NSAIDs and Cancer Resolution: New Paradigms beyond Cyclooxygenase.

Acute inflammation or resolved inflammation is an adaptive host defense mechanism and is self-limiting, which returns the body to a state of homeostasis. However, unresolved, uncontrolled, or chronic inflammation may lead to various maladies, including cancer. Important evidence that links inflammation and cancer is that nonsteroidal anti-inflammatory drugs (NSAIDs), such as aspirin, reduce the risk and mortality from many cancers. The fact that NSAIDs inhibit the eicosanoid pathway prompted mechanistic drug developmental work focusing on cyclooxygenase (COX) and its products. The increased pr

Untangling the web of 5-lipoxygenase-derived products from a molecular and structural perspective: The battle between pro- and anti-inflammatory lipid mediators.

Arachidonic acid (AA) is the precursor to leukotrienes (LT), potent mediators of the inflammatory response. In the 35 + years since cysteinyl-LTs were reported to mediate antigen-induced constriction of bronchi in tissue from asthma patients, numerous cellular responses evoked by the LTs, such as chemoattraction and G protein-coupled receptor (GPCR) activation, have been elucidated and revealed a potential for 5-lipoxygenase (5-LOX) as a promising drug target that goes beyond asthma. We describe herein early work identifying 5-LOX as the key enzyme that initiates LT biosynthesis and the discov

Dysregulated NK cell phenotype in exacerbation-prone severe asthma.

BACKGROUND: Natural killer (NK) cell dysfunction and defective inflammation resolution pathways are associated with chronic airway inflammation in asthma. NK cells are targets for regulation by essential polyunsaturated fatty acid-derived specialized pro-resolving mediators (SPMs), including resolvin D2 (RvD2), to stimulate inflammation resolution. METHODS: Multiparameter flow cytometry was utilized to examine expression patterns of select activating and inhibitory receptors, including the RvD2 receptor DRV2, on circulating NK cells in SARP3 severe and non-severe asthma subjects and healthy c

Modulation of inflammatory pathways by omega-3 fatty acids in knee joint health for the management of post-traumatic osteoarthritis: a review.

The knee joint facilitates movement essential for daily activities. Its intricate anatomy makes it particularly prone to injury, specifically to the anterior cruciate ligament (ACL). ACL injury is a leading cause of post-traumatic osteoarthritis (PTOA), a degenerative joint disease that affects millions and results in pain, decreased mobility, and reduced quality of life. The cascade of initial inflammatory activity triggered by ACL injury plays a critical role in the pathogenesis of PTOA, as elevated levels of pro-inflammatory cytokines and catabolic enzymes lead to ongoing joint damage and d

Maresin 1, a Proresolving Lipid Mediator, Ameliorates Liver Ischemia-Reperfusion Injury and Stimulates Hepatocyte Proliferation in Sprague-Dawley Rats.

Maresin-1 (MaR1) is a specialized pro-resolving mediator, derived from omega-3 fatty acids, whose functions are to decrease the pro-inflammatory and oxidative mediators, and also to stimulate cell division. We investigated the hepatoprotective actions of MaR1 in a rat model of liver ischemia-reperfusion (IR) injury. MaR1 (4 ng/gr body weight) was administered prior to ischemia (1 h) and reperfusion (3 h), and controls received isovolumetric vehicle solution. To analyze liver function, transaminases levels and tissue architecture were assayed, and serum cytokines TNF-α, IL-6, and IL-10, mitotic

Dietary bioactive compounds and inflammaging: Pro-inflammatory triggers and geroprotective countermeasures.

BACKGROUND: Chronic low-grade inflammation ("inflammaging") is a key driver of age-related pathologies including cardiovascular disease, neurodegeneration, and metabolic syndrome. Diet plays a dual role in modulating this process, acting both as a source of pro-inflammatory molecular patterns and as a delivery system for geroprotective compounds. SCOPE AND APPROACH: This review examines the pro-inflammatory dietary components (advanced glycation end products, lipid peroxidation products, oxysterols, trans fats, and microbiome-derived metabolites) that activate pattern recognition receptors an

Resolution-Oriented Immunopharmacological Potential of the Lipid Matrix of Rhynchophorus palmarum Larvae in Viral Respiratory Infections: A Perspective Review.

This review explores how the fatty acid (FA) profile of the Amazonian Rhynchophorus palmarum larva (Chontacuro), traditionally eaten for respiratory ailments, might biochemically mitigate viral respiratory infections. During metamorphosis, this insect utilizes its unique FA matrix to survive severe, self-inflicted oxidative and inflammatory tissue stress. The authors hypothesize that when consumed by humans, these insect lipids undergo chylomicron-mediated lymphatic transport. This pathway bypasses initial liver metabolism, delivering unsaturated FAs directly to the pulmonary microvasculature

How to Deal with Pulpitis: An Overview of New Approaches.

Background: Traditional root canal therapy (RCT) effectively removes diseased or necrotic pulp tissue and replaces it with inorganic materials. Regenerative endodontics is an alternative to conventional RCT by using biologically based approaches to restore the pulp-dentin complex. This review explores emerging techniques, including autogenic and allogenic pulp transplantation, platelet-rich fibrin, human amniotic membrane scaffolds, specialized pro-resolving mediators, nanofibrous and bioceramic scaffolds, injectable hydrogels, dentin matrix proteins, and cell-homing strategies. These methods

Preventive and Therapeutic Effects of Intracerebroventricular Administration of Maresin-1 on Lipopolysaccharide-Induced Depression-Like Behaviors in Mice.

Enhanced inflammatory and immune responses have been observed in patients with major depressive disorder, pointing to anti-inflammatory substances as potential seeds for developing novel antidepressants. Omega-3 polyunsaturated fatty acid metabolites, such as resolvin D and E series, maresins, and protectins (collectively known as specialized pro-resolving mediators) demonstrate anti-inflammatory effects. This study examined the antidepressant-like effects of maresin-1 (MaR1) on lipopolysaccharide (LPS)-induced depression-like behaviors in mice. Using the tail suspension test (TST) and the for

Stretching Reduces Tumor Growth in a Mouse Breast Cancer Model.

There is growing interest in developing non-pharmacological treatments that could boost natural defenses against cancer and contribute to primary and secondary cancer prevention. Recent studies have shown that gentle daily stretching for 10 minutes can reduce local connective tissue inflammation and fibrosis. Because mechanical factors within the stroma can influence the tumor microenvironment, we hypothesized that stretching would reduce the growth of tumors implanted within locally stretched tissues and tested this hypothesis in a mouse orthotopic breast cancer model. Female FVB mice (N = 66

Neutrophils and periodontitis: From pathological mechanisms to therapeutic opportunities.

Periodontitis (PD) is a prevalent inflammatory disease causing gingival destruction and alveolar bone loss. Neutrophils dominate periodontal lesions and, while essential for host defense, become pathogenic when dysregulated. Their hyperactivated antimicrobial functions-phagocytosis, oxidative burst, degranulation, and neutrophil extracellular trap (NET) formation-drive oxidative stress, protease release, and aberrant NETosis, leading to tissue and bone damage. Cytokines, chemokines, microbial virulence factors, and aging further shape neutrophil responses, but chronic overactivation remains ce

Nature of the Association between Rheumatoid Arthritis and Cervical Cancer and Its Potential Therapeutic Implications.

It is now established that patients with rheumatoid arthritis (RA) have an increased risk of developing cervical cancer (CC) or its precursor, cervical intraepithelial neoplasia (CIN). However, the underlying mechanisms of this association have not been elucidated. RA is characterized by unresolved chronic inflammation. It is suggested that human papillomavirus (HPV) infection in RA patients exacerbates inflammation, increasing the risk of CC. The tumor microenvironment in RA patients with CC is also marked by chronic inflammation, which aggravates the manifestations of both conditions. Gut an

Loss of 15-Lipoxygenase in Retinodegenerative RCS Rats.

Retinitis pigmentosa (RP) is a retinal degenerative disease associated with a diversity of genetic mutations. In a natural progression study (NPS) evaluating the molecular changes in Royal College of Surgeons (RCS) rats using lipidomic profiling, RNA sequencing, and gene expression analyses, changes associated with retinal degeneration from p21 to p60 were evaluated, where reductions in retinal ALOX15 expression corresponded with disease progression. This important enzyme catalyzes the formation of specialized pro-resolving mediators (SPMs) such as lipoxins (LXs), resolvins (RvDs), and docosap

Reduced Levels of Plasma Lipoxin A4 Are Associated with Post-Stroke Cognitive Impairment.

BACKGROUND: Specialized pro-resolving mediators (SPMs) are bioactive lipids derived from n-3 and n-6 polyunsaturated fatty acids. SPMs promote resolution of inflammation and are reduced in Alzheimer's disease. It is unknown whether SPMs are associated with post-stroke cognitive impairment (PSCI). OBJECTIVE: In the present report, we aimed to study the levels of SPMs in PSCI patients in the acute phase of ischemic stroke. METHODS: Levels of SPMs in the plasma from 36 patients with PSCI and 33 patients with post-stroke non-cognitive impairment (PSNCI) were measured by enzyme immunoassay. RESU

Receptors for pro-resolving mediators are increased in Alzheimer's disease brain.

Neuroinflammation is a key element of AD pathology and conceivably a result of a disturbed resolution. Resolution of inflammation is an active process which is strictly orchestrated following the acute inflammatory response after removal of the inflammatory stimuli. Acute inflammation is actively terminated by specialized pro-resolving mediators (SPMs) thereby promoting healing and return to homeostasis. Failed resolution may contribute to persistent neuroinflammation and aggravate AD pathology. BLT1 (leukotriene B4 receptor) and ChemR23 (chemerin receptor 23) are receptors for the SPM resolvi

Omega-3 long chain fatty acids and their metabolites in pregnancy outcomes for the modulation of maternal inflammatory- associated causes of preterm delivery, chorioamnionitis and preeclampsia.

Preterm birth is a major cause of perinatal complications and neonatal deaths. Furthermore, in the field of obstetrics many clinical entities like uterine contractions or the occurrence of pre- eclampsia remain to be serious complications during pregnancy and represent a major psychological, financial, and economic burden for society. Several published guidelines, studies and recommendations have highlighted the importance of supplementation of omega-3 long chain polyunsaturated fatty acids (PUFAs) during pregnancy. This narrative review aims at giving an overview on the modern perception of i

Specialized pro-resolving receptors are expressed in salivary glands with Sjögren's syndrome.

Our previous studies demonstrated that resolvin D1 (RvD1) and its aspirin-trigged (AT) form AT-RvD1, are effective in decreasing inflammation while restoring saliva flow rates in a Sjögren's syndrome (SS)-like mouse model before and after disease onset. Resolvins are specialized pro-resolving mediators (SPM) that actively regulate inflammation. However, we only have extensive data within the salivary glands for RvD1 and AT-RvD1, both of which bind to the receptor ALX/FPR2. As such, the presence of other SPM receptors is unknown within salivary glands. Therefore, the goal of this study was to d

Inflammaging as a common ground for the development and maintenance of sarcopenia, obesity, cardiomyopathy and dysbiosis.

Sarcopenia, obesity and their coexistence, obese sarcopenia (OBSP) as well as atherosclerosis-related cardio-vascular diseases (ACVDs), including chronic heart failure (CHF), are among the greatest public health concerns in the ageing population. A clear age-dependent increased prevalence of sarcopenia and OBSP has been registered in CHF patients, suggesting mechanistic relationships. Development of OBSP could be mediated by a crosstalk between the visceral and subcutaneous adipose tissue (AT) and the skeletal muscle under conditions of low-grade local and systemic inflammation, inflammaging.

Interaction of aging with lipoxygenase deficiency initiates hypersplenism, cardiac dysfunction, and profound leukocyte directed non-resolving inflammation.

In the process of physiological cardiac repair, splenic leukocyte-activated lipoxygenases (LOXs) are essential for the biosynthesis of specialized pro-resolving lipid mediators as a segment of an active process of acute inflammation in splenocardiac manner. In contrast, young 12/15LOX-/- mice use a compensatory mechanism that amplifies epoxyeicosatrienoic acid mediators after myocardial infarction, improving cardiac repair, function, and survival. Next, we tested whether deletion of 12/15LOX impacted the genesis of chronic inflammation in progressive aging. To test the risk factor of aging, we
