BACKGROUND: Cystic fibrosis-related diabetes (CFRD) is one of the most common extrapulmonary complications of cystic fibrosis (CF), characterized by abnormal glucose tolerance due primarily to insulin insufficiency. CF-related prediabetes (CFRpD) represents an asymptomatic stage before CFRD onset, requiring routine screening for detection. We aimed to characterize the metabolic profiles underlying abnormal glucose tolerance in people living with CF (pwCF) and identify disrupted pathways associated with CFRD progression.
METHODS: Fasting plasma samples from pwCF with normal glucose tolerance (

Global warming induced heat stress severely impairs porcine oocyte quality due to their high lipid content, disrupting reproductive performance. This study combined transcriptomic and metabolomic analyses of heat-stressed porcine oocytes, identifying glutathione metabolism, arginine and proline metabolism, and purine biosynthesis as key perturbed pathways. We focused on arginine due to the significant depletion of arginine and its downstream metabolites in heat stressed oocytes. Exogenous arginine supplementation was applied to heat-stressed immature oocytes. Results showed arginine protects p

Diquat is a widely used bipyridyl herbicide, and central nervous system injury (CNSI) is a major cause of death in acute poisoning; early serum markers and underlying mechanisms remain unclear. We prospectively classified acutely poisoned patients into healthy controls (JKDZ), diquat patients without CNSI (NCNSI), patients who developed CNSI during hospitalization (CNSI1), and patients presenting with CNSI (CNSI2). Early serum underwent transcriptomic, proteomic and metabolomic profiling. Multivariate and differential analyses with KEGG/GO enrichment and O2PLS integration identified key pathwa

Soil salinization due to anthropogenic activity and climate change is a bottleneck to the global agricultural yield and food security. Conventional approaches for mitigating salt stress, including utilization of chemical fertilizers, have shown limited success, and the use of genetically engineered microbes as bioinoculants or salt-tolerant crop varieties against abiotic salinity stress often faces regulatory challenges with extended timelines. Contrary to the existing approaches, employment of halophilic bacteria and archaea offers a promising platform for eco-friendly and sustainable crop cu

BACKGROUND: Atherosclerosis (AS) is a chronic inflammatory vascular disease characterized by immune dysregulation, metabolic disturbance, and progressive vascular remodeling. Although anoikis resistance has been extensively investigated in cancer biology, its involvement in immune-cell persistence and plaque progression in AS remains poorly understood.
METHODS: We integrated two publicly available bulk transcriptomic datasets (GSE100927 and GSE28829) to identify differentially expressed genes (DEGs) and performed weighted gene co-expression network analysis (WGCNA) to determine disease-associ

BACKGROUND: Idiopathic male infertility (IMI) accounts for 30%-40% of male infertility cases, yet the molecular networks associated with seminal plasma dysfunction and non-invasive biomarkers remain largely unclear. This study aimed to preliminarily explore the transcriptomic and metabolomic alterations in IMI seminal plasma through cross-omics integration, and to screen for exploratory candidate discriminators among the metabolites.
METHODS: A total of 30 IMI patients and 30 normozoospermic controls were recruited for untargeted UHPLC-MS/MS metabolomics of seminal plasma. Candidate different

Osteosarcoma remains a therapeutically challenging malignancy with durable responses limited by frequent treatment resistance. Although immune activity is detectable in many tumors, immunotherapy and combination strategies show limited clinical benefit, indicating immune dysfunction extends beyond tumor-intrinsic mechanisms. Growing evidence identifies immune exclusion as a key immunological barrier in osteosarcoma, characterized by spatial and functional segregation of immune cells from malignant areas despite their presence in the tumor microenvironment. Here, we use immune exclusion as a wo

Global climate change and shrinking freshwater supplies threaten agricultural sustainability. A major translational bottleneck exists: drought-tolerant genotypes developed in labs often fail in open-field conditions due to phenotypic mismatch. Traditional single-gene approaches and static lab tests overlook genotype-by-environment (G × E) interactions, hormonal tradeoffs (involving Abscisic Acid, Jasmonic Acid, Brassinosteroids, and Melatonin), and artificial effects of pots. This review presents an integrated quantitative selection framework to bridge lab and field results. In this review, we

Bisphenol A (BPA) exposure is associated with gestational diabetes mellitus (GDM); however, the underlying molecular mechanisms remain elusive. This study aimed to identify key genes mediating BPA exposure-related GDM through integrative bioinformatics analysis. We integrated BPA-related targets from multiple databases with GDM-associated genes derived from GeneCards and the GSE103552 dataset. Functional enrichment analysis was performed on the intersecting genes. A protein-protein interaction (PPI) network was constructed, and hub genes were identified using four topological algorithms. Endometriosis is a prevalent chronic inflammatory gynecological disorder affecting approximately 10% of reproductive-age women worldwide, characterized by endometrial-like tissue outside the uterine cavity. Ectopic lesion growth tracks closely with immune-inflammatory dysregulation-altered macrophage polarization, impaired natural killer (NK) cytotoxicity, skewed T cell subsets, B cell-related autoimmunity, tolerogenic dendritic cells, mast cell-associated neuroinflammation, and abnormal cytokine networks. Even after many years of study, several regulatory mechanisms in the endometriotic micro

Amyloid-related imaging abnormalities (ARIA) are the principal safety concern limiting anti-amyloid therapies for Alzheimer's disease, yet their biology remains unclear. Here we show, through multi-omic profiling of peripheral blood from three ARIA+ patients and matched controls, that ARIA is associated with coordinated reprogramming of CD8 + T cells. CD8+ effector memory (TEM) and terminally differentiated (TEMRA) subsets were expanded, clonally enriched, and transcriptionally primed for cytotoxicity and vascular trafficking. Transcription factor inference and metabolomics converged on glycol

The pathophysiology of acute pancreatitis (AP), a common clinical emergency, is poorly understood. Previous studies have implicated N6-methyladenosine (m6A) modification in the pathogenesis of AP; however, the precise molecular mechanisms remain unclear. Whether metabolites participate in this process is also an important, unresolved question. To investigate the possible causal pathways, we integrated multi-omics data. We identified m6A related genes from literature and combined these genes with genetic summary statistics of AP, expression quantitative trait loci (eQTL) data, and information o

BACKGROUND: Gallbladder cancer (GBC) is a highly aggressive malignancy with a dismal prognosis, frequently diagnosed at advanced stages. While cholelithiasis is a primary risk factor, the role of the biliary microbiome and its metabolic products in driving carcinogenesis is increasingly recognized. This review synthesizes multi-omics data to elucidate the interplay between microbial dysbiosis and metabolomic shifts in GBC.
METHODS: A systematic literature search was conducted on PubMed (up to January 2026) focusing on biliary bacteria, the gut-bile axis, and multi-omics markers. A narrative s

Osteoarthritis (OA) is a joint disease of unknown etiology that can occur in joints throughout the body, especially with articular cartilage lesions of the knee joint and hip joint. Osteoarthritis not only causes joint pain, deformity, and functional impairment, but also significantly increases the risk of cardiovascular events, deep vein thrombosis in the lower limbs, and hip fractures. Up to now, there have been no specific medications available for the treatment of osteoarthritis, and the disease remains incurable. Therefore, identifying reliable biomarkers is crucial for achieving a more a

BACKGROUND: The piriform cortex (PIR) is a key region of the olfactory system and is increasingly implicated in Alzheimer's disease (AD). However, the cellular heterogeneity of principal neurons (PN) in this region and their selective vulnerability in AD remain poorly understood.
METHODS: Here, we integrated single-cell multi-omics datasets to systematically characterize PIR PN heterogeneity. We identified four transcriptionally and epigenetically distinct PN subtypes with different spatial distributions across the anterior and posterior PIR.
RESULTS: In AD mouse models, the abundance of the

The molecular determinants of phenotypic severity in red cell enzymopathies are often obscured by the disconnect between coding sequence variants and their regulatory landscapes. Here we present a single-patient molecular case study that uses an integrative multi-omic approach-combining short-read WGS, PacBio HiFi long-read sequencing, native CpG methylation profiling, and Iso-Seq full-length transcriptomics-to characterize a severe, transfusion-dependent hemolytic anaemia. We identified a compound heterozygous state in the glucose-6-phosphate isomerase (GPI) gene, with no wild-type allele pre
