Piperazines such as 1-benzylpiperazine (BZP), 1-(3-trifluromethylphenyl)piperazine (3-TFMPP), and 1-(3-chlorophenyl)piperazine (3-CPP) have become recent drugs of abuse. With stimulant effects comparable to amphetamines but with a lower potency and differential global scheduling status, they have been sold as a supposed legal alternative to "Ecstasy". We have synthesized BiNiX, a caged methylxanthine agonist for the ryanodine receptor (RyR) — the major calcium channel that mediates Ca2+ release from intracellular Ca2+ stores in electrically excitable cells. BiNiX is easily loaded into living cells through incubation with its acetoxymethyl (AM) ester. Delivery of focused UV light pulses to the loaded cell releases paraxanthine focally and rapidly to activate RyRs to release Ca2+, thus elevating intracellular Ca2+ concentration to initiate Ca2+-dependent signaling cascades. Intercellular communication mediated by gap junction channels plays an important role in many cellular processes. In contrast to other channels, gap junction channels span two plasma membranes resulting in an intracellular location for both ends of the junctional pore and the regulatory sites for channel gating. This configuration presents unique challenges for detailed experimental studies of junctional channel physiology and ligand-activation in situ. Orotic acid was separated from other urinary constituents by ion-pair formation with tetrabutylammonium, and isocratic elution from a reversed-phase column.
