Integrated biomarker response to assess the effects of pesticide residues on Nile Tilapia in aquatic ecosystems contaminated by cotton-field effluents.

An in-situ study combined with an integrated biomarker response was used to evaluate the impact of agricultural effluents in the physiological responses of Nile tilapia reared in cages and enclosures of water reservoirs in North Benin. Fish were distributed in fish farming systems at two sites: Songhai located outside the cotton basin and Batran located in the most productive commune. They were sampled for blood and organs before (BST), during (DST) and after (AST) pesticide treatment. Pesticide residues were analysed in water, sediments and fish muscles. Several biomarkers were investigated r

Insect Body Defence Reactions against Bee Venom: Do Adipokinetic Hormones Play a Role?

Bees originally developed their stinging apparatus and venom against members of their own species from other hives or against predatory insects. Nevertheless, the biological and biochemical response of arthropods to bee venom is not well studied. Thus, in this study, the physiological responses of a model insect species (American cockroach, Periplaneta americana) to honeybee venom were investigated. Bee venom toxins elicited severe stress (LD50 = 1.063 uL venom) resulting in a significant increase in adipokinetic hormones (AKHs) in the cockroach central nervous system and haemolymph. Venom tre

Sox genes in Agasicles hygrophila (Coleoptera: Chrysomelidae) are involved in ovarian development and oogenesis.

The alligator weed flea beetle, Agasicles hygrophila is a monophagous natural enemy of the invasive alligator weed Alternanthera philoxeroides. Oogenesis plays a vital role in the process of individual development and population continuation of oviparous insects. Sox is an ancient and ubiquitous metazoan gene family that plays a key regulatory role in various physiological processes, including oogenesis, which is closely related to fecundity. In this study, two Sox genes AhDichaete and AhSox3 were cloned and characterized, and then the expression profiles of AhDichaete and AhSox3 were qualifie

Endocrine-disrupting effects of spironolactone in female western mosquitofish, Gambusia affinis.

The discovery of pharmaceuticals in effluent from wastewater treatment plants and drug manufacturing facilities and in receiving waters has raised environmental concern. Because these compounds are ending up in the environment, it is important to investigate the effects of these compounds on wildlife as well as humans. The present study used a fish model to investigate the endocrine-disrupting effects of spironolactone (SPL), an aldosterone antagonist used as a diuretic, but which also exhibits antiandrogenic effects in humans. A dose-response study measured the effects of SPL on anal fin ray

A study of somatolactin actions by ectopic expression in transgenic zebrafish larvae.

Somatolactin (SL) is a fish-specific hormone that belongs to the prolactin (PRL) and GH family. Recently, two forms of SL, SLα and SLβ, have been found in some species, and may have different actions and functions. To investigate the role of SL in fish growth and metabolism, we generated transgenic fish founders with ectopic expression of SLα and SLβ to study the physiological functions and actions of these SLs among several marker genes. We fused the cDNAs encoding the precursor SLs in frame to a zebrafish β-actin gene promoter to generate transgenic zebrafish lines that were coinjected with

Evaluation of the Effect of Adipokinetic Hormone/Corazonin-Related Peptide (ACP) on Ovarian Development in the Mud Crab, Scylla paramamosain.

In this study, we identified Sp-ACP and its putative receptor from the mud crab S. paramamosain and explored its potential role in ovarian development. RT-PCR results suggested Sp-ACP was extensively expressed in nervous tissues, the ovary, the middle gut, and the Y-organ, while Sp-ACPR was highly expressed in the ovary. The expression level of Sp-ACP in the ovary, eyestalk, and cerebral ganglia gradually increased during ovarian development, whereas its receptor exhibited an opposite expression pattern in the ovary. Immunofluorescence revealed that ACP was specifically localized in the follic

Juvenile hormone and its receptor methoprene-tolerant promote ribosomal biogenesis and vitellogenesis in the Aedes aegypti mosquito.

Juvenile hormone (JH) controls many biological activities in insects, including development, metamorphosis, and reproduction. In the Aedes aegypti mosquito, a vector of dengue, yellow fever, chikungunya, and zika viruses, the metabolic tissue (the fat body, which is an analogue of the vertebrate liver) produces yolk proteins for developing oocytes. JH is important for the fat body to acquire competence for yolk protein production. However, the molecular mechanisms of how JH promotes mosquito reproduction are not completely understood. In this study we show that stimulation of the JH receptor m

Transgenic Expression of the piRNA-Resistant Masculinizer Gene Induces Female-Specific Lethality and Partial Female-to-Male Sex Reversal in the Silkworm, Bombyx mori.

In Bombyx mori (B. mori), Fem piRNA originates from the W chromosome and is responsible for femaleness. The Fem piRNA-PIWI complex targets and cleaves mRNAs transcribed from the Masc gene. Masc encodes a novel CCCH type zinc-finger protein and is required for male-specific splicing of B. mori doublesex (Bmdsx) transcripts. In the present study, several silkworm strains carrying a transgene, which encodes a Fem piRNA-resistant Masc mRNA (Masc-R), were generated. Forced expression of the Masc-R transgene caused female-specific lethality during the larval stages. One of the Masc-R strains weakly

Effects of Dietary Exposure to Zearalenone (ZEN) on Carp (Cyprinus carpio L.).

The mycotoxin zearalenone (ZEN) is frequently contaminating animal feeds including feed used in aquaculture. In the present study, the effects of dietary exposure to ZEN on carp (Cyprinus carpio L.) were investigated. ZEN at three different concentrations (low dose: 332 µg kg(-1), medium dose: 621 µg kg(-1) and high dose: 797 µg kg(-1) final feed, respectively) was administered to juvenile carp for four weeks. Additional groups received the mycotoxin for the same time period but were fed with the uncontaminated diet for two more weeks to examine the reversibility of the ZEN effects. No effects

Increased Akt signaling in the mosquito fat body increases adult survivorship.

Akt signaling regulates diverse physiologies in a wide range of organisms. We examine the impact of increased Akt signaling in the fat body of 2 mosquito species, the Asian malaria mosquito Anopheles stephensi and the yellow fever mosquito Aedes aegypti. Overexpression of a myristoylated and active form of A. stephensi and Ae. aegypti Akt in the fat body of transgenic mosquitoes led to activation of the downstream signaling molecules forkhead box O (FOXO) and p70 S6 kinase in a tissue and blood meal-specific manner. In both species, increased Akt signaling in the fat body after blood feeding s

Stressors, glucocorticoids and ovarian function in teleosts.

The purpose of this overview is to re-examine the postulated direct and indirect actions of glucocorticoids on ovarian function in teleosts. The re-examination is undertaken in light of recent advances in the understanding of the stress response itself, the mode of action of the hypothalamus-pituitary gland-ovarian (HPO) axis, the mechanisms of control of oestrogen-dependent hepatic vitellogenin (VtG) secretion and the apparent roles of corticotrophin-releasing hormone (CRH) and CRH-related factors in the regulation of feeding activity. Many of the results of different studies, particularly wh

Physiological changes in reproductively active rainbowfish (Melanotaenia fluviatilis) following exposure to naphthalene.

Naphthalene makes up a substantial fraction of polycyclic aromatic hydrocarbons (PAHs) in crude oil and is an important by-product of industry; however, few studies have investigated the toxicity of naphthalene to aquatic organisms. We examined the toxicity of increasing concentrations (0, carrier control, 130, 200 and 400microg/l) of naphthalene to adult rainbowfish (Melanotaenia fluviatilis) for 3 and 14 days to determine its potential to act as an endocrine disruptor. After exposure for 3 days, no changes in sex steroids were measured. After 14 days, a decrease of serum estradiol in females

Reproductive inhibition and transgenerational toxicity of triphenyltin on medaka (Oiyzias latipes) at environmentally relevant levels.

An increasing number of studies have reported unexpectedly high body burdens of triphenyltin (TPT) in wild fishes around the world. To assess the effects of TPT on fish, we exposed pairs of medaka (Oryzias latipes) to different levels of TPT for 5 weeks, and the reproduction responses and transgenerational effects were studied. The results demonstrated that TPT exposure markedly suppressed the spawning frequency, spawned egg number, egg quality and gonad development, and induced teratogenesis, such as hemorrhaging, eye defects, morphological malformation and conjoined twins, less hatchability,

Synergistic Roles of Linoleic and Linolenic Acids in Regulating Lc-PUFA Biosynthesis and Reproductive Physiology of Striped Murrel (Channa striata) Broodstock.

Fatty acids are crucial for steroidogenesis and gonadal development in fish. A 90-day feeding experiment was conducted before maturation to assess the effects of different dietary levels of linoleic acid (LA) and linolenic acid (LNA) on gonadal development and reproductive performance in Channa striata broodstocks. A total of 300 male and female broodstocks were divided into five treatment groups, each with three replicates, using a completely randomized design. The fish were fed five experimental diets that were isonitrogenous (42%) and isolipidic (12%), containing 0% (B0), 0.5% LA + 1% LNA (

Dual functions of the Aedes aegypti ecdysone receptor in dengue virus replication and reproduction control.

BACKGROUND: The ecdysone receptor (EcR) is a central regulator of mosquito physiology, best known for its role in vitellogenesis. However, its contribution to antiviral defense and dengue virus (DENV) replication in Aedes aegypti remains poorly understood. METHODS: RNA interference was used to silence Aedes aegypti EcR (AaEcR). Effects on DENV replication, immune gene expression, ovarian development, vitellogenin (Vg) synthesis, and target of rapamycin (TOR) pathway activity were assessed using molecular, cellular, and phenotypic analyses. RESULTS: Silencing AaEcR markedly suppressed DENV re

Energy reserves, rather than body size, are associated with ovarian development in two-year-old female European sea bass (Dicentrarchus labrax L.).

In fish, early puberty can lead to divergent growth trajectories and increased size dispersion within populations. In European sea bass, this pattern is evident by the end of the second year, with females of greater body weight generally tending to reach puberty earlier, although this association is not always consistent. This study investigates the factors influencing the relationship between growth and reproductive maturation in this teleost fish. Four female groups were defined based on body weight (small, S; large, L) and gonadal development (immature, I; advanced, A): SI, SA, LI, and LA.

Transcription factor E93 regulates vitellogenesis via the vitelline membrane protein 26Ab gene in Chilo Suppressalis.

BACKGROUND: Ecdysone-induced protein 93 F (E93, also known as Eip93F) plays a crucial role in the reproductive process of numerous insects. This study aims to delineate the function of E93 in Chilo suppressalis and elucidated the regulatory mechanism by which E93 influences the reproduction of C. suppressalis METHODS AND RESULTS: The results of the bioinformatics analysis indicate that C. suppressalis E93 shows the highest homology with E93 from Bombyx mori. We used qPCR to evaluate the expression profile of CsE93 from different developmental stages and tissues, revealed that CsE93 had the hig

Identification and analysis of JHBP/TO family genes and their roles in the reproductive fitness cost of resistance in Frankliniella occidentalis (Pergande).

The juvenile hormone binding protein (JHBP) and takeout (TO) genes, mediated by the juvenile hormone (JH), play a crucial role in regulating the reproductive physiology of insects. Our previous study revealed that spinosad-resistant Frankliniella occidentalis (NIL-R) exhibited reduced fecundity and significant changes in JHBP/TO family gene expression. We hypothesized that these genes were involved in regulating the fitness costs associated with resistance. In this study, 45 JHBP/TO genes were identified in F. occidentalis, among which FoTO2 and FoTO10 were duplicates. Additionally, eight gene
