Effects of Bonito Elastin HC on Skin Dryness, Wrinkles, and Pigmentation In Vitro and In Vivo.

We investigated the effects of bonito fish (Katsuwonus pelamis) elastin HC (KE) on skin dryness, wrinkles, and pigmentation in vitro and in vivo. In vitro, we evaluated the expression of mRNA genes and proteins related to skin dryness, wrinkles, and pigmentation. HaCaT and HS27 cells were exposed to ultraviolet B radiation (UVB) (50 mJ/cm2), and B16F10 cells were stimulated with 3-isobutyl-1-methylxanthine (IBMX, 250 μg/mL) for 72 h to induce melanin synthesis. All cells were treated with KE (50-400 μg/mL) for 24 h. We found that KE increased the expression of long-chain base 1, dihydroceramid

Genetic loci associated with skin pigmentation in African Americans and their effects on vitamin D deficiency.

A recent genome-wide association study (GWAS) in African descent populations identified novel loci associated with skin pigmentation. However, how genomic variations affect skin pigmentation and how these skin pigmentation gene variants affect serum 25(OH) vitamin D variation has not been explored in African Americans (AAs). In order to further understand genetic factors that affect human skin pigmentation and serum 25(OH)D variation, we performed a GWAS for skin pigmentation with 395 AAs and a replication study with 681 AAs. Then, we tested if the identified variants are associated with serum…

Deciphering skin re-pigmentation patterns in vitiligo: an update on the cellular and molecular events involved.

Current treatment of vitiligo is still a great challenge, since most cases of vitiligo have variable re-pigmentation outcomes due to their unpredictable responses to existing therapeutic regimens. There is an urgent need to identify this re-pigmentation process and to develop novel therapies. This review illustrates the most current research and latest understanding of vitiligo skin re-pigmentation and related regulatory mechanisms. Literature was collected from PubMed until January 2020, using the search terms including "vitiligo," "re-pigmentation," "phototherapy," "narrow-band ultraviolet B

The Sequelae of Metallosis Resulting in Skin Pigmentation and Tattooing: A Case Presentation and Literature Review.

With advancing technologies in orthopedics and increasing demands of the population for orthopedic interventions, younger patients are now receiving joint replacements. One of the potential risks of joint replacement is metallosis, or the local and systemic release of metal ions. Metallosis is caused by the release of metallic debris, secondary to hardware failure. The phenomenon is most commonly associated with failed metal-on-metal hip prostheses and is characterized locally by heavy staining of surrounding soft tissue, metallic synovitis, joint effusion, and gradual loosening of the prosthe

Possible contributions of skin pigmentation and vitamin D in a polyfactorial model of seasonal affective disorder.

Seasonal affective disorder (SAD) is a polyfactorial and polygenetic disorder that involves biological and psychological sub-mechanisms that … ses the circadian timing systems and much of its neural circuitry. We also consider the variable of skin pigmentation as this may affect levels of vitamin D in the body. We hypothesize that people with darker skin pigmentation may experience greater risks for lower vitamin D levels that, especially following their migration to regions of higher latitude, could cont …

Genome-wide association studies of quantitatively measured skin, hair, and eye pigmentation in four European populations.

Pigmentation of the skin, hair, and eyes varies both within and between human populations. Identifying the genes and alleles underlying this … ean genetic differentiation. The country of sampling explains approximately 35% of the variation in skin pigmentation, 31% of the variation in hair pigmentation, and 40% of the variation in eye pigmentation. All three quantitative phenotypes are correlated with each other. In our two-stage association study, we reproduce the association of rs1667394 at the OCA2/HER …

Double trouble from sunburn: UVB-induced erythema is associated with a transient decrease in skin pigmentation.

BACKGROUND: Ultraviolet B (UVB) radiation may cause an immediate inflammatory response followed by a delayed increase in skin pigmentation. The early time course of erythema and pigmentation has so far not been monitored simultaneously by photometric measurements. METHODS: Test areas on the volar forearms of 15 volunteers were irradiated with 210 mJ/cm(2) UVB. Skin erythema and pigmentation were determined photometrically at time 0, after 6 h, and after 1, 2, 3 and 7 days. Punch biopsies were taken before irradiation, after 6 h and after 7 days. Melanocytes were stained using the DOPA method.…

Isosexual pseudoprecocious puberty in a 2(1/2)-year-old girl presenting with intense skin pigmentation.

BACKGROUND: Ovarian follicular cyst producing estradiol is a rare cause of isosexual pseudoprecocious puberty. Intense pigmentation of breast papillae, areolae, and labia minora is also rarely reported in the literature. CASE: We describe a 2(1/2) year old girl presenting with signs of precocious puberty and advanced bone age due to a large follicular cyst. Estradiol and Dehydro-epiandrosterone sulfate (DHEAS) levels were remarkably elevated. Hyperpigmentation was also noted. Salpingoophorectomy resulted in regression of precocity and depigmentation, but DHEAS serum levels remained elevated.

Pigmentation and skin reaction to sun as risk factors for cutaneous melanoma: Western Canada Melanoma Study.

Between 1 April 1979 and 31 March 1981, 904 residents of the four western provinces of Canada (population 6.5 million), were diagnosed as suffering from primary cutaneous malignant melanoma. Of 801 patients aged 20-79 years, 665 (83%) were interviewed along with control subjects chosen at random from the general population and matched for age, sex, and province. After exclusion of 70 subjects with lentigo maligna or acral lentiginous melanoma, comparisons of the 595 case-control pairs showed that light hair, skin, and eye colour, a history of heavy freckling in adolescence, and a tendency to b

Monitoring of Pigmented Skin Lesions Using 3D Whole Body Imaging.

BACKGROUND AND OBJECTIVES: Advanced artificial intelligence and machine learning have great potential to redefine how skin lesions are detected, mapped, tracked and documented. Here, we propose a 3D whole-body imaging system known as 3DSkin-mapper to enable automated detection, evaluation and mapping of skin lesions. METHODS: A modular camera rig arranged in a cylindrical configuration was designed to automatically capture images of the entire skin surface of a subject synchronously from multiple angles. Based on the images, we developed algorithms for 3D model reconstruction, data processing

Analytical Characteristics of a Noninvasive Gene Expression Assay for Pigmented Skin Lesions.

We previously reported clinical performance of a novel noninvasive and quantitative PCR (qPCR)-based molecular diagnostic assay (the pigmented lesion assay; PLA) that differentiates primary cutaneous melanoma from benign pigmented skin lesions through two target gene signatures, LINC00518 (LINC) and preferentially expressed antigen in melanoma (PRAME). This study focuses on analytical characterization of this PLA, including qPCR specificity and sensitivity, optimization of RNA input in qPCR to achieve a desired diagnostic sensitivity and specificity, and analytical performance (repeatability a

Molecular changes in skin pigmented lesions of the coral trout Plectropomus leopardus.

A high prevalence of skin pigmented lesions of 15% was recently reported in coral trout Plectropomus leopardus, a commercially important marine fish, inhabiting the Great Barrier Reef. Herein, fish were sampled at two offshore sites, characterised by high and low lesion prevalence. A transcriptomic approach using the suppressive subtractive hybridisation (SSH) method was used to analyse the differentially expressed genes between lesion and normal skin samples. Transcriptional changes of 14 genes were observed in lesion samples relative to normal skin samples. These targeted genes encoded for s

Determination of border irregularity in dermoscopic color images of pigmented skin lesions.

Malignant melanoma, which is the most dangerous type of skin cancer, is commonly diagnosed in all people, regardless of age, gender, or race. In the last several years an increasing melanoma incidence and mortality rate has been observed worldwide. In this research we present a new approach to the detection and classification of border irregularity, one of the major parameter in a widely used diagnostic algorithm ABCD rule of dermoscopy. Accurate assessment of irregular borders is clinically important due to a significantly different occurrence in benign and malignant skin lesions. In this pap

Pigmented Basal Cell Carcinoma in Skin of Color: A Systematic Review of the Literature.

BACKGROUND: Though basal cell carcinoma (BCC) is most often studied in Fitzpatrick Skin Types (FST) I-III, BCC in skin of color (SOC) is characterized by unique clinical features and a propensity toward pigmentation. While cutaneous malignancy occurs with decreased frequency in FST IV - VI, the diagnosis is generally associated with disproportionately increased morbidity and mortality. Accordingly, it is increasingly vital for dermatologists to recognize clinical presentations of skin cancer in individuals with SOC to improve outcomes and mitigate gaps in care. OBJECTIVE: We systematically re

Presentations of Cutaneous Disease in Various Skin Pigmentations: Chronic Atopic Dermatitis.

Description Atopic dermatitis is a chronic inflammatory skin disorder classically affecting flexural areas of the body. It is present in children and adults, including those with darker skin pigmentation. Chronic lesions are hyperpigmented plaques that are dry, cracked, and/or scaly often with lichenification. Differential diagnoses include psoriasis, seborrheic dermatitis, ichthyosis, and pityriasis rosea. This article will showcase clinical images with varying presentations of chronic atopic dermatitis in a range of age groups and skin colors according to the Fitzpatrick scale.

Color Biophysics and the Human Color Perception in Relation to Pigments and Tattooed Skin.

Perception of skin color in the natural state, or with tattoo pigment installed in the dermis is highly complex and includes both optical requisites and eye and brain functions connected to advanced cognitive and psychological functions. Permanent makeup (PMU) is miniaturized tattooing on critical anatomical location aiming at fulfilment of a person's esthetic ideal. It is a special challenge since a satisfactory outcome depends on perfectionated technique and very critical selection of pigments. The eye can be compared to a digital camera, and the center of central vision in the brain to a co
