Expression of Six1 and Six4 in mouse taste buds.

Members of the Six gene family are expressed in various tissues including sensory organs, such as the inner ear and olfactory epithelium. We examined the expression of Six1 and Six4 mRNAs in mouse taste buds by using in situ hybridization. Six1 was detected immunohistochemically in the nuclei of taste bud cells, in a subset of type-II cells, as shown by double-immunolabeling with anti-Six1 together with anti-PLCβ2 or anti-IP(3)R3 antibodies. Six1-immunoreactive (IR) nuclei appeared at embryonic day 17.5 in the dorsal epithelium, and in the trench wall epithelium of circumvallate papillae at po…

The effect of taste and palatability on lingual swallowing pressure.

There is evidence that a strong, unpalatable, sour bolus improves swallowing in neurogenic dysphagia. It is not known whether other tastes may alter swallowing physiology. This study investigated the effect of moderate versus high taste concentrations (sweet, sour, salty, bitter) and barium taste samples on lingual swallowing pressure in ten healthy young adults, using a three-bulb lingual pressure array secured to the hard palate. Palatability of the samples was analyzed using the nine-point hedonic scale. Results showed that moderate sucrose, high salt, and high citric acid elicited signific…

[Assessment of the correlation between taste and smell functioning].

UNLABELLED: There are few studies examining relationship between taste and smell. However their clinical unity is more often indicated. Smell perception abnormalities cause modification in taste of food intensity, disabling perception of taste as a consequence of taste's sniffing failure. Thus disturbances of these senses, which often commonly coexist could cause great discomfort in patients, however taste sense is estimated as less important comparing to smell sense. The aim of that study was to compare the taste perception in patients with normal smell function and patients with hyposmia and…

Assessment of smell and taste in patients with allergic rhinitis.

We examined the senses of smell and taste in 240 patients with verified hypersensitivity reactions of the respiratory tract. Olfactometry was performed according to Elsberg and Levy's method and followed by electrogustometry. The examinations revealed that the incidence of smell and taste disorders in patients with allergic rhinitis is 21.4% and 31.2%, respectively. No significant relationships between smell and taste dysfunction were found. We evaluated statistically positive correlations between the olfactory and gustatory thresholds compared with 78 qualitative and quantitative factors asse…

Keratins as markers of differentiated taste cells of the rat.

Cytokeratins in taste buds were immunocytochemically evaluated with monoclonal antibodies. In each of six different epithelial sites in the rat oral cavity, intragemmal cells of taste buds were immunoreactive for keratin polypeptides 8, 18, and 19, as well as for keratin 7, which has not been previously reported in taste buds. Keratin-18-like immunoreactivity was present in fewer than half of the intragemmal cells, whereas all intragemmal cells were immunopositive for keratins 7, 8, and 19. Apart from some salivary duct cells, no other cells in the tongue were immunoreactive for any of these f…

The effect of unilateral chorda tympani damage on taste.

Patients with localized damage to the taste system often experience no subjective change in real-world taste experience. In an effort to understand this, eight patients who recently underwent acoustic neuroma removal were evaluated for taste loss. Localized taste testing showed that taste intensities decreased in the distribution of cranial nerve VII ipsilateral to tumor removal as expected, but asymmetries occurred for IX. Intensities were greater on the side contralateral to the tumor removal. In addition, palatal taste, also thought to be mediated by VII, was not totally abolished. It is co…

The sense of taste: neurobiology, aging, and medication effects.

The sense of taste is an oral chemical sense in mammals that is involved in the choice of foods. Initial transduction of taste stimuli occurs in taste buds, which are distributed in four discrete fields in the oral cavity. Medications can affect the taste buds and ion channels in taste-bud cell membranes involved in stimulus transduction. The sense of taste gradually declines with aging, with bitter taste most affected. Neural circuits that mediate taste in primates include cranial nerves VII, IX, and X, the solitary nucleus in the brain stem, the ventroposteromedial nucleus of the thalamus, a…

Receptor cell regeneration and connectivity in olfaction and taste.

The capacity of adult mammalian gustatory and olfactory receptor cells to regenerate and make synaptic reconnections provides examples that may be useful in initiating replacement of other kinds of sensory receptor cells. The sensory code for taste quality may not be degraded by taste receptor cell turnover because axons probably recouple to the appropriate type of new receptor cell by axon-receptor cell affinity. Experiments on the development and regeneration of taste receptor cells suggest that they regenerate and turn over by recapitulating the late but not the early steps in taste bud dev…

Taste bud distribution in the rat pharynx and larynx.

The present study evaluated the number and distribution of taste buds in the rat pharynx and larynx using light microscopic techniques. The average number of taste buds in this region was 141.9 (S.D. = 29.4, n = 10). Pharyngeal and laryngeal taste buds could be grouped into three subpopulations: those associated with laryngeal structures (LA), the nasopharynx (NP), or the palatopharyngeal eminence (PE). Laryngeal taste buds were most numerous (mean = 103.2, S.D. = 23.6). They were observed on the laryngeal surface of the epiglottis and extended caudally along the aryepiglottic folds, reaching …

Purification, sequence identification, and taste-presenting mechanism analysis of four novel taste-active peptides enzymatically prepared from shiitake mushroom (Lentinula edodes).

Shiitake mushroom-derived taste-active peptides were prepared by enzymatic hydrolysis and purified step-by-step using D001 macroporous adsorption resin, Sephadex G-100 gel filtration, and reversed-phase high-performance liquid chromatography. The initial hydrolysate contained 16.12 mg/mL peptides with saltiness and umami scores of 6. The active fraction was screened by electronic tongue and sensory evaluation, and four novel peptides (PDDER, QPEEQDER, SPDDER, and WEPGPQ) were identified by liquid chromatography tandem mass spectrometry-based de novo sequencing, all exhibiting strong binding to…

Evaluation of taste score and fungiform papillae quantification using digital image analysis in COVID-19 patients with smell and taste dysfunction.

BACKGROUND: The COVID-19 pandemic which is caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has created a major global health crisis in recent years. Despite this, there have been few studies that have utilized reliable methods to assess changes in taste and smell perception. Therefore, our study aims at the number of fungiform papillae and objective measures of taste perception relationship among COVID-19 patients with olfactory and gustatory disorders. MATERIALS AND METHODS: This was a cross-sectional analytical study in which 57 COVID-19 patients were recruited wh…

Smell and taste of milk during tube feeding of preterm infants: neurodevelopmental follow-up of the randomized TASTE trial, study protocol.

BACKGROUND: The Taste And Smell To Enhance nutrition (TASTE) trial investigated the effects of smell and taste of milk with tube feeding compared to routine care on the growth of preterm infants. There was no difference between groups in growth (weight, head circumference, length) z-scores at discharge from the hospital. Infants in the intervention group had higher head circumference and length z-scores at 36 weeks postmenstrual age, both secondary outcomes. The objective of this follow-up study is to assess 2-year neurodevelopmental and growth outcomes after exposure of preterm infants to the…

Taste assessment for paediatric drug Development: A comparison of bitterness taste aversion in children versus Naïve and expert young adult assessors.

Medicines for children often taste bitter, presenting a significant challenge to treatment compliance. However, most studies on paediatric drug development rely on adult volunteers for sensory research, and the level of expertise required from these assessors is unclear. This study aimed to address this gap by investigating perceived bitterness aversion to taste strips impregnated with different concentrations of quinine hydrochloride in 439 school-aged children. Expert (n = 26) and naïve (n = 65) young adult assessors evaluated quinine solutions as well as taste strips, for methodological bri…
