Development of a Trivalent Construct Omp18/AhpC/FlgH Multi Epitope Peptide Vaccine Against Campylobacter jejuni.

Campylobacter jejuni (C. jejuni) is one of the major pathogens contributing to the enteritis in humans. Infection can lead to numerous complications, including but not limited to Guillain-Barre syndrome, reactive arthritis, and Reiter's syndrome. Over the past two decades, joint efforts have been made toward developing a proper strategy of limiting the transmission of C. jejuni to humans. Nevertheless, except for biosecurity measures, no available vaccine has been developed so far. Judging from the research findings, Omp18, AhpC outer membrane protein, and FlgH flagellin subunits of C. jejuni

PIK-24 Inhibits RSV-Induced Syncytium Formation via Direct Interaction with the p85α Subunit of PI3K.

Phosphoinositide-3 kinase (PI3K) signaling regulates many cellular processes, including cell survival, differentiation, proliferation, cytoskeleton reorganization, and apoptosis. The actin cytoskeleton regulated by PI3K signaling plays an important role in plasma membrane rearrangement. Currently, it is known that respiratory syncytial virus (RSV) infection requires PI3K signaling. However, the regulatory pattern or corresponding molecular mechanism of PI3K signaling on cell-to-cell fusion during syncytium formation remains unclear. This study synthesized a novel PI3K inhibitor PIK-24 designed

Comparative Antibody Responses to the Live-Attenuated and Recombinant Herpes Zoster Vaccines.

Two herpes zoster (HZ) vaccines licensed in the United States are recommended by the Advisory Committee on Immunization Practices (ACIP): (i) live-attenuated vaccine (ZVL) using vOka strain varicella-zoster virus (VZV) and (ii) recombinant adjuvanted vaccine (RZV) containing recombinant varicella-zoster virus (VZV) glycoprotein E (gE). Two phase 3 clinical trials of RZV led the Advisory Committee on Immunization Practices (ACIP) to recommend it with preferred status. VZV T cell-mediated immunity (CMI), but not humoral immunity, is considered essential for protection against HZ. Published studi

Immunoproteomics and reverse vaccinology identify a protective T-cell epitope from Fonsecaea pedrosoi for experimental chromoblastomycosis.

Chromoblastomycosis is a chronic neglected tropical mycosis caused predominantly by the melanized fungus Fonsecaea pedrosoi. Current treatments are prolonged and frequently ineffective, highlighting the need for new therapeutic strategies. Here, we applied an integrated immunoproteomic and reverse vaccinology approach to identify immunogenic antigens from F. pedrosoi and evaluate their vaccine potential. Fungal proteins were separated by two-dimensional electrophoresis and immunoreactive proteins were detected using sera from infected mice. Mass spectrometry identified several antigenic protei

Integrated Computational and In Vivo Assessment of a Novel Multi-Epitope Vaccine Against Dengue Virus Serotype 4 Capsid Protein.

Dengue Virus Serotype 4 (DENV4) continues to contribute substantially to global dengue morbidity, yet current tetravalent vaccines provide inconsistent protection and may pose risks of antibody-dependent enhancement (ADE). This study aimed to design and evaluate a serotype-specific multi-epitope vaccine (MEV-DV4) targeting the conserved capsid protein of DENV4 using integrated reverse vaccinology and immunoinformatics approaches. Conserved, antigenic, non-allergenic, and non-toxic B-cell, cytotoxic T lymphocyte (CTL), and helper T lymphocyte (HTL) epitopes were identified and assembled with a

Evaluation of the surface lipoprotein assembly modifier (Slam) as a vaccine candidate against Neisseria gonorrhoeae infection.

The surface lipoprotein assembly modifier (Slam) plays an essential role in the biogenesis and assembly of outer membrane components in Gram-negative bacteria. This study evaluated the potential of recombinant NG Slam as a vaccine candidate. Bioinformatics confirmed its high conservation (including with its Neisseria meningitidis homolog) and outer membrane localization. Immunofluorescence demonstrated antibody accessibility to Slam in fixed bacterial cells. Antigenic epitope mapping revealed that dominant B- and T-cell epitopes are primarily within α-helical and random coil regions. Molecular

Exploring Leptospiral proteomes to identify potential candidates for vaccine design against Leptospirosis using an immunoinformatics approach.

Leptospirosis is the most widespread zoonotic disease, estimated to cause severe infection in more than one million people each year, particularly in developing countries of tropical areas. Several factors such as variable and nonspecific clinical manifestation, existence of large number of serovars and asymptomatic hosts spreading infection, poor sanitation and lack of an effective vaccine make prophylaxis difficult. Consequently, there is an urgent need to develop an effective vaccine to halt its spread all over the world. In this study, an immunoinformatics approach was employed to identify

A broad-spectrum SARS-CoV-2 RBD vaccine with selected high-impact mutations and novel adjuvant induces durable T cell response and broad protection in mice.

INTRODUCTION: The emergence of new SARS-CoV-2 variants with immune evasion capabilities underscores the importance of developing a broad-spectrum and effective vaccine. The receptor binding domain (RBD) of the Spike protein has been widely utilized in vaccine due to its high immunogenicity. However, the Spike protein, particularly the RBD region, exhibits significant variability in the evolution of SARS-CoV-2, leading to viral immune evasion and reduced vaccine effectiveness. METHODS: A broad-spectrum antigen (M5-RBD) was developed via mutation patching, incorporating key high-impact mutation

The global distribution and diversity of protein vaccine candidate antigens in the highly virulent Streptococcus pnuemoniae serotype 1.

Serotype 1 is one of the most common causes of pneumococcal disease worldwide. Pneumococcal protein vaccines are currently being developed as an alternate intervention strategy to pneumococcal conjugate vaccines. Pre-requisites for an efficacious pneumococcal protein vaccine are universal presence and minimal variation of the target antigen in the pneumococcal population, and the capability to induce a robust human immune response. We used in silico analysis to assess the prevalence of seven protein vaccine candidates (CbpA, PcpA, PhtD, PspA, SP0148, SP1912, SP2108) among 445 serotype 1 pneumo

D614G Mutation Alters SARS-CoV-2 Spike Conformation and Enhances Protease Cleavage at the S1/S2 Junction.

The severe acute respiratory coronavirus 2 (SARS-CoV-2) spike (S) protein is the target of vaccine design efforts to end the coronavirus disease 2019 (COVID-19) pandemic. Despite a low mutation rate, isolates with the D614G substitution in the S protein appeared early during the pandemic and are now the dominant form worldwide. Here, we explore S conformational changes and the effects of the D614G mutation on a soluble S ectodomain construct. Cryoelectron microscopy (cryo-EM) structures reveal altered receptor binding domain (RBD) disposition; antigenicity and proteolysis experiments reveal st

A bivalent inactivated influenza vaccine incorporating epitope-optimized surface proteins confers cross-protective immunity against H9N2 influenza virus.

The H9N2 avian influenza virus (AIV) has caused substantial economic losses to the global poultry industry and poses a zoonotic threat to humans. Vaccination constitutes a pivotal strategy for the prevention and control of H9N2 AIVs. However, the ongoing antigenic evolution of the viruses pose a persistent challenge to the protective efficacy of existing vaccines. Therefore, the development of a broadly protective H9N2 influenza vaccine capable of eliciting cross-reactive immune responses is crucial for mitigating both the disease burden and the risk of pandemics. Here, we developed a bivalent

Revolutionizing leishmaniasis control: leveraging immunoinformatics for precision-driven in-silico vaccine design.

Leishmaniasis is an endemic disease in many countries that affects vulnerable populations of humans, dogs, and cats. This study employs immunoinformatics to design a multi-epitope vaccine construct for leishmaniasis by identifying vaccine targets on the protein Pteridine Reductase. T-cell and B-cell epitopes were screened for antigenicity, toxicity, and allergenicity. Shortlisted T-cell targets were confirmed to have appropriate IC values (≤ 50 nM and ≤ 500 nM for MHC I and MHC II, respectively). One B-cell epitope, six MHC class I epitopes, and 25 MHC class II epitopes were selected for inclu

Circular mRNA-LNP vaccine encoding self-assembled E2-TMD-mi3 nanoparticles licit enhanced CSFV-specific immunity over commercial subunit vaccine.

The E2 subunit vaccine is crucial for eliminating Classical Swine Fever Virus (CSFV) due to its favorable biosafety and Differentiating Infected from Vaccinated Animals (DIVA) capability. However, low immunogenicity and high costs limit its broader application. To overcome these bottlenecks, we leveraged mRNA-LNP technology to design next-generation E2 glycoprotein vaccines with enhanced immunogenicity and cost-effectiveness. We designed different E2 glycoprotein coding sequences incorporating CD154 adjuvants and mi3 self-assembled nanoparticles, delivered via cmRNA-LNP formulations in murine

Rational design and in silico characterization of a multiepitope mRNA vaccine candidate against human metapneumovirus (hMPV) using reverse vaccinology and immunoinformatics approaches.

Emerging groups/subgroups of hMPV, a respiratory pathogen, pose a public health concern. Despite its global prevalence and recent outbreaks, no vaccines or targeted therapies exist. Using an immunoinformatics approach, chimeric messenger RNA (mRNA) vaccines targeting the major structural proteins of hMPV, including the glycoprotein (G), fusion protein (F), small hydrophobic protein (SH), and matrix protein (M), was aimed to be developed. The target protein sequences, formatted in FASTA, were sourced from the NCBI Virus Resource, followed by redundancy removal using CD-Hit. Epitope prediction f

A novel TamA + LtgC bivalent vaccine induces potent immunogenic and bactericidal antibodies against Neisseria gonorrhoeae.

Development of an effective vaccine against Neisseria gonorrhoeae comes with unique challenges. After five decades of research, the lack of an FDA-approved vaccine underscores the need for novel immunization strategies. Therefore, in this study, we evaluated a bivalent vaccine compared to two single-antigen formulations containing LtgC or TamA. The tamA and ltgC genes were cloned, expressed and purified. Serum IgG levels against recombinant proteins in gonorrhea patients were assessed by the enzyme-linked immunosorbent assay (ELISA). Mice were immunized with TamA and/or LtgC formulations along

A porcine reproductive and respiratory syndrome virus vaccine candidate based on PRRSV glycoprotein 5 and the Toll-like receptor 5 agonist Salmonella typhimurium flagellin.

Glycoprotein 5 (GP5) from porcine reproductive and respiratory syndrome virus (PRRSV) is a key inducer of neutralizing antibodies. A truncated GP5 gene lacking the signal peptide and transmembrane sequences was amplified via an overlap PCR method and inserted into prokaryotic expression vectors, pET32a or pGEX-6p-1, to add an His or GST tag, respectively. His-tagged GP5 was induced with IPTG, verified by SDS-PAGE and Western blotting, and purified to serve as an immunogen accompanied with the Salmonella typhimurium flagellin (FliC), a Toll-like receptor 5 (TLR5) agonist. Levels of TLR5 and cyt

Efficacy of a potential trivalent vaccine based on Hc fragments of botulinum toxins A, B, and E produced in a cell-free expression system.

Botulinum toxins produced by the anaerobic bacterium Clostridium botulinum are the most potent biological toxins in nature. Traditionally, people at risk are immunized with a formaldehyde-inactivated toxin complex. Second generation vaccines are based on the recombinant carboxy-terminal heavy-chain (Hc) fragment of the neurotoxin. However, the materialization of this approach is challenging, mainly due to the high AT content of clostridial genes. Herein, we present an alternative strategy in which the native genes encoding Hc proteins of botulinum toxins A, B, and E were used to express the re

Efficacy, safety and immunogenicity of a pneumococcal protein-based vaccine co-administered with 13-valent pneumococcal conjugate vaccine against acute otitis media in young children: A phase IIb randomized study.

BACKGROUND: Native American populations experience a substantial burden of pneumococcal disease despite use of highly effective pneumococcal conjugate vaccines (PCVs). Protein-based pneumococcal vaccines may extend protection beyond the serotype-specific protection elicited by PCVs. METHODS: In this phase IIb, double-blind, controlled trial, 6-12 weeks-old Native American infants randomized 1:1, received either a protein-based pneumococcal vaccine (dPly/PhtD) containing pneumolysin toxoid (dPly, 10 µg) and pneumococcal histidine triad protein D (PhtD, 10 µg) or placebo, administered along wit

An engineered probiotic co-expressing dual melanoma epitopes for therapeutic cancer vaccination.

Therapeutic cancer vaccines face challenges in tumor targeting, adjuvant dependency, and inefficient antigen cross-presentation. We engineered a probiotic vaccine using Escherichia coli Nissle 1917 co-expressing MHC-I epitopes from melanoma antigens Pmel17 and TYRP2. The fusion protein, designed via AlphaFold3, incorporating an IgG Fc domain for stability and Listeriolysin O (LLO) for cytosolic delivery, with expression controlled by an L-arabinose-inducible promoter. The strain retained probiotic properties, was efficiently phagocytosed by antigen-presenting cells, and enhanced their maturati
