Multiple viruses induce reactive oxygen intermediate (ROI) generation during infection that plays an important role in growth. We have examined the importance of ROI during lymphocytic choriomeningitis virus (LCMV) infection of immortalized BHK-21 cells and murine peritoneal macrophages. Two new iron nitrosyls derived from two designed pentadentate ligands N,N-bis(2-pyridylmethyl)-amine-N'-(2-pyridylmethyl)acetamide and N,N-bis(2-pyridylmethyl)-amine-N'-[1-(2-pyridinyl)ethyl]acetamide (PcPy(3)H and MePcPy(3)H, respectively, where H is the dissociable amide proton) have been structurally characterized. The superoxide dismutase-mimetic activity and the electrochemical behavior of the binuclear complex [Cu(TSA)2py]2 is reported. The complex exhibits a marked SOD activity in the nitroblue tetrazolium assay. Its redox response is in agrement with two copper atoms partially coupled through the carboxylate moieties. The electrode reactions lie in the range of the superoxide/peroxide and oxygen/superoxide couples. This fact is indicative that this complex can act as catalyst for the SOD reaction.

A nonspecific staining occurs in serum in isoenzyme methods based on tetrazolium staining reactions. The artefact is pronounced when a large application volume, a prolonged incubation time, distinctly alkaline conditions, or excess of phenazine methosulphate is used. Only phenazine methosulphate and a tetrazolium stain are required for the appearance. Inhibition studies and histochemical staining reactions give evidence that protein-bound sulphydryl groups are responsible for the artefact in serum.

Oxidative stress contributes significantly to graft failure, morbidity and mortality in renal transplant recipients (RTR).
