TORC1 selectively regulates synaptic maturation and input convergence in the developing visual system.

Newly synthesized proteins support the development of functional neural circuits and previous work has suggested that dysregulated translation mediates certain forms of autism spectrum disorder (ASD). Here, we investigated the role of Target of Rapamycin Complex 1 (TORC1) in synaptic and dendritic development in vivo in the retinotectal system of Xenopus laevis tadpoles. We found that TORC1 signaling regulates dendritic growth and branching and that acute over-activation of TORC1 by Rheb overexpression drove enhanced maturation of excitatory synapses by recruiting AMPA receptors. Interestingly

Normal correspondence of tectal maps for saccadic eye movements in strabismus.

The superior colliculus is a major brain stem structure for the production of saccadic eye movements. Electrical stimulation at any given point in the motor map generates saccades of defined amplitude and direction. It is unknown how this saccade map is affected by strabismus. Three macaques were raised with exotropia, an outwards ocular deviation, by detaching the medial rectus tendon in each eye at age 1 mo. The animals were able to make saccades to targets with either eye and appeared to alternate fixation freely. To probe the organization of the superior colliculus, microstimulation was ap

Dissociation between the panicolytic effect of cannabidiol microinjected into the substantia nigra, pars reticulata, and fear-induced antinociception elicited by bicuculline administration in deep layers of the superior colliculus: The role of CB1-cannabinoid receptor in the ventral mesencephalon.

Many studies suggest that the substantia nigra, pars reticulata (SNpr), a tegmental mesencephalic structure rich in γ-aminobutyric acid (GABA)- and cannabinoid receptor-containing neurons, is involved in the complex control of defensive responses through the neostriatum-nigral disinhibitory and nigro-tectal inhibitory GABAergic pathways during imminently dangerous situations. The aim of the present work was to investigate the role played by CB1-cannabinoid receptor of GABAergic pathways terminal boutons in the SNpr or of SNpr-endocannabinoid receptor-containing interneurons on the effect of in

Development of tectal connectivity across metamorphosis in the bullfrog (Rana catesbeiana).

In the bullfrog (Rana catesbeiana), the process of metamorphosis culminates in the appearance of new visual and visuomotor behaviors reflective of the emergence of binocular vision and visually-guided prey capture behaviors as the animal transitions to life on land. Using several different neuroanatomical tracers, we examined the substrates that may underlie these behavioral changes by tracing the afferent and efferent connectivity of the midbrain optic tectum across metamorphic development. Intratectal, tectotoral, tectotegmental, tectobulbar, and tecto-thalamic tracts exhibit similar traject

Saccadic trajectory in Huntington's disease.

Trajectories of saccadic eye movements can be modulated by the presence of a competing visual distractor. It is proposed that the superior colliculus (SC) controls the initial deviation through competitive lateral interactions. Given the ramifications of connections between basal ganglia (BG) thalamo-cortical circuitry and the SC, it was anticipated that this modulation would be differentially effected in those with Huntington's disease, which in its early stages is primarily a disorder of the BG. Horizontal deviation was determined for exogenously driven and endogenously driven vertical sacca

Pattern of striate cortical projections to the pretectal complex in the guinea pig.

The primary goal of this study was to determine whether the striate cortex (Oc 1) of the guinea pig projects to the pretectal nucleus of the optic tract (NOT), the first postretinal station of the horizontal optokinetic pathway, and, if so, to analyze the anatomical organization of this cortico-NOT projection. Other goals of this investigation are to identify other pretectal nuclear projections from the visual cortex in the guinea pig, and to determine whether there is any visuotopic organization in this pathway. Axonal tracers (biocytin or 3H-leucine) were injected into the striate cortex (Oc

Activity-driven sharpening of the retinotectal projection in goldfish: development under stroboscopic illumination prevents sharpening.

Blocking or synchronizing activity during regeneration of the retinotectal projection prevents both the sharpening of the retinotopic map recorded on tectum and the refinement of the structure of individual arbors within the plane of the map, and this refinement is triggered by N-methyl-D-aspartate (NMDA) receptors. We tested whether activity-driven refinement also occurs during development of the projection in larval and young adult goldfish. Shortly after hatching, larval goldfish were placed into tanks within light-tight chambers illuminated by a xenon strobe at 1 Hz for 14 h of each daily

Albino gene dosage and retinal decussation patterns in the pigmented ferret.

We have examined the retinal decussation patterns in pigmented ferrets that were either wild-type sable or heterozygous with one albino gene. Unilateral injections of horseradish peroxidase were made into the optic tract and labeled ganglion cells visualized in retinal wholemounts. In both wild-type and heterozygous ferrets, those ganglion cells in the temporal retina with the largest cell bodies projected only to the contralateral side of the brain. The total number of ipsilaterally projecting ganglion cells did not differ with the genotype of the animal. The numbers ranged from 5471-6759 cel

Complementary and non-matching afferent compartments in the cat's superior colliculus: innervation of the acetylcholinesterase-poor domain of the intermediate gray layer.

Three tectal afferent-fiber systems were experimentally labeled in the cat to learn how their distributions within the superior colliculus were related to the prominent compartments of high acetylcholinesterase activity found in the intermediate gray layer. Presumptive somatic sensory afferents were labeled by injections of horseradish peroxidase-wheatgerm agglutinin conjugate placed at the bulbospinal junction and in the ventral anterior ectosylvian cortex corresponding to somatic sensory area SIV. Vision-related afferents were labeled by injections of the same tracer substance into the later

Optic axons ignore foreign denervated sites in goldfish tectum even after removal of some of their normal termination sites.

Previous experiments have shown that optic axons ignore foreign denervated territory in the goldfish tectum and preferentially reinnervate their normal layers of termination. In the present experiments ultrastructural morphometry was used to test whether this was still the case if optic axons were deprived of some of their normal termination sites. The stratum fibrosum marginale (SFM) of the tectum was partially denervated to provide foreign synaptic sites and the caudal half of the tectum was ablated at the same time. This removed approximately half the normal sites of optic termination. In t

A quantitative study of the reinnervation of the goldfish optic tectum following optic nerve crush.

Stereological and quantitative morphometric methods were used to study changes in the stratum fibrosum et griseum superficialis (SFGS), the major retinal target, in optic tectum of goldfish, during regeneration of the optic nerve. Orthograde transport of HRP by optic axons was used to characterize the retinal projection in SFGS. Profiles of HRP-labeled optic terminals contained rounded vesicles, contacted small dendrites, and were distributed uniformly throughout the area of SFGS sampled; labeling density estimates indicate that at least 37% of the total terminal population in SFGS is retinal

Target regulation of synaptic number in the compressed retinotectal projection of goldfish.

In order to determine the morphological consequences of the formation of a compressed retinotectal projection, the optic neuropil lamina (stratum fibrosum et griseum superficialis, SFGS) was examined in large goldfish 3 months to 4 years after ablation of the caudal half of the tectum both with crush of the optic nerve (HTX) without (HT). In semithin sections, the SFGS, as delineated with orthograde HRP labeling, shows a persistent hypertrophy of about 25% in HTX and HT groups. Comparison of ultrastructural stereological data with similar data on control and regenerated projections to intact t

Different loci suggested to mediate tilt and spiral motion aftereffects.

Interocular transfer of two figural aftereffects was examined in orthotropes and strabismic subjects. Within both groups there were persons with an appreciable degree of stereoacuity and others who had little or none. Experimental evidence from a variety of sources has suggested that stereopsis depends upon binocularity of units in the geniculostriate system. For the tilt aftereffect, interocular transfer correlated with stereoacuity among both orthotropes and strabismics. For the spiral motion aftereffect, interocular transfer did not correlate with stereoacuity; it was present among orthotro

Topography of visual and somatosensory projections to mouse superior colliculus.

In adult mice of the C57BL/6J strain the projection of the visual field was systematically mapped under direct vision. As in other vertebrate species the nasal (anterior) field projected anterolaterally, and the inferior field posterolaterally. Values of magnification-1 (m-1, or degrees of visual field per millimeter tectal surface) were calculated over most of the tectum, for measurements in the coronal and sagittal planes. Whereas m-1 was fairly constant for measurement pairs in sagittal planes, for coronal planes there was a rather large, elongated, horizontally oriented area in the upper f

Simultaneous neuron evidence for much higher covariation with saccadic reaction time of superior colliculus than primary visual cortex visual responses.

Visual response strength in the primate superior colliculus (SC) has recently been shown to inversely correlate with trial-by-trial saccadic reaction time in a much stronger way than visual response strength in the primary visual cortex (V1). However, for any given visual stimulus onset, populations of neurons in each brain area are concurrently activated, leaving open the question of how V1 visual response strength can predict trial-by-trial saccadic reaction time when multiple simultaneously recorded neurons are taken into account. Using a classic visually guided saccade task, here we assess

Retinal Input to Macaque Superior Colliculus Derives from Branching Axons Projecting to the Lateral Geniculate Nucleus.

The superior colliculus receives a direct projection from retinal ganglion cells. In primates, it remains unknown if the same ganglion cells also supply the lateral geniculate nucleus. To address this issue, a double-label experiment was performed in two male macaques. The animals fixated a target while injection sites were scouted in the superior colliculus by recording and stimulating with a tetrode. Once suitable sites were identified, cholera toxin subunit B-Alexa Fluor 488 was injected via an adjacent micropipette. In a subsequent acute experiment, cholera toxin subunit B-Alexa Fluor 555

M2 Cortex Circuitry and Sensory-Induced Behavioral Alterations in Huntington's Disease: Role of Superior Colliculus.

Early and progressive cortico-striatal circuit alterations have been widely characterized in Huntington's disease (HD) patients. Cortical premotor area, M2 cortex in rodents, is the most affected cortical input to the striatum from early stages in patients and is associated to the motor learning deficits present in HD mice. Yet, M2 cortex sends additional long-range axon collaterals to diverse output brain regions beyond basal ganglia. Here, we aimed to elucidate the contribution of M2 cortex projections to HD pathophysiology in mice. Using fMRI, M2 cortex showed most prominent functional conn

Optogenetic Stimulation of the Superior Colliculus Confers Retinal Neuroprotection in a Mouse Glaucoma Model.

Glaucoma is characterized by a progressive loss of retinal ganglion cells (RGCs) in the eye, which ultimately results in visual impairment or even blindness. Because current therapies often fail to halt disease progression, there is an unmet need for novel neuroprotective therapies to support RGC survival. Various research lines suggest that visual target centers in the brain support RGC functioning and survival. Here, we explored whether increasing neuronal activity in one of these projection areas could improve survival of RGCs in a mouse glaucoma model. Prolonged activation of an important

Comparative Brain Morphology of the Greenland and Pacific Sleeper Sharks and its Functional Implications.

In cartilaginous fishes, variability in the size of the brain and its major regions is often associated with primary habitat and/or specific behavior patterns, which may allow for predictions on the relative importance of different sensory modalities. The Greenland (Somniosus microcephalus) and Pacific sleeper (S. pacificus) sharks are the only non-lamnid shark species found in the Arctic and are among the longest living vertebrates ever described. Despite a presumed visual impairment caused by the regular presence of parasitic ocular lesions, coupled with the fact that locomotory muscle power

Asphyxia induced by umbilical cord occlusion alters glutamatergic and GABAergic synaptic transmission in neurons of the superior colliculus in fetal rats.

Using optical recordings, we studied the effects of asphyxia on intracellular Cl(-) and Ca(2+) concentrations ([Cl(-)]i; [Ca(2+)]i) in the superior colliculus of fetal rats, which were connected via the umbilical cord to the dam. Acute asphyxia was induced by umbilical cord occlusion. The number of fetal superior colliculus neurons showing GABA-mediated increases in [Cl(-)]i (leading to hyperpolarization) following local synaptic electrical stimulation had decreased by 3 h post-asphyxiation, while the number showing GABA-mediated decreases in [Cl(-)]i (leading to depolarization) increased.
