AMPK as a central regulator of necroptosis: mechanistic insights and therapeutic implications.

Adenosine monophosphate-activated protein kinase (AMPK) is an evolutionarily conserved serine/threonine kinase that links cellular energy stress with metabolic adaptation, autophagy, redox homeostasis, and cell fate decisions. Necroptosis is a regulated lytic form of cell death driven by receptor-interacting serine/threonine kinases 1 and 3 (RIPK1 and RIPK3), with mixed lineage kinase domain-like protein (MLKL) serving as the terminal executor. Increasing evidence suggests that AMPK modulates necroptosis through multiple interconnected mechanisms. AMPK directly phosphorylates RIPK1, thereby in

Relationship between vitamin D and surrogate parameters (leukocyte telomerase length, telomerase activity), and genes (ACTN3, FOXO3A, VDR, SIRT1, MSTN) for ageing in Asian Indian with prediabetes.

INTRODUCTION: Vitamin D has been implicated in cellular aging through its effects on telomere maintenance, but the extent of its influence on leukocyte telomere length (LTL), telomerase activity (TA), and interactions with aging-related genetic polymorphisms remains underexplored in Asian Indians with prediabetes. This study investigated the relationship between serum vitamin D levels, LTL, TA, and variants in aging-associated genes-ACTN3, FOXO3A, VDR, SIRT1, and MSTN. METHODS: A cross-sectional study was conducted among 290 prediabetic subjects (aged 20-60 years). Anthropometry, biochemical

Perfluorooctanoic acid (PFOA) induces apoptosis by disrupting mitochondrial function via the SIRT1/FOXO1-SOD2 pathway in human granulosa cells: Implications for PCOS.

Perfluorooctanoic acid (PFOA), a widely used perfluoroalkyl substance (PFAS), has been associated with adverse reproductive health outcomes, including polycystic ovary syndrome (PCOS). However, the molecular mechanisms remain poorly understood. In this study, we explored the effects of PFOA exposure on granulosa cell apoptosis, a key contributor to PCOS pathogenesis. Human ovarian granulosa-like tumor cell line (KGN) was exposed to PFOA (0-100 μM), resulting in a significant increase in cell apoptosis and impaired mitochondrial function, as evidenced by elevated reactive oxygen species (ROS) l

UCP2 Upregulates ACSL3 to Enhance Lipid Droplet Release from Acinar Cells and Modulates the Sirt1/Smad3 Pathway to Promote Macrophage-to-Myofibroblast Transition in Chronic Pancreatitis.

Chronic pancreatitis (CP) is a progressive inflammatory disease characterized by pancreatic fibrosis and functional decline. Here, we identify macrophage-to-myofibroblast transition (MMT) as a novel feature of CP and investigate the role of mitochondrial uncoupling protein 2 (UCP2) in this process. Using mouse models, human pancreatic specimens, and cell lines, we show that UCP2 is markedly upregulated in CP, primarily in acinar cells. UCP2 knockout reduces MMT and alleviates fibrosis, whereas macrophage depletion reverses this protective effect, confirming the central role of MMT. Metabolomic

Nuciferine, a lotus leaf extract with low bioavailability, shows beneficial effects on hepatic metabolism and gut microbiota, but its anti-inflammatory mechanisms in non-alcoholic steatohepatitis (NASH) are unclear. This study aimed to clarify how nuciferine mitigates hepatic inflammation in NASH by exploring its interactions with immune pathways and gut microbiota. Initially, a NASH mouse model was induced using a methionine- and choline-deficient diet, with nuciferine administered orally. Furthermore, liver damage was assessed, and hepatic M1 (CD11B+ pro-inflammatory) and M2 (CD163+ anti-inf

System biology-based assessment of the molecular mechanism of epigallocatechin gallate in Parkinson's disease: via network pharmacology, in-silico evaluation & in-vitro studies.

Epigallocatechin gallate (EGCG) compound (IMPHY000226) has the potential to modulate multiple molecular mechanisms involved in Parkinson's disease. Multiple targets such as SIRT3, FOXO1, PRKAA1, PPARGC1A, and CREBBP directly regulate reactive oxygen species levels and oxidative stress, suggesting that targeting these genes could help prevent further cellular damage. EGCG targets were identified using Swiss target prediction, revealing 31 targets modulated by EGCG. Specific keywords were used to identify 4663 targets related to PD modulation. The network was constructed and analyzed using the n

TACC3 enhances glycolysis in bladder cancer cells through inducing acetylation of c-Myc.

The proliferation of bladder cancer (BC) cells is driven by metabolic reprogramming, marked by a glycolytic dependency to sustain uncontrolled growth. While Transforming Acidic Coiled-Coil Containing Protein 3 (TACC3) is known to promote BC progression and correlate with poor prognosis, the mechanisms underlying its upregulation and role in aerobic glycolysis remain unclear. Here, we identify E2F3 as a direct transcriptional activator of TACC3, with its amplification in BC driving elevated TACC3 expression. TACC3 overexpression enhances glycolysis, increasing glucose consumption, lactate produ

Harmine alleviated STZ-induced rat diabetic nephropathy: A potential role via regulating AMPK/Nrf2 pathway and deactivating ataxia-telangiectasia mutated (ATM) signaling.

Diabetic nephropathy (DN) is a serious kidney disorder driven by diabetes and affects people all over the world. One of the mechanisms promoting NF-κB-induced renal inflammation and injury has been theorized to be ATM signaling. On the other hand, AMPK, which can be activated by the naturally occurring alkaloid harmine (HAR), has been proposed to stop that action. As a result, the goal of this study was to evaluate the therapeutic effectiveness of HAR against streptozotocin (STZ)-induced DN in rats through AMPK-mediated inactivation of ATM pathways. Twenty male Wistar rats were grouped into 4

Sirt3 Protects Against Ischemic Stroke Injury by Regulating HIF-1α/VEGF Signaling and Blood-Brain Barrier Integrity.

Sirtuin 3 (Sirt3) is a member of the Sirtuin family proteins and known to regulate multiple physiological processes such as metabolism and aging. As stroke is an aging-related disease, in this work, we attempt to examine the role and potential mechanism of Sirt3 in regulating ischemic stroke by using a permanent middle cerebral artery occlusion (pMCAO) model in wild type (WT) and Sirt3 knockout (KO) mice, coupled with oxygen glucose deprivation (OGD) experiments in cultured primary astrocytes. Sirt3 deficiency aggravated neuronal cell apoptosis and neurological deficits after brain ischemia. I

Dexmedetomidine postconditioning suppresses myocardial ischemia/reperfusion injury by activating the SIRT1/mTOR axis.

Источник пометил публикацию как отозванную

Myocardial ischemia/reperfusion (MI/R) triggers a complicated chain of inflammatory reactions. Dexmedetomidine (Dex) has been reported to be important in myocardial disorders. We evaluated the role of Dex in MI/R injury via the silent information regulator factor 2-related enzyme 1 (SIRT1)/mammalian target of rapamycin (mTOR) signaling pathway. First, Dex was immediately injected into rat models of MI/R injury during reperfusion. After Evans Blue-triphenyl tetrazolium chloride (TTC) and Hematoxylin-Eosin (H-E) staining, MI/R injury was observed. The extracted serum and myocardial tissues were

HSF1/HSP pathway in the hippocampus is involved in SIRT1-mediated caloric restriction-induced neuroprotection after surgery in aged mice.

Postoperative cognitive dysfunction is common in the elderly. Endoplasmic reticulum stress (ER-stress) increases neuronal apoptosis after surgery, and chaperone molecules, such as heat shock proteins (HSPs), help reduce unfolded protein reactions, thereby promoting protein homeostasis. Mammal sirtuin1 (SIRT1)-mediated deacetylation of heat shock factor 1 (HSF1) upregulates HSF1 binding to the HSP70 promoter. Caloric restriction (CR) improves cognition in many neurodegenerative models. In this study, we evaluated whether CR improves impaired learning and memory after surgery by attenuating ER-s

Hepatitis C virus core protein induces dysfunction of liver sinusoidal endothelial cell by down-regulation of silent information regulator 1.

Hepatic fibrosis is a frequent feature of chronic hepatitis C virus (HCV) infection. Some evidence has suggested the potential role of silent information regulator 1 (SIRT1) in organ fibrosis. The aim of this study was to investigate the effect of HCV core protein on expression of SIRT1 of liver sinusoidal endothelial cell (LSEC) and function of LSEC. LSECs were co-cultured with HepG2 cells or HepG2 cells expressing HCV core protein and LSECs cultured alone were used as controls. After co-culture, the activity and expression levels of mRNA and protein of SIRT1 in LSEC were detected by a SIRT1
