Gastroesophageal reflux and chronic cough.

Gastroesophageal reflux (GOR) disease is one of the 3 commonest causes of chronic cough. It can be difficult to diagnose as the traditionally recognised symptoms of GOR, such as heartburn and acid regurgitation, are often absent. More subtle indicators of a link between the cough and the oesophagus should therefore be sought. These include cough which occurs in relation to eating or phonation, cough which settles at night and does not tend to wake the patient from sleep and symptoms suggestive of laryngopharyngeal reflux. Investigations such as oesophageal manometry and 24 hour pH monitoring c

Pseudomonas putida B2: a tod-lux bioluminescent reporter for toluene and trichloroethylene co-metabolism.

A tod-lux transcriptional fusion bioluminescent reporter strain, Pseudomonas putida B2, was developed to permit on-line analysis of trichloroethylene (TCE) transformation by toluene dioxygenase (todC1C2BA) in Pseudomonas putida F1. Strain B2 was exposed to toluene in growing and resting cell bioluminescence assays. The growing cells showed a direct correlation between bioluminescence and toluene concentration, while resting cells showed reproducible bioluminescence with repeated toluene exposures. In addition, P. putida B2 was encapsulated in alginate beads and used in a packed bed flow-throug

A cell biosensor specific for formaldehyde based on pH-sensitive transistors coupled to methylotrophic yeast cells with genetically adjusted metabolism.

A cell biosensor specific for formaldehyde was developed using double-mutant cells of the methylotrophic yeast Hansenula polymorpha A3-11. The activities of some of the enzymes in the metabolic pathway of the wild-strain cells were deliberately suppressed by introducing respective genetic blocks to optimize the selectivity and acidification rate. Mutant yeast cells produced in this way were immobilized in Ca-alginate gel on the gate of a pH-sensitive field effect transistor. The local acidification of the extracellular medium due to specific conversion of formaldehyde was recorded. The steady-

Microencapsulated parathyroid cells as a bioartificial parathyroid. In vivo studies.

Parathyroid cells were isolated from healthy rats, encapsulated in alginate-polylysine membranes, and injected intraperitoneally into rats on which total parathyroidectomies had been performed. Three days posttransplant, serum calcium and PTH-M concentrations had increased to near-normal levels in the recipient animals. Similar results were observed in a separate group of parathyroidectomized rats 3 days after free parathyroid cells were implanted, but within 4 weeks serum calcium and PTH-M concentrations had decreased almost to pretransplant levels in these rats. In the rats with encapsulated

Signal peptides for engineered protein secretion by free and hydrogel-encapsulated cyanobacteria.

Cyanobacteria provide a promising platform for light-driven heterologous protein production. In heterologous production systems, cells are typically lysed for product purification or to facilitate interaction with external substrates, unless the product is secreted. Fusion to molecular "shipping labels" known as signal peptides can target chimeric proteins to native membrane translocation pathways and result in secretion. In this work, we identified signal peptides from the model cyanobacterium Synechococcus elongatus PCC 7942 (S. elongatus) and genetically fused to two heterologous enzymes: t

Human colon tumor-on-a-chip based on magnetic microcontainers encapsulated cancer spheroids.

Tumor-on-a-chip platforms are increasingly recognized as promising preclinical tools for developing personalized therapies. However, effectively recapitulating tumor heterogeneity, three-dimensional architecture, and the dynamic in vivo microenvironment remains a significant challenge. Here, we developed magneto-responsive microcontainers for controllable culture of the patient-derived colorectal tumor cells (pCRCs) spheroids. Using microfluidic electrospray technology, microcontainers with a biocompatible carboxymethyl cellulose (CMC) core and a magnetic Fe3O4@alginate shell were fabricated f

Digital Light Processing of Three-dimensional Liquid Metal Hydrogels.

Liquid metal (LM) hydrogels emerge as promising materials for flexible electronics and soft robotics, but they are challenged by simplistic structure, limited precision, and insufficient functional designability. Herein, we demonstrate the first digital light processing of three-dimensional (3D) LM hydrogels with micrometer precision (10 µm) and arbitrarily elaborated architectures. A universal strategy is developed for photo-curable LM-based various resins by encapsulation of LM droplets with sodium alginate (SA). The SA coating not only prevents the ink from sedimentation and self-polymeriza

Chairside digital workflow to obtain an optimized 3D model of a partially edentulous mandible.

Intraoral scanners generate accurate optical impressions for dentate areas, but wide edentulous spans remain challenging due to limited stable landmarks and mobile mucosa. This article describes a single-visit, chairside workflow that combines an intraoral scan with a digitized irreversible hydrocolloid impression to obtain an optimized three-dimensional model of a partially edentulous mandible suitable for computer aided design-based removable partial denture fabrication other prosthetically driven procedures. The workflow includes intraoral scanning, conventional alginate impression taking f

Bacteriophages and polymeric delivery systems: a mapping review.

Bacteriophages are viruses that specifically infect bacteria and have emerged as promising agents for bacterial control in human, animal, and environmental fields. On the other hand, their practical application is limited by instability outside the bacterial hosts, which compromises their efficacy. The incorporation of bacteriophages into polymeric materials has been explored as a strategy to protect the viruses from rapid environmental inactivation and, in some cases, to allow controlled release. In this review, association maps provide a broad overview of the main polymers and fabrication ap

Design of an injectable magnetic hydrogel with porous structure and electrocatalytic activity for the sensitive electrochemical detection of nitrite in foods.

Herein, we synthesized a novel injectable porous magnetic hydrogel (MHG) at room temperature using carboxymethyl chitosan (CMCS), polydopamine (PDA), sodium alginate (SA), polyethyleneimine (PEI) and copper ferrite (CuFe2O4) as building blocks. The CMCS and SA as monomers provided good film-forming and anti-fouling properties for MHG. The PDA-coated CuFe2O4 as a cross-linking agent improved the homogeneity, adsorption and electrocatalytic performance of MHG, but also generated a macroporous hydrogel structure which was beneficial for sensing applications. The positive PEI enhanced the adsorpti

Multifunction hydrogen-bonded organic framework aerogel platform for detection and removal of heavy metal ions in pear juice.

Heavy metal ions (HMIs) in food pose significant health risks due to their bioaccumulation and toxicity, necessitating effective detection and remediation methods. This study explores a multifunctional hydrogen-bonded organic framework (HOF-16) wrapped in sodium alginate (SA) to form a stable HOF-16/SA aerogel for detecting and removing HMIs in pear juice. The HOF-16 fluorescence probe exhibits excellent sensitivity with limits of detection of 0.49, 0.60, and 0.44 μmol/L for Pb(II), Cu(II), and Cd(II), respectively. The maximum adsorption capacities of HOF-16/SA aerogel for Pb(II), Cu(II), and

Development of a tissue-specific bioscaffold for intestinal stem cell culture.

The generation of a tissue-specific intestinal hydrogel derived from the native intestine has the potential to support and promote the growth of intestinal organoids. In this study, we aimed to develop hydrogels derived exclusively from intestinal extracellular matrix (ECM) or composites comprised of intestinal ECM combined with alginate that allow for greater tuning of the hydrogel properties. A novel mouse intestinal decellularization protocol was developed and the ECM characterized. Our analyses demonstrate that our protocol effectively removed cellular and nuclear content while preserving

Fabrication of curcumin-loaded bovine serum albumin (BSA)-polymannuronate nanoparticles and the therapeutic effects for ulcerative colitis.

In this study, alginate-derived polymannuronate (PM) was covalently conjugated to Bovine serum albumin (BSA) via the Maillard reaction to increase the stability of BSA nanoparticles (NPs) and improve the delivery efficiency of curcumin (Cur) to alleviate ulcerative colitis (UC). First, the BSA-PM glycoprotein was synthesized and characterized, and its ability to inhibit the heat-induced structural denaturation of BSA was demonstrated. Cur was subsequently encapsulated in the NPs to yield Cur-BSA-PM NPs, which displayed a spherical morphology and demonstrated increased physical stability during

Efficient active hydrogen delivery for drug-free radiation enteritis therapy in mice.

Radiation enteritis, affecting over 90% of pelvic/abdominal radiotherapy patients, is primarily caused by radiation-induced reactive oxygen and nitrogen species (RONS). Active hydrogens, with broad-spectrum RONS scavenging ability, show radioprotective potential but face delivery challenges due to the intestinal mucus barrier and short lifespan. Here, we show drinkable, self-thermophoretic sodium alginate/chitosan oligosaccharide-coated hydrogenated molybdenum oxide nanomachines (HxMoO3@SA@COSs) that exhibit near-infrared (NIR)-driven directional motility and sustained active hydrogen release.

Cardiac repair using regenerating neonatal heart tissue-derived extracellular vesicles.

Neonatal mammalian hearts are capable of regenerating by inducing cardiomyocyte proliferation after injury. However, this regenerative capability in adult mammalian hearts almost disappears. Extracellular vesicles (EVs) have been shown to play vital cardioprotective roles in heart repair. Here, we report that EVs from regenerating neonatal heart tissues, after apical resection surgery (AR-Neo-EVs), exhibit stronger pro-proliferative, anti-apoptotic, and pro-angiogenesis activities than EVs from neonatal mouse cardiac tissues (Neo-EVs), effects which are absent in adult mouse heart-derived EVs

Oral microsphere formulation of M2 macrophage-mimetic Janus nanomotor for targeted therapy of ulcerative colitis.

Oral medication for ulcerative colitis (UC) is often hindered by challenges such as inadequate accumulation, limited penetration of mucus barriers, and the intricate task of mitigating excessive ROS and inflammatory cytokines. Here, we present a strategy involving sodium alginate microspheres (SAMs) incorporating M2 macrophage membrane (M2M)-coated Janus nanomotors (denominated as Motor@M2M) for targeted treatment of UC. SAM provides a protective barrier, ensuring that Motor@M2M withstands the harsh gastric milieu and exhibits controlled release. M2M enhances the targeting precision of nanomot

Vibrio cholerae virulence is blocked by chitosan oligosaccharide-mediated inhibition of ChsR activity.

Vibrio cholerae causes cholera, an important cause of death worldwide. A fuller understanding of how virulence is regulated offers the potential for developing virulence inhibitors, regarded as efficient therapeutic alternatives for cholera treatment. Here we show using competitive infections of wild-type and mutant bacteria that the regulator of chitosan utilization, ChsR, increases V. cholerae virulence in vivo. Mechanistically, RNA sequencing, chromatin immunoprecipitation with sequencing and molecular biology approaches revealed that ChsR directly upregulated the expression of the virulenc
