Competence for transformation is widespread among bacterial species. In the case of Gram-negative systems, a key step to transformation is the import of DNA across the outer membrane. Although multiple factors are known to affect DNA transport, little is known about the dynamics of DNA import. Here, we characterized the spatio-temporal dynamics of DNA import into the periplasm of Neisseria gonorrhoeae. DNA was imported into the periplasm at random locations around the cell contour. Subsequently, it was recruited at the septum of diplococci at a time scale that increased with DNA length. We fou

In vitro studies in model cell lines have implicated the GTPase Rho in the control of diverse cellular responses including the control of the actin cytoskeleton and the regulation of cell cycle progression. It is also reported that the transformation of fibroblasts via oncogenic Ras requires intact Rho signalling. An invaluable tool used to investigate Rho function is the bacterial toxin C3 transferase derived from Clostridium botulinum. C3 transferase ribosylates Rho in its effector domain thereby abolishing interaction with downstream effectors. We have previously reported the use of C3 tran

In order to search for an occult cytotoxic enzymatic activity of the toxic shock syndrome toxin 1 (TSST-1), we placed the gene encoding TSST-1 (tstH) under the control of an inducible promoter in the eukaryotic yeast Saccharomyces cerevisiae. Under similar circumstances, the known bacterial enzymatic cytotoxins Shiga-like toxin and diphtheria toxin are both highly lethal to the yeast host. Although full-length stable TSST-1 was demonstrated within the yeast cells and although it retained mitogenicity for human T cells, it had no apparent effect on the yeast cells' growth kinetics or on their g

The lymphocyte transformation test (LTT) was given to 20 healthy subjects and 43 patients with generalized psoriasis vulgaris: it was given right after stimulation with PHA (spontaneous) and after stimulation with allogenic and autogenic protein factor (NPF). NPF was isolated from secondary lysosome granules of peripheral blood neutrophils. The results were analyzed using computer statistic tests. No distinct differences were noticed between the spontaneous transformation test in psoriatic patients compared to the controls. After stimulation with PHA, the percentage of blast cells was signific

Urban mangrove sediments receive continuous inputs of industrial and domestic pollutants, yet the microbial basis of pollutant transformation in these chronically impacted systems remains insufficiently resolved. This study investigated xenobiotic degradation potential in sediments from two urban mangrove locations along the Mandovi estuary, Goa, India, through reanalysis of previously generated shotgun metagenomic data. The assembled metagenomes showed enrichment of degradation pathways associated with aromatic and aliphatic pollutants commonly linked to urban contamination. A high representa

Twenty dentulous subjects undergoing clinical therapy for severe periodontitis were used to determine the longitudinal effects of bacterial plaque reduction in vitro lymphocyte transformation. The therapy consisted of either complete extractions or partial extractions and periodontal surgery combined with rigorous oral hygiene. Prior to therapy lymphocytes from these subjects responded significantly to Streptolysin O (SLO) but were not transformed significantly by solubilized dental plaque. However, after therapy lymphocytes from these same subjects responded significantly to both solubilized

Efficient nitrogen removal from low-C/N aquaculture wastewater remains challenging because carbon availability can constrain nitrogen conversion. A biochar-assisted algal-bacterial system was developed using a consortium capable of direct ammonium oxidation (Dirammox) and heterotrophic nitrification-aerobic denitrification. Four sequencing batch reactors were operated to compare the individual and combined effects of bioaugmentation and biochar. The combined system achieved a maximum total nitrogen (TN) removal efficiency of 88 % and a mean stable effluent TN concentration of 2.8 mg·L-1, compa

Recent work suggests that soil-borne viruses play an important role in controlling carbon (C) cycling and stocks. However, the contribution of individual prophage (i.e., temperate phages residing within bacterial hosts during lysogenic cycle) to C degradation remains largely undocumented at global scale. Here, we generated a global gene catalog of prophage-encoded carbohydrate-active enzyme (pCAZymes), including 20,131 soil bacterial genomes, 3548 metagenomes, and 951 metatranscriptomes derived from pre-existing databases. The catalog includes 4708 pCAZymes associated with the degradation of l

Human beings release thousands of antibiotics into the environment, which could generate the related transformation products (TPs), most of which have yet to be identified and lack rigorous microbial risk information. This study aimed to investigate the impact and mechanisms of 4-nitro sulfamethoxazole, N4-acetylated sulfamethoxazole, and N4-acetylated sulfadiazine, three typical sulfonamide (SAs) metabolites, on the risk of antibiotic resistance genes (ARGs) transmission. The results revealed that TPs significantly enhance the risk of conjugative transfer of RP4 plasmid at clinically and envi

This study developed dual-function tannin-based flocculants, namely tannin-graft-acrylamide-diallyl dimethyl ammonium chloride (TGCC-A/TGCC-C), endowed with enhanced flocculation-sterilization properties. The impacts of these flocculants on proliferation and transformation of antibiotic resistance genes (ARGs) among bacteria during the flocculation-deposition process were examined. TGCC-A/TGCC-C exhibited remarkable flocculation capacities towards both Escherichia coli and Staphylococcus aureus, encompassing a logarithmic range of initial cell density (108-109 CFU/mL) and a broad pH spectrum (

Many epidemiological studies have demonstrated a strong association between H. pylori (Helicobacter pylori) infection and human gastric cancer development. The precise mechanisms accounting for gastric cancer development induced by H. pylori infection are still not completely understood. However, it should be reasonable to assume that there are two distinct molecular pathways for gastric carcinogenesis by H. pylori infection; the direct action of the bacteria itself on gastric epithelial cells, and the accumulation of genetic changes caused by prolonged bacterial infection and chronic inflamma

Live attenuated Shigella vaccines have shown promise in inducing protective immune responses in human clinical trials and as carriers of heterologous antigens from other mucosal pathogens. In the past, construction of Shigella vaccine strains relied on classical allelic exchange systems to genetically engineer the bacterial genome. These systems require extensive in vitro engineering of long homologous sequences to create recombinant replication-defective plasmids or phage. Alternatively, the lambda red recombination system from bacteriophage facilitates recombination with as little as 40 bp o

Accurate computer-aided diagnosis of pediatric pneumonia remains challenging due to limited annotated data. To systematically address this challenge, a spatially transformed and adversarially realigned vision transformer (STAR-ViT) is proposed. STAR-ViT integrates spatial transformation consistency and adversarial cross-modal realignment within a single architecture. The spatial transformation module enforces feature consistency under random perturbations to enhance translation robustness, while the adversarial realignment module employs a domain discriminator to extract modality-invariant fea

Microbes in the intestine transform bile acids during transit, altering their functional and signaling capacities before recirculation via the portal vein. Sex differences in the gut microbiota have been noted, but their consequence on bile acid composition is unclear. Here, we investigated the composition and functional potential of microbes in the small and large intestines together with portal and systemic bile acid levels. Female and male mice exhibit distinct microbial diversity throughout the length of the intestine, leading to dimorphism in genes related to bile acid transformation. Of

Carcinoma ex pleomorphic adenoma (Ca ex PA) is a rare malignant transformation of a benign primary pleomorphic adenoma (PA). We report the case of a 62-year-old male who presented with a swelling over his left temple. Imaging revealed a lytic lesion over the left orbital wall with soft tissue extension suggestive of malignancy. He underwent an en bloc resection of the mass with orbital exenteration, craniotomy, and reconstruction. Pathology demonstrated a lacrimal gland ductal adenocarcinoma arising from a PA which led to the diagnosis of ductal adenoCa ex PA. Postoperatively, he received chem

In concern with frequent recurrence of anthrax in endemic areas and inadvertent use of its spores as biological weapon, the development of an effective anthrax vaccine suitable for both human and veterinary needs is highly desirable. A simple oral delivery through expression in plant system could offer promising alternative to the current methods that rely on injectable vaccines extracted from bacterial sources. In the present study, we have expressed protective antigen (PA) gene in Indian mustard by Agrobacterium-mediated transformation and in tobacco by plastid transformation. Putative trans

Antibiotic and herbicide resistance genes are currently the most frequently used selectable marker genes for plant research and crop development. However, the use of antibiotics and herbicides must be carefully controlled because the degree of susceptibility to these compounds varies widely among plant species and because they can also affect plant regeneration. Therefore, new selectable marker systems that are effective for a broad range of plant species are still needed. Here, we report a simple and inexpensive system based on providing transgenic plant cells the capacity to convert a nonmet

Mercury pollution is a major environmental problem that arises as a result of natural processes as well as from anthropogenic sources. In response to toxic mercury compounds, microbes have developed astonishing array of resistance systems to detoxify them. To address this challenge, this study was aimed in screening bacterial isolates for their tolerance against varied concentrations of phenylmercuric acetate. Mercury transformation by bacteria being sensitive to factors such as available carbon source, etc. that affect mer-mediated transformation, screened mercury tolerant bacteria were also

UNLABELLED: Traditional reverse genetics strategies for large-genome RNA viruses are hindered by multiple technical bottlenecks, including dependence on specific restriction enzyme sites, cumbersome multi-step cloning, and genetic instability of oversized DNA in bacterial systems. Herein, we established a universal reverse genetics platform for infectious bronchitis virus (IBV) through systematic optimization of the transformation-associated recombination (TAR) technology. By dividing the IBV genome into seven overlapping fragments and employing Saccharomyces cerevisiae for seamless assembly,

Several 1- and 2-carbon halogenated aliphatic organic compounds present at low concentrations (less than 100 micrograms/liter) were degraded under methanogenic conditions in batch bacterial cultures and in a continuous-flow methanogenic fixed-film laboratory-scale column. Greater than 90% degradation was observed within a 2-day detention time under continuous-flow methanogenic conditions with acetate as a primary substrate. Carbon-14 measurements indicated that chloroform, carbon tetrachloride, and 1,2-dichloroethane were almost completely oxidized to carbon dioxide, confirming removal by bioo
