptive platform, cross-linked lipoic acid nanoadjuvant (cLAN), designed to strategically navigate this dilemma. Upon cellular internalization, cLAN dissociates into the lipoic acid (LA)/dihydrolipoic acid (DHLA) redox pair, which elicits distinct biological responses in normal vs tumor cells. In irradiated normal cells, the LA/DHLA system primarily scavenges radiation-induced

Micronuclei expression in tumors as a test for radiation sensitivity.

Dose-effect curves were determined for the frequency of micronuclei and impairment of cell clonogenicity from two types of tumours of different sensitivity irradiated in situ. Micronucleated cells were measured 24, 48 and 72 h after treatment. The quantitative relationships between cell reproductive death and the induction of micronuclei are the same for both tumours.

[Thyroid and radiation].

The topic "Thyroid and Radiation" is both an old and a new area to be solved by human beings. The thyroid is an organ that is usually susceptible to exposure to ionizing radiation, both by virtue of its ability to concentrate radioiodine (internal radiation) and by routine medical examination: Chest X-ray, Dental X-ray, X-irradiation of cervical lymphnodes etc. (external radiation). Iodine-131 is widely used for the therapy of Graves' disease and thyroid cancers, of which the disadvantage is radiation-induced hypothyroidism but not complications of thyroid tumor. The thyroid gland is comparati

Brain-Specific Relative Biological Effectiveness of Protons Based on Long-term Outcome of Patients With Nasopharyngeal Carcinoma.

PURPOSE: Uncertainties in relative biological effectiveness (RBE) constitute a major pitfall of the use of protons in clinics. An RBE value of 1.1, which is based on cell culture and animal models, is currently used in clinical proton planning. The purpose of this study was to determine RBE for temporal lobe radiographic changes using long-term follow-up data from patients with nasopharyngeal carcinoma. METHODS AND MATERIALS: Five hundred sixty-six patients with newly diagnosed nasopharyngeal carcinoma received double-scattering proton therapy or intensity modulated radiation therapy at our i

Overcoming Radiation Resistance: Ferroptosis Induction to Sensitize Solid Tumors to Radiation Therapy.

Radiation therapy (RT) is a mainstay of treatment for a myriad of cancers, often utilized for tumors that are unable to be resected, as well as an adjunct to surgery and chemotherapy. Unfortunately, many cancers are resistant to RT-induced damage and subsequent cell death. This has spurred the pursuit of novel radiation sensitizers that could potentiate the effects of this widely used and important treatment modality. Since its discovery as a regulated cell death mechanism in 2012, ferroptosis has been studied for its connection to cancer and other known oxidative pathways. With this, the inte

Mathematical models of radiation action on living cells: From the target theory to the modern approaches. A historical and critical review.

Cell survival is conventionally defined as the capability of irradiated cells to produce colonies. It is quantified by the clonogenic assays that consist in determining the number of colonies resulting from a known number of irradiated cells. Several mathematical models were proposed to describe the survival curves, notably from the target theory. The Linear-Quadratic (LQ) model, which is to date the most frequently used model in radiobiology and radiotherapy, dominates all the other models by its robustness and simplicity. Its usefulness is particularly important because the ratio of the valu

[Pharmacological targeting of Mdm2: rationale and perspectives for radiosensitization].

The central role of p53 after exposure to ionizing radiation has been widely demonstrated. Mdm2, the main cellular regulator of p53, is a promising target for radiosensitizing purposes. In this article, we review the most recent data on the pharmacological targeting of Mdm2, with focus on strategies of radiosensitization. Antitumor activity of Mdm2 inhibitors has been related with activation of p53-dependant apoptosis, action on DNA repair systems, and antiangiogenic activity. Preliminary data suggested a synergic interaction between Mdm2 inhibitors and ionizing radiations. However, no clinica

Thorium-induced oxidative stress mediated toxicity in mice and its abrogation by diethylenetriamine pentaacetate.

PURPOSE: Thorium ((232)Th, IV) preferentially accumulates in the liver, femur and spleen, which necessitates evaluation of its toxic effect in these organs. The present study was aimed at evaluation of liver function, oxidative stress and histological alterations in these organs. MATERIALS AND METHODS: Swiss albino mice were administered either with Thorium nitrate (10 mg/kg body weight/day equivalent to 1,090 pCi/kg body weight/day) for 30 days (1/40th dose of LD(50/30); the dose of thorium required to kill 50% of the test cohort within 30 days) intraperitoneally or with calcium salt of diet

A critical look at the association of human genetic syndromes with sensitivity to ionizing radiation.

Individuals with inherited heterozygosity for mutations involving tumor suppressor genes may be at greater risk for ionizing radiation-induced cancer even though their cells may not show increased cytotoxicity or chromosome damage. In addition, many human genetic syndromes have been reported to show increased sensitivity to ionizing radiation. In most instances the effects are small and/or not reproducible. Only in the genetic disease ataxia-telangiectasia (AT) is radiosensitivity consistently outside the normal range. Even AT heterozygotes (0.68 to 7.7% of the population) appear to be slightl

[Restoration of the skin of mice following damages cause by ionizing radiation and hyperthermia].

The half-period of mouse skin regeneration after sublethal injuries caused by hyperthermia (44 degrees C) was 2.9 h and completed within 16-24 h. The half-period of regeneration after sublethal injuries caused by ionizing radiation was 2.1 h at a dose of 5 Gy and 4.3 at a dose of 20 Gy. The rate of mouse skin regeneration after sublethal injuries caused by exposure to ionizing radiation only and in combination with hyperthermia at similar levels of injury did not differ.

Effect of X irradiation on the rectum of the rat.

It is demonstrated how a definite part of the rectum of the female Wistar rat can be selectively irradiated without surgery. The effect of X irradiation on the rectum is in the acute phase (second week after irradiation) a haemorrhagic radiation proctitis and in the chronic phase a rectal obstruction. The dose-effect relationship using rectal obstruction as the end point is a sigmoid curve with a ED10 of 17.50 Gy, ED50 of 21.5 Gy and ED90 of 27.5 Gy. Latency time decreases exponentially with increasing dose and is 150 days at the ED50. Histological findings show mucosal glands and cysts within

[Application of the NSD-concept for the ascertainment of tolerance doses in the region of the lung (author's transl)].

A clinical application of the NSD-concept using iso-ret plans for the recording of radiation pneumonitis is reported. The conditions of an ascertainment of tolerance values for single organs by help of the Ellis formula are discussed. Of the examined patients with Hodgkin's disease who had been exposed to irradiation of the unaffected mediastinum, 42% suffered from radiogenic lung complications. The calculated iso-ret plans reveal a limitation of the pneumonitic shadow zone identical to the 1200-ret isoline. The data in literature are in accordance with these results.
