Factors limiting motor recovery after facial nerve transection in the rat: combined structural and functional analyses.

It is believed that a major reason for the poor functional recovery after peripheral nerve lesion is collateral branching and regrowth of ax … of motoneurons, more than two-fold as compared with control animals. However, biometric analysis of vibrissae movements did not show positive effects on functional recovery, suggesting that polyneuronal reinnervation--rather than collateral branching --may be the critical limiting factor. In support of this hypothesis, we found that motor end-plates with mo …

Deletion of the GATA domain of TRPS1 causes an absence of facial hair and provides new insights into the bone disorder in inherited tricho-rhino-phalangeal syndromes.

GATA transcription factors mediate cell differentiation in diverse tissues, and their dysfunction is associated with certain congenital huma … th findings for TRPS, whereas TRPS1(Deltagt/Deltagt) mice additionally reveal a complete absence of vibrissae. Unexpectedly, TRPS1(Deltagt/Deltagt) mice die of neonatal respiratory failure resulting from abnormalities of the thoracic spine and ribs. Heterozygotes also develop thoracic kyphoscoliosis with age and reveal structural deficits in cortical and tr …

Niche partitioning and individual specialisation in resources and space use of sympatric fur seals at their range margin.

Ecological theory predicts niche partitioning between high-level predators living in sympatry as a mechanism to minimise the selective press … space. We found some niche overlap, yet clear partitioning in diet trophic level (δ15N values from vibrissae), spatial niche space (horizontal and vertical telemetry data) and circadian activity patterns (timing of dives) between males of each species, suggesting competition may remain an active driver of niche partitioning amongst individuals even in smal …

Agmatine treatment and vein graft reconstruction enhance recovery after experimental facial nerve injury.

The rate of nerve regeneration is a critical determinant of the degree of functional recovery after injury. Here, we sought to determine whe … th unsutured vein graft followed by agmatine treatment. Functional recovery, as assessed by grading vibrissae movements and by recording nerve conduction velocity and numbers of regenerated axons, indicated that either vein reconstruction or agmatine treatment resulted in accelerated and more complete recovery as compared with controls. Somatosensory evoked potentials in rat cerebral cortex before and after middle cerebral artery occlusion.

We recorded somatosensory evoked potentials in pentobarbital-anesthetized rats before and after middle cerebral artery occlusion. Trigeminal (vibrissae), median (forelimb), and sciatic (hind limb) nerve stimuli produced consistent, robust, and sharply localized responses in the trigeminal, forelimb, and hind limb regions of the somatosensory cortex of 18 rats. These regions are situated at sequentially greater distances from the center of infarcts produced by middle cerebral artery occlusion. In eight rats, occlusion 1-2 mm below the rhinal fissure abolished somatosensory evoked potentials in …

Topography of distance-modulated multisensory object location encoding in mouse area RL.

For animals to interact coherently with the external world, their brains must integrate object location information across sensory modalities, an inherently complex process. Each sensory modality samples distinct regions of external space, with object location encoded in fundamentally different reference frames: for example, visual representation of space begins with retinotopic maps, and tactile representation of space in the rodent whisker system starts with somatotopic maps.1,2,3,4,5,6 To form an integrated representation of objects in space, the brain must reconcile differences in spatial

The rabbit as a behavioral model system for magnetic resonance imaging.

BACKGROUND: fMRI requires that subjects not move during image acquisition. This has been achieved by instructing people not to move, or by anesthetizing experimental animal subjects to induce immobility. We have demonstrated that a surgically implanted headbolt onto the skull of a rabbit allows their brain to be imaged comfortably while the animal is awake. This article provides a detailed method for the preparation. NEW METHOD: We took advantage of the rabbit's tolerance for restraint to image the brain while holding the head at the standard stereotaxic angle. Visual stimulation was produced

Rabbit facial nerve regeneration in autologous nerve grafts after antecedent injury.

OBJECTIVE: The effect of incomplete antecedent injuries on subsequent facial nerve regeneration within cable graft repairs is not known. The purpose of this study is to compare facial nerve regeneration after an immediate and delayed neural cable graft repair. METHOD: Rabbit facial nerve regeneration after complete transectional injuries of the buccal division was compared in two experimental models. In one, a 10-mm segment of the nerve was transected, rotated 180 degrees, and immediately repaired as a cable graft (N=8). In the second, a preliminary nerve crush was allowed to recover over a 4

Abnormal Whisker-Dependent Behaviors and Altered Cortico-Hippocampal Connectivity in Shank3b-/- Mice.

Abnormal tactile response is an integral feature of Autism Spectrum Disorders (ASDs), and hypo-responsiveness to tactile stimuli is often associated with the severity of ASDs core symptoms. Patients with Phelan-McDermid syndrome (PMS), caused by mutations in the SHANK3 gene, show ASD-like symptoms associated with aberrant tactile responses. The neural underpinnings of these abnormalities are still poorly understood. Here we investigated, in Shank3b-/- adult mice, the neural substrates of whisker-guided behaviors, a key component of rodents' interaction with the surrounding environment. We asse

Neuroprotective effects of kolaviron against psycho-emotional stress induced oxidative brain injury in rats: The whisker removal model.

BACKGROUND: The study investigated the neuroprotective potentials of kolaviron (a biflavonoid complex of Garcinia kola) against psycho-emotional stress induced oxidative brain injury in Wistar rats. METHODS: Twenty-four adult Wistar rats (180-220g) randomly divided into four groups (1-1V,n=6) were used for the study . Group 1 served as control (non stressed), group 11 consisted of stressed rats induced by complete removal' of the whiskers around the mouth and the nose without anaesthesia. The rats in group 111 were pre- treated with 200mg/kg kolaviron per oral (p.o), daily for seven days befo

[Functional mapping of the motor cortex in the white mouse by microstimulation].

Motor representations of the face muscles and muscles of limbs were determined by means of intracortical microstimulation (ICMS) in 33 anaesthetized white mice. ICMS evoked mostly ipsilateral motor responses of facial muscles and contralateral motor responses of forelimb and hindlimb. Clear asymmetry of the face muscles's motor representation was found using low-threshold stimulation in left and right hemispheres. Hindlimb's motor responses to ICMS in rostral areas of frontal neocortex were observed and it indicates the presence of multiple motor representations in the mouse neocortex.

Electrophysiological examination of the representation of the face in the suprasylvian gyrus of the ferret: a correlative study with cytoarchitecture.

Using high-resolution microelectrode mapping methods, we explored the organization of the face representation within the primary somatosensory cortex of ferrets, finding evidence for at least two and probably four representations of the face distributed consecutively from anterior to posterior along the long axis of the suprasylvian gyrus. Examination of the cytoarchitecture (Rice et al., this issue) revealed that these four areas corresponded to four different cytoarchitectonic fields within the crown of the suprasylvian gyrus. The two central, most completely defined representations were ori

Fasciculation in the early mouse trigeminal nerve is not ordered in relation to the emerging pattern of whisker follicles.

Detailed reconstructions of lengths of the embryonic mouse maxillary nerve were made from serial light and electron microscope sections to determine whether there is any correspondence between the arrangement of fasciculi in the developing nerve and the emerging pattern of whisker follicles on the snout. There was neither correspondence nor obvious pattern in the arrangement of fasciculi at either E11, when trigeminal nerve fibers first contact the presumptive whisker pad, or at E12, when the pattern of developing whisker follicles becomes apparent. Fasciculi merged and branched to form an int

Development and lesion induced reorganization of the cortical representation of the rat's body surface as revealed by immunocytochemistry for serotonin.

Immunocytochemistry with an antiserum directed against serotonin (5-HT) was used to assess the development of the representation of the body surface in the rat's primary somatosensory cortex (S-I). Within 1 hour of birth (P-O), 5-HT-positive fibers were present in the marginal zone, the cortical plate, and developing layers V and VI. Immunoreactivity in the marginal zone consisted of a thin band of coarse fibers oriented parallel to the pia. Only a small number of isolated fibers were visible in the cortical plate. A denser network of both coarse and fine fibers could be seen in presumptive la

Functional role of peripheral P2X3 receptor expression in trigeminal ganglion neurons: a microarray-based evaluation of the antagonist effect of P2X3.

The P2X receptor is an extracellular adenosine triphosphate (ATP)-gated ion channel that plays critical roles in cellular and developmental processes, including cell proliferation, survival, and differentiation. Specifically, P2X3 is essential for ATP to function as a neurotransmitter in the peripheral nervous system. Despite the role of P2X3 in nociception, its specific contribution to the development of the trigeminal ganglion (TG), particularly across different developmental stages, remains undetermined. Therefore, in this study, we measured P2X3 expression in the mouse TG and peripheral ti

Hormonal mechanisms in the paraventricular nuclei associated with hyperalgesia in Parkinson's disease model rats.

Pain is a major non-motor symptom of Parkinson's disease (PD). The relationship between hyperalgesia and neuropeptides originating from paraventricular nucleus (PVN) in 6-hydroxydopamine (6-OHDA) rats has already been investigated for oxytocin (OXT), but not yet for arginine vasopressin (AVP) and corticotropin-releasing hormone (CRH). The present study aimed to investigate the alterations in these neuropeptides following nociceptive stimulation in PD model rats and to examine the mechanisms of hyperalgesia. Dopaminergic nigrostriatal lesions were induced by injecting 6-OHDA into the medial for

A brainstem circuit for the expression of defensive facial reactions in rat.

The brainstem houses neuronal circuits that control homeostasis of vital functions. These include the depth and rate of breathing1,2 and, critically, apnea, a transient cessation of breathing that prevents noxious vapors from entering further into the respiratory tract. Current thinking is that this reflex is mediated by two sensory pathways. One known pathway involves vagal and glossopharyngeal afferents that project to the nucleus of the solitary tract.3,4,5 Yet, apnea induced by electrical stimulation of the nasal epithelium or delivery of ammonia vapors to the nose persists after brainstem

Hair follicle associated pluripotent (HAP) stem cells jump from transplanted whiskers to pelage follicles and stimulate hair growth.

Stimulation of hair growth in hair loss has been a difficult goal to achieve. Hair follicle-associated pluripotent (HAP) stem cells express nestin and have been shown to differentiate to multiple cell types including keratinocytes, neurons, beating cardiac muscles and numerous other cell types. HAP stem cells originate in the bulge area of the hair follicle and have been shown to migrate within and outside the hair follicle. In the present study, the upper part of vibrissa follicles from nestin-driven green-fluorescent protein (GFP) transgenic mice, containing GFP-expressing HAP stem cells, we
