Trends and Perspectives in the Targeting of Brain Through Ethosomal Formulations.

Neurological diseases such as Alzheimer's disease, Schizophrenia, anxiety, Parkinson's disease, and migraine are serious conditions that continue to threaten mankind. The cases of brainrelated disorders are increasing worldwide and are closely related to physiological, genetic, and environmental factors. Direct drug delivery to the brain is crucial for the effective treatment and prevention of these conditions. However, due to the presence of a lipophilic barrier, i.e., the bloodbrain barrier, the entry of therapeutic agents into the brain is restricted, resulting in a lower concentration at t

Understanding the Potential of Antibody-Drug Conjugates Functionalized Engineered Exosomes in Hepatocellular Carcinoma Therapy: A Comprehensive Narrative Review.

BACKGROUND: Exosomes are naturally secreted nanovesicles with high biocompatibility, low immunogenicity, and the ability to traverse biological barriers, making them attractive theragnostic carriers. When engineered to present tumor-specific antibodies and to deliver antibody-drug conjugates (ADCs), they may enhance selective targeting of hepatocellular carcinoma (HCC). METHODS: This narrative review synthesizes the multidisciplinary field of exosome bioengineering, ADCs, and HCC diagnosis and therapy, covering topics from click chemistry to translational challenges. A comprehensive search of

Dysfunction of Primary Cilium in Huntington's Disease: How Mutant Huntingtin Disrupts This Cellular Signaling Hub.

Primary cilia are solitary, antenna-like organelles that project from the surface of most vertebrate cells. They consist of a microtubule-based axoneme extending from a modified centriole (basal body) and enclosed by a lipid bilayer membrane. For several decades after their discovery, the functions of primary cilia had remained speculative; they were even considered vestigial structures. Currently, primary cilium is recognized as essential sensory and signaling structure involved in both chemo- and mechanosensation. Its anchoring at the centrosome surrounded by a radially organized network of

Advances in the extracellular vesicles treatment of alopecia.

BACKGROUND: Alopecia is a prevalent condition that significantly impacts appearance, mental health, and overall quality of life. Current clinical treatments for alopecia include medication, hair transplantation, low-level light therapy, and stem cell therapy, among others. However, the effectiveness of these treatments remains far from ideal. Extracellular vesicles (EVs) have emerged as a promising therapeutic strategy, yet challenges, such as mass production and clinical application persist. This review aims to explore recent advancements in the use of EVs for the treatment of alopecia. METH

Monocellular and Multicellular Parasites Infesting Humans: A Review of Calcium Ion Mechanisms.

Calcium (Ca2+) is a signal messenger for ion flow in and out of microbial, parasitic, and host defense cells. Manipulation of calcium ion signaling with ion blockers and calcineurin inhibitors may improve host defense while decreasing microbial/parasitic resistance to therapy. Ca2+ release from intracellular storage sites controls many host defense functions (cell integrity, movement, and growth). The transformation of phospholipids in the erythrocyte membrane is associated with changes in deformability. This type of lipid bilayer defense mechanism helps to prevent attack by Plasmodium. The isoprenyl chain length of coenzyme Q mediates the nutritional resistance of fungi to amoeba predation.

UNLABELLED: Amoebae are environmental predators feeding on bacteria, fungi, and other eukaryotic microbes. Predatory interactions alter microbial communities and impose selective pressure toward phagocytic resistance or escape which may, in turn, foster virulence attributes. The ubiquitous fungivorous amoeba Protostelium aurantium has a wide prey spectrum in the fungal kingdom but discriminates against members of the Saccharomyces clade, such as Saccharomyces cerevisiae and Candida glabrata. Here, we show that this prey discrimination among fungi is solely based on the presence of ubiquinone a

Extracellular Vesicles in Sickle Cell Disease: Plasma Concentration, Blood Cell Types Origin Distribution and Biological Properties.

Prototype of monogenic disorder, sickle cell disease (SCD) is caused by a unique single mutation in the β-globin gene, leading to the production of the abnormal hemoglobin S (HbS). HbS polymerization in deoxygenated condition induces the sickling of red blood cells (RBCs), which become less deformable and more fragile, and thus prone to lysis. In addition to anemia, SCD patients may exhibit a plethora of clinical manifestations ranging from acute complications such as the frequent and debilitating painful vaso-occlusive crisis to chronic end organ damages. Several interrelated pathophysiologic

Protective effect of fungal extracellular vesicles against murine candidiasis.

Extracellular vesicles (EVs) are lipid bilayered compartments released by virtually all living cells, including fungi. Among the diverse molecules carried by fungal EVs, a number of immunogens, virulence factors and regulators have been characterised. Within EVs, these components could potentially impact disease outcomes by interacting with the host. From this perspective, we previously demonstrated that EVs from Candida albicans could be taken up by and activate macrophages and dendritic cells to produce cytokines and express costimulatory molecules. Moreover, pre-treatment of Galleria mellon

Investigating the Influence of Membrane Composition on Protein-Glycolipid Binding Using Nanodiscs and Proxy Ligand Electrospray Ionization Mass Spectrometry.

This work describes a versatile analytical approach, which combines the proxy ligand electrospray ionization mass spectrometry (ESI-MS) assay and model membranes of defined composition, to quantify the influence of lipid bilayer composition on protein-glycolipid binding in vitro. To illustrate the implementation of the assay (experimental design and data analysis), affinities of the monosialoganglioside ligand GM1, incorporated into nanodiscs (NDs), for cholera toxin B subunit homopentamer (CTB5) were measured. A series of NDs containing GM1 and cholesterol were prepared using three different

An ion gating mechanism of gastric H,K-ATPase based on molecular dynamics simulations.

Gastric H,K-ATPase is an electroneutral transmembrane pump that moves protons from the cytoplasm of the parietal cell into the gastric lumen in exchange for potassium ions. The mechanism of transport against the established electrochemical gradients includes intermediate conformations in which the transferred ions are trapped (occluded) within the membrane domain of the pump. The pump cycle involves switching between the E1 and E2P states. Molecular dynamics simulations on homology models of the E2P and E1 states were performed to investigate the mechanism of K(+) movement in this enzyme. We p

Short-term exposure to cigarette smoke induces endothelial dysfunction in small intrapulmonary arteries: analysis using guinea pig precision cut lung slices.

Recent studies indicate that low concentrations of acetaldehyde may function as the primary factor in alcoholic cardiomyopathy by disrupting Ca(2+) handling or disturbing cardiac excitation-contraction coupling. By producing reactive oxygen species, acetaldehyde shifts the intracellular redox potential from a reduced state to an oxidized state. We examined whether the redox state modulates acetaldehyde-induced Ca(2+) handling by measuring Ca(2+) transient using a confocal imaging system and single ryanodine receptor type 2 (RyR2) channel activity using the planar lipid bilayer method. The structure and function of band 3 (AE1): recent developments (review).

This review discusses recent advances in our understanding of the structure, function and molecular genetics of the membrane domain of red cell anion exchanger, band 3 (AE1), and its role in red cell and kidney disease. A new model for the topology of band 3 has been proposed, which suggests the membrane domain has 12 membrane spans, rather than the 14 membrane spans of earlier models. The major difference between the models is in the topology of the region on the C-terminal side of membrane spans 1-7. Two dimensional crystals of the deglycosylated membrane domain of band 3 have yielded two an

Galactose-functionalized engineered outer membrane vesicles for targeted delivery of enterocin Gr17 ameliorate Salmonella Typhimurium-induced colitis.

Bacteriocins, antimicrobial peptides from lactic acid bacteria (LAB), represent promising agents against Salmonella Typhimurium-induced colitis. However, their proteinaceous nature leads to poor bioavailability and insufficient concentrations at infection sites, which significantly limits their application potential. Enterocin Gr17 exhibits potent antibacterial activity against S. typhimurium and effectively ameliorates pathogen-induced intestinal barrier damage. Herein, we innovatively developed a targeted oral delivery system termed OMV-Gr17/Gal, based on galactose (Gal)-modified engineered

A bibliometric systematic review of extracellular vesicles in cutaneous malignant melanoma from 2005 to 2025.

INTRODUCTION: Cutaneous melanoma (CM) is among the most aggressive human malignancies characterised by its strong propensity for metastasis. Recent advances in melanoma research have reframed the disease as a systemic condition driven by dynamic interactions between tumour cells, the tumour microenvironment and the immune system. Increasing evidence indicates that these interactions are largely mediated by extracellular vesicles (EVs), small lipid-bilayer-enclosed particles that facilitate intercellular communication. EVs can be isolated from all biofluids, making them attractive minimally inv

A novel scalable method for the production of rennet-treated milk-derived extracellular vesicles for improved curcumin oral delivery.

BACKGROUND: Extracellular vesicles (EVs) are lipid bilayer-enclosed nanoparticles secreted by all cell types and are gaining increasing attention as natural, biocompatible drug delivery nanovehicles. Among the various sources, bovine milk-derived EVs (MEVs) represent an abundant, cost-effective, and eco-friendly alternative, with demonstrated potential in delivering chemotherapeutics, bioactive natural compounds, miRNAs, and other therapeutic agents. However, the effective and scalable production of MEVs still requires protocol optimization for contaminant removal, especially casein micelles a

Lactoferrin-modified organic-inorganic hybrid mesoporous silica for co-delivery of levodopa and curcumin in the synergistic treatment of Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) is a chronic neurodegenerative disorder primarily characterized by oxidative stress and dopaminergic neuron damage. While levodopa remains the cornerstone of PD treatment, its efficacy is limited by poor bioavailability and neuroprotective effects. Curcumin, a potent antioxidant derived from turmeric, demonstrates neuroprotective promise but also suffers from low bioavailability, hindering its therapeutic application. The combined therapeutic use of levodopa and curcumin offers a potential synergistic approach, though its neuroprotection potential through b

A life in light - in honor of David Mauzerall on his 95th birthday.

David Mauzerall was born on July 22, 1929 to a working-class family in the small, inland textile town of Sanford, Maine. Those humble origins instilled a lifelong frugality and an innovative spirit. After earning his PhD degree in 1954 in physical organic chemistry with Frank Westheimer at the University of Chicago, he joined The Rockefeller Institute for Medical Research (now University) as a postdoctoral fellow that summer, rose to the rank of professor, and remained there for the rest of his career. His work over more than 60 years encompassed porphyrin biosynthesis, photoinduced electron-t
