Nitric oxide-induced potentiation of CA1 hippocampal synaptic transmission during baseline stimulation is strictly frequency-dependent.

Nitric oxide (NO) has been hypothesised to serve a signalling role in certain types of synaptic plasticity. If so, exogenously applied NO should be able to elicit those same plastic changes under appropriate conditions. In the case of hippocampal long-term potentiation (LTP), however, existing evidence is discrepant. Field recordings of synaptic transmission in the CA1 area of rat hippocampal slices were used to re-examine this issue. Under 0.2 Hz afferent fibre stimulation, NO (delivered using two different NONOates) produced, concentration-dependently, a depression of synaptic transmission. …

Cannabinoid receptor mediated inhibition of excitatory synaptic transmission in the rat hippocampal slice is developmentally regulated.

The cannabinoid (CB) receptor agonist WIN55,212-2 (500 nM) had no effect on the first of a pair of population spikes evoked in the CA1 regio … agonist AM281 (500 nM). The results indicate that activation of CB(1) receptors inhibits excitatory synaptic transmission in neonatal, but not adult rat hippocampus. This developmental regulation of CB(1) receptor mediated control of excitatory transmission may help explain some, but not all, of the previous discrepancies in the literature.

The role of adenosine A2a receptors in regulating GABAergic synaptic transmission in striatal medium spiny neurons.

We demonstrated an adenosine A2a receptor-mediated disinhibition of medium spiny projection neurons using intracellular recording and the whole-cell patch-clamp recording applied to these cells, visually identified in thin rat striatal slices. The A2a receptor agonist 2-[p-(2-carboxyethyl) phenylethylamino]-5'-N- ethylcarboxamido adenosine (CGS-21680; 0.3-10 microM) suppressed GABAergic synaptic transmission onto these cells in a manner inhibited by the A2a receptor-selective antagonist (E)-8-(3,4-dimethoxystyryl)-1,3-dipropyl-7-methylxanthine (0.1-1.0 microM). The A1 receptor antagonists had …

Nickel induces oscillatory behavior and enhanced synaptic and electrotonic transmission between stomatogastric neurons of Panulirus interruptus.

The pyloric pattern generator network of the stomatogastric ganglion uses a mixture of burst-inducing plateau potentials, synaptic transmission, and electrical coupling to produce its patterned output. This study examines the effects of two divalent, calcium channel blockers, nickel and cadmium, on voltage oscillations, synaptic transmission, and electrical coupling between the two pyloric dilator (PD) neurons and lateral pyloric (LP) neuron of Panulirus interruptus. The in vitro stomatogastric ganglion was bathed in saline containing tetrodotoxin (TTX) to eliminate Na-spikes and the spontaneo…

Tyramine modulates synaptic transmission in vertebrate neuromuscular junction by releasing catecholamines and activating alpha-adrenergic receptors.

Tyramine is a biogenic amine whose role is closely linked to the sympathetic nervous system. Sympathetic axons densely innervate the neuromuscular junction regions of skeletal muscles. We hypothesized that tyramine might be the involvement in modulation of acetylcholine (ACh) release in the neuromuscular junctions. On the mouse Levator Auris Longus muscle it was found that tyramine enhances the loss of fluorescent false monoamine neurotransmitter FNN 511, preloaded into sympathetic varicosities located near the neuromuscular junctions. This indicates tyramine-induced release of endogenous cate

A biophysically grounded model of glutamatergic synaptic transmission integrating glutamate transport, receptor kinetics, and electrotonic effects.

Gluatamatergic synaptic transmission, critical for learning and memory, relies on precise regulation of extracellular glutamate levels by astrocytic transporters, particularly EAAT2. While existing models of AMPA and NMDA receptor kinetics often oversimplify glutamate dynamics or become computationally intractable, this study develops a balanced, biophysically grounded model that integrates glutamate transport, receptor sensitivity, and electrotonic effects. Using rat hippocampal slices, we recorded postsynaptic currents in CA1 pyramidal neurons under control conditions and during glutamate tr…

Multiplexed changes in synaptic transmission underlie stress-induced reduction of persistent firing in the parietal cortex.

Repeated exposure to stress disrupts cognitive processes, including attention and working memory. A key mechanism supporting these functions … -induced reduction in persistent firing. Rather, a combination of altered excitatory and inhibitory synaptic transmission was necessary to impact sustained activity. These data suggest that a multitude of converging changes in neural and circuit function underpin the effects of stress on cognitive processes.

The elevated open platform stress suppresses excitatory synaptic transmission in the layer V anterior cingulate cortex.

There are various forms of stress including; physical, psychological and social stress. Exposure to physical stress can lead to physical sen … m the ACC. However, it is still unclear whether or not EOP exposure alters intrinsic properties and synaptic transmission in layer V pyramidal neurons. This is essential because these neurons are known to be a primary output to subcortical structures which may ultimately impact the behavioral stress response. Here, we studied both intrinsic properties and excitatory/in …

Hypoxia-induced depression of synaptic transmission becomes irreversible by intracellular accumulation of non-excitatory amino acids.

The intracellular accumulation of some amino acids (AAs), mainly glutamine, can contribute to brain edema observed during liver failure. We recently demonstrated that individual applications of high concentrations (10 mM) of some non-excitatory AAs increase the electrical resistance of hippocampal slices, indicating cell swelling. Therefore, we pondered whether an AA mixture's application might cause cell swelling at a physiological concentration range. In rat hippocampal slices, we carried out extra- and intracellular electrophysiological recordings and AAs analysis to address this question.

Danish and British dementia ITM2b/BRI2 mutations reduce BRI2 protein stability and impair glutamatergic synaptic transmission.

Mutations in integral membrane protein 2B (ITM2b/BRI2) gene cause familial British and Danish dementia (FBD and FDD), autosomal dominant disorders characterized by progressive cognitive deterioration. Two pathogenic mechanisms, which may not be mutually exclusive, have been proposed for FDD and FBD: 1) loss of BRI2 function; 2) accumulation of amyloidogenic mutant BRI2-derived peptides, but the mechanistic details remain unclear. We have previously reported a physiological role of BRI2 in excitatory synaptic transmission at both presynaptic termini and postsynaptic termini. To test whether pat…

Plastic changes in spinal synaptic transmission following botulinum toxin A in patients with post-stroke spasticity.

OBJECTIVE: The therapeutic effects of intramuscular injections of botulinum toxin-type A on spasticity can largely be explained by its blocking action at the neuromuscular junction. Botulinum toxin-type A is also thought to have a central action on the functional organization of the central nervous system. This study assessed the action of botulinum toxin-type A on spinal motor networks by investigating post-activation depression of the soleus H-reflex in post-stroke patients. Post-activation depression, a presynaptic mechanism controlling the synaptic efficacy of Ia-motoneuron transmission, i

A mouse model for MeCP2 duplication syndrome: MeCP2 overexpression impairs learning and memory and synaptic transmission.

Rett syndrome and MECP2 duplication syndrome are neurodevelopmental disorders that arise from loss-of-function and gain-of-function alterations in methyl-CpG binding protein 2 (MeCP2) expression, respectively. Although there have been studies examining MeCP2 loss of function in animal models, there is limited information on MeCP2 overexpression in animal models. Here, we characterize a mouse line with MeCP2 overexpression restricted to neurons (Tau-Mecp2). This MeCP2 overexpression line shows motor coordination deficits, heightened anxiety, and impairments in learning and memory that are accom

Diabetes depresses synaptic transmission in sympathetic ganglia by inactivating nAChRs through a conserved intracellular cysteine residue.

Most people with diabetes develop severe complications of the autonomic nervous system; yet, the underlying causes of many diabetic-induced dysautonomias are poorly understood. Here we explore the idea that these dysautonomias results, in part, from a defect in synaptic transmission. To test this idea, we investigated cultured sympathetic neurons and show that hyperglycemia inactivates nAChRs through a mechanism involving an elevation in reactive oxygen species and an interaction with highly conserved cysteine residues located near the intracellular mouth of the nAChR channel. Consistent with …

Distinct physiological mechanisms underlie altered glycinergic synaptic transmission in the murine mutants spastic, spasmodic, and oscillator.

Spastic (spa), spasmodic (spd), and oscillator (ot) mice have naturally occurring glycine receptor (GlyR) mutations, which manifest as motor deficits and an exaggerated "startle response." Using whole-cell recording in hypoglossal motoneurons, we compared the physiological mechanisms by which each mutation alters GlyR function. Mean glycinergic miniature IPSC (mIPSC) amplitude and frequency were dramatically reduced (>50%) compared with controls for each mutant. mIPSC decay times were unchanged in spa/spa (4.5 +/- 0.3 vs 4.7 +/- 0.2 ms), reduced in spd/spd (2.7 +/- 0.2 vs 4.7 +/- 0.2 ms), and

Brain-derived neurotrophic factor acutely depresses excitatory synaptic transmission to GABAergic neurons in visual cortical slices.

Brain-derived neurotrophic factor (BDNF) acutely modulates synaptic transmission to excitatory neurons in hippocampus and neocortex. The question of whether BDNF acts similarly on excitatory synaptic transmission to GABAergic neurons was eluded in previous studies using cortical slices. To address this question, we used transgenic mice in which expression of green fluorescence protein (GFP) is regulated by glutamic acid decarboxylase 67 (GAD67) promoter. In cortical slices prepared from these GAD67-GFP knock-in mice, we could detect GABAergic neurons under a fluorescent microscope. An applicat…

Acute intrahippocampal infusion of BDNF induces lasting potentiation of synaptic transmission in the rat dentate gyrus.

The effect of acute intrahippocampal infusion of brain-derived neurotrophic factor (BDNF) on synaptic transmission in the dentate gyrus was investigated in urethan-anesthetized rats. Medial perforant path-evoked field potentials were recorded in the dentate hilus and BDNF-containing buffer was infused (4 microl, 25 min) immediately above the dentate molecular layer. BDNF led to a slowly developing increase of the field excitatory postsynaptic potential (fEPSP) slope and population spike amplitude. The potentiation either reached a plateau level at approximately 2 h after BDNF infusion or conti…
