Low succinate dehydrogenase (SDH) activity in a patient with a hereditary myopathy with paroxysmal myoglobinuria.

An unusual hereditary myopathy with paroxysmal myoglobinuria has been described previously. We have studied muscle biopsy specimens taken before and after exercise to exhaustion (24 min at 20-25 W) in a young woman with this condition. Marked glycogenolysis with lactate production and marked phosphagen breakdown (ATP + CP) were observed after exercise, and almost all type I fibres were found to be depleted of glycogen. Succinate dehydrogenase (SDH) activity was low, while the activities of 3-OH-acyl-CoA-dehydrogenase, phosphofructokinase, phosphorylase and lactate dehydrogenase were normal. On

Methylprednisolone sodium succinate (Solu-Medrol) in the treatment of typhoid perforation (a preliminary report).

It is now universally accepted that treatment of typhoid perforation must be surgical. In spite of this and the use of antibiotics, the mortality rate is still high. Previous observations that some patients with typhoid perforation present with features suggestive of septicaemia and septic shock made us conduct a clinical trial with methylprednisolone sodium succinate (Solu-Medrol) in treating this disease. Our data show that use of this drug in addition to antibiotics and early surgical intervention reduces the mortality rate in typhoid perforation.

Human brain aldehyde reductases: relationship to succinic semialdehyde reductase and aldose reductase.

Human brain contains multiple forms of aldehyde-reducing enzymes. One major form (AR3), as previously shown, has properties that indicate its identity with NADPH-dependent aldehyde reductase isolated from brain and other organs of various species; i.e., low molecular weight, use of NADPH as the preferred cofactor, and sensitivity to inhibition by barbiturates. A second form of aldehyde reductase ("SSA reductase") specifically reduces succinic semialdehyde (SSA) to produce gamma-hydroxybutyrate. This enzyme form has a higher molecular weight than AR3, and uses NADH as well as NADPH as cofactor.

Metabolic and muscular adaptations to dietary succinate in Tan sheep: Insights from transcriptomics.

Succinate (SUC), a dietary feed additive, is known to influence muscle fiber structure and lipid metabolism, thereby enhancing meat quality in livestock. This study evaluated the effects of SUC supplementation on growth performance, meat quality, muscle fiber morphology, and transcriptomic profiles of the liver and longissimus thoracis (LT) muscle in Tan sheep. Thirty 5-month-old male Tan sheep (31.48 ± 0.25 kg) were randomly allocated to three dietary treatments (n = 10 per group): control (CON) group (basal diet), SUC1 group (basal diet + 1.0% SUC), and SUC2 group (basal diet + 2.0% SUC). Succinate receptor 1 restricts hematopoiesis and prevents acute myeloid leukemia progression.

Despite intriguing roles for the Succinate receptor (Sucnr1) in inflammation, few studies have explored its role in hematopoiesis. Here, we show that low SUCNR1 represents a marker for reduced overall and progression-free survival in acute myeloid leukemia (AML) patients. Succinic acid, which displays Sucnr1-dependent and independent effects, promotes disease in mouse models of pre-leukemic myelopoiesis, AML and AML xenografts, expressing low SUCNR1. In vivo global or hematopoietic deletion of Sucnr1 induces expansion of hematopoietic stem and progenitor cells (HSPC) and hematopoiesis, whilst

Gut-derived succinic acid potentiates high-altitude-related spermatogenesis dysfunction.

Exposure to high altitude (HA) is linked to male spermatogenesis impairment and microbial imbalance; however, the association and underlying mechanisms remain unexplored. Herein, we demonstrate that HA-induced gut microbiota changes in humans and mice lead to reduced sperm quality. Specifically, we observe an increase in intestinal Clostridium symbiosum colonization in HA human populations and mice exposed to HA-mimicking conditions. Furthermore, C. symbiosum causes a decline in sperm quality through succinic acid (su) production. Mechanistically, su targets G-protein-coupled receptor 91(GPR91

Succinate drives macrophage inflammation by promoting GPR91 mitochondrial signaling in rheumatoid arthritis.

High succinate concentrations are implicated in rheumatoid arthritis (RA) and other inflammatory diseases through G protein-coupled receptor 91 (GPR91)-mediated signaling. Despite the therapeutic potential of targeting GPR91, conflicting reports on the receptor's inflammatory roles have hindered treatment development. Here, we report that the effects of succinate on GPR91 signaling are biphasic and concentration dependent. At physiological succinate concentration, membrane-localized GPR91 promotes M2 polarization through Gq-mediated activation of phospholipase C and intracellular calcium mobil

Succinate-driven PKM2 succinylation and dimerization accelerates age-associated cardiac fibrosis.

Age-related cardiac fibrosis is a key driver of heart failure and hallmark of aging whose mechanisms remain incompletely understood. Here we show elevated succinate levels in aged mice and humans drive cardiac fibrosis by enhancing fibroblast activation and collagen production. This process is mediated through succinate-dependent succinylation of PKM2 at lysine 125, promoting its transition from tetrameric to dimeric states. Using SUCNR1-/- mice, we establish that succinate signaling through SUCNR1/GPR91 promotes PKM2 succinylation and dimerization, creating a profibrotic network associated wi

Increase in plasma succinate is associated with aerobic lactate production in a model of endotoxic shock.

The Krebs or tricarboxylic acid (TCA) cycle plays a key role in the regulation of immune responses and adaptations to hypoxia that occur during sepsis. Although the concentrations of some of these intermediates have been reported to be increased in large cohorts of septic patients, a detailed analysis of their changes during sepsis is still lacking. Here, we investigated the plasma concentrations of several TCA intermediates in a swine model of endotoxic shock and the relationship between these TCA cycle intermediates and lactate production. Nine female swine were administered lipopolysacchari

Recent Advances in Succinate Dehydrogenase Deficient Gastrointestinal Stromal Tumor Systemic Therapies.

Gastrointestinal stromal tumors (GIST) are the most common gastrointestinal soft tissue sarcomas, with an incidence of about 15 cases per million person-years. Approximately 15% of GIST develop due to succinate dehydrogenase deficiency (SDH-Def), and such tumors do not respond well to the tyrosine kinase inhibitors (TKIs) used to treat other GIST. Due to its indolent nature SDH-Def GIST can often be surveilled if asymptomatic. In our current practice we typically treat advanced symptomatic SDH-Def GIST with the anti-angiogenic TKIs, sequentially treating with sunitinib, regorafenib and pazopan

TIGAR reduces neuronal ferroptosis by inhibiting succinate dehydrogenase activity in cerebral ischemia.

Ischemia Stroke (IS) is an acute neurological condition with high morbidity, disability, and mortality due to a severe reduction in local cerebral blood flow to the brain and blockage of oxygen and glucose supply. Oxidative stress induced by IS predisposes neurons to ferroptosis. TP53-induced glycolysis and apoptosis regulator (TIGAR) inhibits the intracellular glycolytic pathway to increase pentose phosphate pathway (PPP) flux, promotes NADPH production and thus generates reduced glutathione (GSH) to scavenge reactive oxygen species (ROS), and thus shows strong antioxidant effects to ameliora

Succinate-mediated symbiosis between Dialister hominis and an uncharacterized Segatella-like pectinophile.

OBJECTIVES: Syntrophy has been documented between pectinophiles and methanol-utilizing bacteria, along with instances of cross-feeding between pectinophiles and methanogens. However, studies on the ecology of pectinophiles in anaerobic digestion (AD) are lacking. Therefore, in this study, we aimed to elucidate the ecology of pectinophiles by isolating novel pectinophile forms and conducting a comprehensive analysis of their physiology and ecology. METHODS: Complex microbial communities from AD systems were enriched in a pectin-containing medium; subsequently, specific strains were isolated us

Pheochromocytoma associated with a succinate dehydrogenase subunit B mutation: A minireview and a case report.

Objective. Pheochromocytomas and paragangliomas are rare neuroendocrine tumors that arise from the chromaffin cells of the adrenal medulla or extra-adrenal tissues. These tumors are characterized by an excessive secretion of catecholamines, which are responsible for the clinical manifestation of the disease. Although most of these tumors are sporadic, underlying genetic abnormalities may be present in up to 24% of the cases. A succinate dehydrogenase subunit B (SDHB) mutation represents one of the rare presentations of the disease. In this study, we represent a rare case of pheochromocytoma as

Understanding Ischemic Retinopathies: The Role of Succinate and Its Receptor in Retinal Pigment Epithelium.

Retinopathy is the general name for all condition of the eyes in which blood vessels that supply oxygen to the retina are damaged. These include diabetic retinopathy, retinopathy of prematurity, hypertensive retinopathy, and arteriosclerotic retinopathy. Although the initial trigger that leads to insufficient perfusion of the retina may be different, once a critical level of ischemia is achieved, all types of retinopathies seem to follow a common sequence-oxidative stress, followed by hypoxia-induced formation of morphologically abnormal vessels. This preretinal vascular growth is the most sev

Microbial metabolite succinate activates solitary chemosensory cells in the human sinonasal epithelium.

BACKGROUND: Succinate, although most famous for its role in the Krebs cycle, can be released extracellularly as a signal of cellular distress, particularly in situations of metabolic stress and inflammation. Solitary chemosensory cells (SCCs) express SUCNR1, the succinate receptor, and modulate type 2 inflammatory responses in helminth and protozoal infections in the small intestine. SCCs are the dominant epithelial source of interleukin-25, as well as an important source of cysteinyl leukotrienes in the airway, and have been implicated as upstream agents in type 2 inflammation in chronic rhin

SNORA14A inhibits hepatoblastoma cell proliferation by regulating SDHB-mediated succinate metabolism.

Hepatoblastoma (HB) is the most common paediatric liver malignancy. Dysregulation of small nucleolar RNAs (snoRNAs) is a critical inducer of tumour initiation and progression. However, the association between snoRNAs and HB remains unknown. Here, we conducted snoRNA expression profiling in HB by snoRNA sequencing and identified a decreased level of SNORA14A, a box H/ACA snoRNA, in HB tissues. Low expression of SNORA14A was correlated with PRETEXT stage and metastasis in patients. Functionally, overexpression of SNORA14A suppressed HB cell proliferation and triggered cell apoptosis and G2/M pha
