Breath Analysis for Early Detection of Malignant Pleural Mesothelioma: Volatile Organic Compounds (VOCs) Determination and Possible Biochemical Pathways.

Malignant pleural mesothelioma (MPM) is a rare neoplasm, mainly caused by asbestos exposure, with a high mortality rate. The management of patients with MPM is controversial due to a long latency period between exposure and diagnosis and because of non-specific symptoms generally appearing at advanced stage of the disease. Breath analysis, aimed at the identification of diagnostic Volatile Organic Compounds (VOCs) pattern in exhaled breath, is believed to improve early detection of MPM. Therefore, in this study, breath samples from 14 MPM patients and 20 healthy controls (HC) were collected an

Development and External Validation of a Machine Learning-Derived Risk Stratification Score for Metastatic Castration-Resistant Prostate Cancer.

PURPOSE: Metastatic castration-resistant prostate cancer (mCRPC) shows marked clinical heterogeneity and poor long-term survival, highlighting the need for simple and reliable risk stratification tools beyond the limited usability of existing complex prognostic models. In this study, we developed and validated PROGRESS (Prostate Cancer Global Risk Evaluation and Stratification Score), a simplified prognostic score derived through machine learning‑guided feature selection distilled into a simple laboratory-based tool. METHODS: Baseline data from 2,035 patients with mCRPC (training set) were em

Prediction of cognitive impairment in Parkinson's disease using TabPFN: an integrative analysis of clinical data, longitudinal trajectories, and lipidomics.

BACKGROUND: Cognitive impairment is a common and disabling non-motor manifestation of Parkinson's disease (PD). Early identification of patients at high risk remains challenging, and the metabolic mechanisms underlying cognitive decline are not fully understood. METHODS: We conducted a retrospective cohort study including PD patients who were cognitively normal at baseline. A machine learning model based on a Tabular Prior-data Fitted Network (TabPFN) was developed using routinely collected clinical and laboratory data to predict future cognitive impairment. Model performance was evaluated us

Multidimensional feature fusion in longitudinal physical examination data: a machine learning framework for classification of type 2 diabetes.

BACKGROUND: Machine learning-based risk classification of type 2 diabetes mellitus (T2DM) has become a prevailing research direction in intelligent healthcare. Nevertheless, current studies suffer from two key limitations. First, most existing models are built on cross-sectional data and cannot capture the dynamic progression characteristics of T2DM. Second, traditional longitudinal time-series models are commonly adopted without sufficiently exploring the deep interactive relationships among multi-temporal physical examination features, limiting model predictive performance. METHODS: To tack

The relationship between protein modified folding molecular network and Alzheimer's disease pathogenesis based on BAG2-HSC70-STUB1-MAPT expression patterns analysis.

BACKGROUND: Alzheimer's disease (AD) is the most common cause of dementia and cognitive decline, while its pathological mechanism remains unclear. Tauopathies is one of the most widely accepted hypotheses. In this study, the molecular network was established and the expression pattern of the core gene was analyzed, confirming that the dysfunction of protein folding and degradation is one of the critical factors for AD. METHODS: This study analyzed 9 normal people and 22 AD patients' microarray data obtained from GSE1297 in Gene Expression Omnibus (GEO) database. The matrix decomposition analy

An evaluation of existing text de-identification tools for use with patient progress notes from Australian general practice.

INTRODUCTION: Digitized patient progress notes from general practice represent a significant resource for clinical and public health research but cannot feasibly and ethically be used for these purposes without automated de-identification. Internationally, several open-source natural language processing tools have been developed, however, given wide variations in clinical documentation practices, these cannot be utilized without appropriate review. We evaluated the performance of four de-identification tools and assessed their suitability for customization to Australian general practice progre

Clinical Decision Support Using Speech Signal Analysis: Systematic Scoping Review of Neurological Disorders.

BACKGROUND: Digital biomarkers are increasingly used in clinical decision support for various health conditions. Speech features as digital biomarkers can offer insights into underlying physiological processes due to the complexity of speech production. This process involves respiration, phonation, articulation, and resonance, all of which rely on specific motor systems for the preparation and execution of speech. Deficits in any of these systems can cause changes in speech signal patterns. Increasing efforts are being made to develop speech-based clinical decision support systems. OBJECTIVE:

Deciphering Shared Gene Signatures and Immune Infiltration Characteristics Between Gestational Diabetes Mellitus and Preeclampsia by Integrated Bioinformatics Analysis and Machine Learning.

Gestational diabetes mellitus (GDM) and preeclampsia (PE) are common and serious disorders of pregnancy that threaten maternal safety and perinatal outcomes. Generally, GDM is recognized as an independent risk factor for the development of preeclampsia, while a history of preeclampsia in primiparous women is also a risk factor for GDM in subsequent pregnancies. However, the intricate underlying mechanisms of GDM and PE remain elusive. This study developed a diagnostic prediction model for GDM and PE. It investigated the correlation between shared signature genes and immune infiltration charact

Machine Learning Interpretability to Assess the Association Between Time in Tight Range and Mortality in Cardiogenic Shock.

BACKGROUND: Cardiogenic shock (CS) is a critical condition of end-organ hypoperfusion with high mortality. Fluctuations in blood glucose (BG) levels may exacerbate cardiovascular instability in critically ill patients. Time In Tight Range (TITR), defined as the percentage of time in the target BG range of 3.9-7.8 mmol/L (70-140 mg/dL), has become an increasingly important index of glycaemic status, but its impact on mortality in CS remains unclear. Interpretable machine learning (ML) models provide transparent, quantitative and visual insights into the prognostic importance of TITR, clarifying

The novel diagnostic markers for systemic lupus erythematosus and periodontal disease.

BACKGROUND AND AIMS: Systemic lupus erythematosus (SLE) is one of the most prevalent systemic autoimmune diseases, characterized by aberrant activation of the immune system that leads to diverse clinical symptoms; periodontal disease (PD) is an inflammatory oral disorder caused by immune-mediated damage against subgingival microflora. Although clinical evidence suggests a potential association between SLE and PD, their shared pathogenic mechanisms remain unclear. This study aims to explore common genetic markers in SLE and PD that hold diagnostic and therapeutic implications. METHODS: Microar

Patient-Related Metadata Reported in Sequencing Studies of SARS-CoV-2: Protocol for a Scoping Review and Bibliometric Analysis.

BACKGROUND: There has been an unprecedented effort to sequence the SARS-CoV-2 virus and examine its molecular evolution. This has been facilitated by the availability of publicly accessible databases, such as the GISAID (Global Initiative on Sharing All Influenza Data) and GenBank, which collectively hold millions of SARS-CoV-2 sequence records. Genomic epidemiology, however, seeks to go beyond phylogenetic (the study of evolutionary relationships among biological entities) analysis by linking genetic information to patient characteristics and disease outcomes, enabling a comprehensive underst

Fcer1g and St3gal1: Macrophage-associated angiogenesis biomarkers and therapeutic targets in sepsis-induced acute lung injury.

BACKGROUND: Acute lung injury (ALI) involves the release of growth factors and inflammatory mediators from damaged pulmonary tissues, fostering endothelial cell proliferation, migration, and vascular lumen formation, thereby driving pathological angiogenesis. Macrophages contribute to angiogenesis and vascular homeostasis, but their dysregulation in pathological states worsens vascular dysfunction. This study aims to identify macrophage-associated angiogenesis-related genes as novel diagnostic biomarkers and therapeutic targets for sepsis-associated ALI (SALI). METHODS: Transcriptomic dataset

Towards multicenter β-amyloid PET imaging in mouse models: A triple scanner head-to-head comparison.

AIM: β-amyloid (Aβ) small animal PET facilitates quantification of fibrillar amyloidosis in Alzheimer's disease (AD) mouse models. Thus, the methodology is receiving growing interest as a monitoring tool in preclinical drug trials. In this regard, harmonization of data from different scanners at multiple sites would allow the establishment large collaborative cohorts and may facilitate efficacy comparison of different treatments. Therefore, we objected to determine the level of agreement of Aβ-PET quantification by a head-to-head comparison of three different state-of-the-art small animal PET
