VGF expression in the brain.

VGF is a neuronal polypeptide first identified as a cDNA clone in a gene library from nerve growth factor-stimulated PC12 cells. In the present paper, the expression of VGF is examined for the first time throughout the adult rat central nervous system with immunocytochemistry and Northern blot analysis. VGF RNA was found in all brain regions studied, including hypothalamus, hippocampus, cerebellum, olfactory bulb, and cortex. In contrast to the relatively strong immunostaining of hypothalamic neurons, the level of VGF RNA expression in the hypothalamus was relatively low in comparison with oth…

Combined olfactory bulbectomy and zinc deficiency induce complex neurobiological alterations resistant to classical antidepressant treatments: Insights from proteomic and neurochemical analyses.

Major depression (MDD) is a complex and multifactorial mental illness. Despite advances in treatment, about 30% of patients still do not see improvement, underscoring the need for new insights into its pathophysiology. Clinical studies suggest that zinc deficiency may be an essential risk factor for the development of depressive disorder and may also contribute to reduced antidepressant efficacy. We developed a novel animal model combining olfactory bulbectomy (OB) with dietary zinc deficiency (ZnD; 3 mg Zn/kg) to further explore this relationship. After three weeks of OB + ZnD treatment, rats…

Spinal cord injury alters the therapeutic potential of olfactory ensheathing cells harvested in an autologous transplantation model.

Spinal cord injury (SCI) leads to irreversible loss of motor, sensory, and autonomic functions and currently has no cure. While cell therapies in animal models have shown benefits such as scar reduction and neuroprotection, their translation to humans remains limited, partly because clinical applications rely on autologous rather than healthy donor cells. This prompted us to investigate how the lesion context alters the neuro-reparative potential of olfactory ensheathing cells (OECs). OECs were cultured from healthy animals or from animals 1 week after SCI, and both populations were transplant

Transplantation of olfactory ensheathing cells embedded in the GelMA-F127DA hydrogel promotes the repair of spinal cord injury.

Spinal cord injury (SCI) is a highly disabling condition. Olfactory ensheathing cells (OECs) are important transplantable cells for treating spinal cord injury (SCI). However, directly transplanting cells into the injured area may result in the loss of transplanted cells. Additionally, secondary inflammatory responses reduce the survival of transplanted cells. In this study, a hydrogel with favorable properties, including plasticity, strong adhesion, mechanical stability, shear-thinning behavior, and pore size suitable for OEC growth and axon extension, was fabricated from methacryloylated gel

Olfactory ensheathing cell transplantation alleviates cancer pain by inhibiting P2X7 receptor expression-mediated activation of microglia.

Peripheral nerve invasion can occur during cancer progress, which seriously affects the patient's quality of life and brings pain. There is currently no effective treatment method. Therefore, in this study, we established a model of breast cancer bone metastasis and transplanted olfactory sheath cells (OECs) into rats to explore a new cell therapy strategy for treating cancer pain. The results showed that compared with the cancer pain group and the vehicle group (DMEM/F12 medium solution), OEC transplantation improved tibial bone destruction and osteolytic lesions, increased the mechanical wit

Chitosan biomaterial enhances the effect of OECs on the inhibition of sciatic nerve injury-induced neuropathic pain.

Neuropathic pain is a common symptom experienced by most clinical diseases at different levels, and its treatment has always been a clinical difficulty. Therefore, it is particularly important to explore new and effective treatment methods. The role of olfactory ensheathing cells (OECs) in nerve injury and pain is recognized by different studies. Our previous study found that transplantation of OECs alleviated hyperalgesia in rats. However, single-cell transplantation lacks medium adhesion and support, and exerts limited analgesic effect. Therefore, on the basis of the previous study, this stu

Olfactory Dysfunction in the COVID-19 Outbreak.

The first cases of coronavirus 2019 disease (COVID-19) occurred in Wuhan, China, and the disease rapidly become a public health emergency of international proportions. COVID-19 can cause mild-to-severe acute respiratory syndrome (SARS) and is caused by the SARS-CoV-2 coronavirus. The clinical manifestations of COVID-19 include fever, dry cough, fatigue, sputum production, shortness of breath, sore throat, and headache. We performed this narrative review to analyze the current literature on postviral olfactory dysfunction related to the SARSCoV- 2 pandemic. Since the initial anecdotal reports f

Pharmacoimaging of Blood-Brain Barrier Permeable (FDG) and Impermeable (FLT) Substrates After Intranasal (IN) Administration.

To illustrate the use of imaging to quantify the transfer of materials from the nasal cavity to other anatomical compartments, specifically, transfer to the brain using the thymidine analogue, [18F]fluorothymidine (FLT), and the glucose analogue, [18F]fluorodeoxyglucose (FDG). Anesthetized rats were administered FLT or FDG by intranasal instillation (IN) or tail-vein injection (IV). PET/CT imaging was performed for up to 60 min. Volumes-of-interest (VOIs) for the olfactory bulb (OB) and the remaining brain were created on the CT and transferred to the co-registered dynamic PET. Time-activity c…

Olfactory bulbectomy-induced changes in phospholipids and protein profiles in the hippocampus and prefrontal cortex of rats. A preliminary study using a FTIR spectroscopy.

BACKGROUND: Dysfunctions in neuronal proteins can lead to development of depression. Lipids determine the localization and function of proteins in the cell membrane and thereby regulate synaptic throughput in neurons. The aim of study was to examine changes in the phospholipid-protein balance in the olfactory bulbectomy (OB) model of depression in rats using a Fourier transform infra-red (FTIR) spectroscopy. METHODS: Sprague Dawley rats were subjected to the OB procedure and treated with amitriptyline (AMI, 10mg/kg). The FTIR measurements were performed in the hippocampus and prefrontal corte…

Transplantation of olfactory ensheathing cells promotes axonal regeneration and functional recovery of peripheral nerve lesion in rats.

INTRODUCTION: Olfactory ensheathing cells (OECs) hold promise for cell therapy because they may promote regeneration of the central nervous system. However, OECs have been less studied after peripheral nerve injury (PNI). The purpose of this investigation was to determine the effect of OEC transplantation on a severe sciatic nerve (SN) lesion. METHODS: OECs were injected in rats after section and 2-cm resection of the SN. RESULTS: Three months after therapy, muscle strength and morphometric studies showed complete restoration of the contractile properties of the gastrocnemius and complete re

In vivo MRI of olfactory ensheathing cell grafts and regenerating axons in transplant mediated repair of the adult rat optic nerve.

The purpose of the present study was to use magnetic resonance imaging (MRI) as a tool for monitoring transplant-mediated repair of the adult rat visual pathway. We labelled rat olfactory ensheathing cells (OECs) using micron-sized particles of iron oxide (MPIO) and transplanted them by: i) intravitreal injection (ivit) and ii) intra-optic nerve (ON) injection (iON) in adult rats with ON crush (ONC) injury. We applied T(2)-weighted MRI and manganese-enhanced MRI (MEMRI) to visualise transplanted cells and ON axons at specific times after injury and cell engraftment. Our findings demonstrate th

Further observations on subacute sclerosing encephalitis in adult hamsters: the effects of intranasal infections with Langat virus, measles virus and SSPE-measles virus.

Passage by i.c. inoculations of suckling hamsters enhanced the virulence for adult hamsters of Langat virus (TP21), neurotropic strain of measles virus (HNT) and SSPE-measles virus (HBS), not only for i.c. infections but also for intranasal instillations. The various viral strains passaged in hamsters showed a great similarity of behaviour including the ability of producing in a proportion of apparently unaffected survivors a subacute sclerosing encephalitis, leading to atrophy of parts of the brain especially the rhinencephalon. When large groups of animals were used for transmission experime

The olfactory bulbectomised rat as a model of depression.

Bilateral olfactory bulbectomy results in changes in behavior, and in the endocrine, immune and neurotransmitter systems, that simulates many of those seen in patients with major depression. The olfactory system in the rat forms a part of the limbic region in which the amygdala and hippocampus contribute to the emotional and memory components of behavior. However, the loss of olfaction alone, which results from bulbectomy, is not the major factor that contributes to the behavioral abnormalities as peripherally induced anosmia does not cause the same behavioral changes. Thus it would appear tha…
