Generation of AQP2-Cre transgenic mini-pigs specifically expressing Cre recombinase in kidney collecting duct cells.

The important differences in physiological parameters and anatomical characteristics of the kidney between humans and mice make it difficult to replicate the precise progression of human renal cystic diseases in gene modification mouse models. In contrast to mice, pigs are a better animal model of human diseases, as they are more similar in terms of organ size, structure, and physiological parameters. Here, we report the generation and initial examination of an AQP2-Cre transgenic (Tg) Chinese miniature (mini)-pig line that expresses Cre recombinase exclusively in kidney collecting duct cells.

Development and evaluation of a recombinase-aided amplification-lateral flow dipstick assay for rapid and sensitive detection of Mycoplasma synoviae.

BACKGROUND: Mycoplasma synoviae (MS) is a critical avian pathogen responsible for enormous economic losses in global poultry production. Timely prevention and control of MS epidemics rely heavily on rapid, sensitive and accurate diagnostic approaches, while conventional detection methods remain time-consuming and complicated. Therefore, we developed a visual on-site detection system based on recombinase-aided amplification combined with lateral flow dipstick for MS (RAA-LFD-MS assay). METHODS: According to the design rules for RAA primers and exo-probes, specific primers and a probe were desi

An Enzymatic Recombinase Amplification Coupled Lateral Flow Assay for Rapid, Visual, and Cost-Effective Detection of Bordetella pertussis.

Bordetella pertussis, the causative agent of pertussis, is a highly contagious respiratory pathogen. Rapid and reliable diagnostics are essential for early detection and disease control. Here, we developed an enzymatic recombinase amplification–lateral flow dipstick (ERA-LFD) assay targeting the IS1663 gene of B. pertussis. The assay achieves high sensitivity with detection limit of 10 copies/µL, and analytical specificity reached 100%. It was performed at 32 ℃ for 15 min, with visual results obtainable within 20 min. Clinical evaluation of 50 nasopharyngeal swab samples demonstrated a sensiti

[Development and application of a triplex basic- recombinase polymerase amplification method to detect Brucella, Babesia, and Pasteurella multocida in cattle].

To combat three major pathogens-Brucella spp.,Babesia bovis, and Pasteurella multocida-threatening cattle health, we developed a rapid and accurate triplex basic-recombinase polymerase amplification (Basic-RPA) detection system. Targeting bcsp31, rap-1, and kmt1, we designed and screened specific RPA primers and optimized the reaction temperature, time, and primer concentration. The best reaction conditions were 38 ℃ for 25 min, with primers at 10 μmol/L in a 1:1:1 ratio. We then used reference and non-target strains to evaluate the specificity of the established method and serial DNA dilution

Advances in the recombinase polymerase amplification platform for urogenital schistosomiasis diagnosis (ShDraI-RPA) towards implementation at the point-of-care.

Accurate diagnosis of schistosomiasis is crucial to achieve disease elimination as a public health problem. Rapid and highly sensitive diagnostic tools that can be used in decentralized environments and/or at the point-of-care are needed. This work optimises and simplifies an existing isothermal molecular diagnostic platform (recombinase polymerase amplification, RPA) for urogenital schistosomiasis, the ShDraI-RPA, with a focus on delivering a more accurate diagnosis in endemic settings. The standard ShDraI-RPA oligonucleotides were modified, incorporating a phosphorothioate backbone into the

Development of a Recombinase-Mediated Cassette Exchange System for Gene Knockout and Expression of Non-Native Gene Sequences in Rickettsia.

BACKGROUND/OBJECTIVES: Incidence of vector-borne diseases, including rickettsioses and anaplasmosis, has been increasing in many parts of the world. The obligate intracellular nature of rickettsial pathogens has hindered the development of robust genetic tools for the study of gene function and the identification of therapeutic targets. Transposon mutagenesis has contributed to recent progress in the identification of virulence factors in this important group of pathogens. METHODS: Combining the efficiency of the himar1 transposon method with a recombinase-mediated system, we aimed to develop

A multiplexed, allele-specific recombinase polymerase amplification assay with lateral flow readout for sickle cell disease detection.

Isothermal nucleic acid amplification tests have the potential to improve disease diagnosis at the point of care, but it remains challenging to develop multiplexed tests that can detect ≥3 targets or to detect point mutations that may cause disease. These capabilities are critical to enabling informed clinical decision-making for many applications, such as sickle cell disease (SCD). To address this, we describe the development of a multiplexed allele-specific recombinase polymerase amplification (RPA) assay with lateral flow readout. We first characterize the specificity of RPA using primer de

G9a inactivation in progenitor cells with Isl1-Cre with reduced recombinase activity models aspects of Dandy-Walker complex.

G9a, also known as EHMT2, is essential for embryogenesis and has specific functions in multiple developmental processes. G9a inactivation affects development of the nervous system, which is formed with contribution of descendants of progenitor cells expressing the transcription factor Isl1. However, the function of G9a in Isl1-expressing progenitors is unknown. Here, we show that G9a is required for proper development of multiple structures formed with contribution of Isl1-expressing progenitors. A Cre-dependent GFP reporter revealed that the recombinase activity of the Isl1-Cre used in this s

Leak-proof probe for accurate detection of Neisseria gonorrhoeae by recombinase polymerase amplification-mediated lateral flow strip.

Neisseria gonorrhoeae is the only pathogen contributing to gonorrhea, a common infectious disease. Clinically, approximately 50-80% of female and 40% of male patients are asymptomatic, and these carriers are the key to gonorrhea transmission. The rapid detection of N. gonorrhoeae recessive infection is vital to curb the spread of gonorrhea. Therefore, the development of a specific, sensitive, rapid, and convenient method for the diagnosis of N. gonorrhoeae is a priority. In this study, we identified the highly conserved fitA gene of N. gonorrhoeae as a detection target through bioinformatics a

A novel mouse line with epididymal initial segment-specific expression of Cre recombinase driven by the endogenous Lcn9 promoter.

Spermatozoa released from testes undergo a maturation process and acquire the capacity to fertilize ova through epididymal transit. The epididymis is divided into four regions, each with unique morphology, gene profile, luminal microenvironment and distinct function. To study the functions of relevant genes in the epididymal initial segment (IS), a novel IS-specific mouse model, Lcn9-Cre knock-in (KI) mouse line was generated via CRISPR/Cas9 technology. The TAG stop codon was replaced by a 2A-NLS-Cre cassette, resulting in the co-expression of Lcn9 and Cre recombinase. IS-specific Cre expressi
