Metabolic Profiling of Circulating and Tumour-Infiltrating T Cells Uncovers Bioenergetic Checkpoints Linked With Immune and Clinical Skewing in Melanoma Patients.

Metabolism is critical for immune cell functions. Tumours shape their microenvironment to render it metabolically hostile for infiltrating immune cells. While targeting immunometabolism emerges as a promising way to reinvigorate anticancer immunity, a deeper understanding of the metabolic disturbances of immune cells is needed. Here, we explored how the metabolic status of T cells governs immune skewing from circulating and tumour-infiltrating CD4+ and CD8+ T cells of melanoma patients at a single-cell level using the SCENITH method and targeted metabolomics. Circulating and tumour-infiltratin

Metabolic licensing at the dendritic cell-NK cells immune synapse in viral asthma exacerbations.

Asthma exacerbations are predominantly triggered by respiratory viral infections, yet current therapies largely fail to restore effective antiviral immunity. Emerging data indicate that this failure is tightly coupled to dysregulated immunometabolism within the asthmatic lung. This review advances the concept of a dendritic cell-natural killer (DC-NK) metabolic checkpoint, whereby the metabolic state of DCs, regulated by autophagy and AMPK/mTOR signaling, licenses NK cells for antiviral effector function. In type 2-high, type 2-low, and obesity-related asthma endotypes, chronic hypoxia, HIF-1α

Function of HIF-1α in Regulating Sphingolipid Metabolism and Alleviating Oxidative Stress Damage in Callosobruchus chinensis Under Hypoxia.

It has been suggested that hypoxia-inducible factor 1α (HIF1α) serves as a key regulatory factor in insect metabolism remodelling under hypoxia, in which lipid metabolism appears to play an important role. However, the underlying mechanism remains incompletely understood. In this paper, the function of HIF1α in regulating lipid metabolism under hypoxia in Callosobruchus chinensis (Coleoptera: Bruchidae), a typical stored-product insect, was investigated. Based on the amplification and cloning, the sequence of the CcHIF1α gene was analysed and found to be highly conserved throughout evolution.

Optimization of untargeted lipidomics analysis of human plasma after Dangguijakyak-san administration.

Untargeted lipidomics enables comprehensive characterization of lipid profiles; however, analytical variability in sample preparation, chromatographic conditions, and data processing often limits reproducibility. Thus, this study aimed to optimize untargeted lipidomics analysis by evaluating chromatographic columns, mobile-phase additives, and lipid extraction methods to maximize feature detection, reproducibility, and overall data quality. Chromatographic performance was compared between ethylene-bridged hybrid (BEH) and charged-surface hybrid columns. For mobile-phase additives, 10 mM ammoni

Dysregulated sphingolipid metabolismdrives pancreatic carcinogenesis through plasma membrane Kras enrichment.

BACKGROUND: As key constituents of cellular sphingolipid pools, sphingomyelin (SM) and ceramide (CER) are central to the regulation of cancer cell death and survival. The metabolic flux between these two lipids is a vital component of the cellular stress response, yet the underlying regulatory mechanisms in cancer remain elusive. Acid sphingomyelinase (SMPD1) facilitates the conversion of SM to CER, functioning as a key enzymatic driver of CER-mediated signalling. OBJECTIVES: Herein, we aim to evaluate the role of SMPD1-driven sphingolipid metabolism in pancreatic carcinogenesis. DESIGN: A t

Skin health in oncology: evidence-based skin care for cancer patients.

BACKGROUND: Cancer and its treatments frequently compromise skin integrity, leading to xerosis, pruritus, dermatitis, radiation-induced skin damage, and secondary infections. These cutaneous adverse events (cAEs) are not merely superficial; they profoundly impact patient quality of life (QoL), contributing to physical discomfort, psychosocial distress, and poorer treatment adherence. Even mild skin toxicities elevate dermatology-specific QoL measures, affecting mood and social function. Additionally, new targeted therapies, such as daraxonrasib-a KRAS inhibitor under development for pancreatic

Ultrasound-Activated Microbubbles for Precision Chemo-Radioenhancement in Prostate Cancer: Sonoporation, Therapeutic Synergy, and Translational Pathways.

Background/Objectives: Radiotherapy and docetaxel are mainstays of prostate cancer management, yet both are constrained-radiotherapy by normal-tissue tolerance and docetaxel by systemic toxicity and acquired resistance. Ultrasound-activated microbubbles (USMB) convert acoustic energy into spatially localized mechanical bioeffects that can potentiate both. This review examines USMB as a precision chemo- and radio-enhancer in prostate cancer. Methods: We conducted a scoping review with mechanistic and translational synthesis, informed by PRISMA-ScR reporting principles. We identified sources thr

Spatial multi-omics unveils sphingolipid metabolic reprogramming within the retinal pathological niche.

Retinal ischemia-reperfusion (RIR) injury is a central mechanism underlying irreversible vision loss in glaucoma and other retinal diseases, yet the spatial organization of the pathogenic microenvironment remains poorly understood. Here, we constructed a high-resolution, whole-eye spatial multi-omics atlas integrating Stereo-seq transcriptomics and MALDI-MSI-based metabolomics to capture the early molecular events in the RIR mouse model. Spatially, retinal ganglion cell (RGC) interactions with surrounding cells were markedly reduced after injury, whereas immune-glial interactions were enhanced

Metabolite biomarkers in lung cancer: unlocking the potential of body fluid analysis for early detection and prognosis-a narrative review.

BACKGROUND AND OBJECTIVE: Lung cancer is a leading cause of cancer-related mortality, due to delayed diagnosis and the complexity of selecting the optimal treatment method, given the genetic diversity and heterogeneity of the disease. Traditional invasive techniques, such as tissue biopsy, carry risks of severe complications and are often costly. Therefore, there is increasing interest in non-invasive alternatives, particularly liquid biopsy. This review aims to propose promising circulating metabolite biomarkers for lung cancer and their clinical applications. METHODS: A PubMed search [2014-

Interrogating mediators of single-cell transcriptional changes in the acute damaged cerebral cortex: Insights into endothelial-astrocyte interactions.

Traumatic brain injury (TBI) induces complex cellular and molecular changes, challenging recovery and therapeutic development. Although molecular pathways have been implicated in TBI pathology, the cellular specificity of these mechanisms remains underexplored. Here, we investigate the role of endothelial cell (EC) EphA4, a receptor tyrosine kinase receptor involved in axonal guidance, in modulating cell-specific transcriptomic changes within the damaged cerebral cortex. Utilizing single-cell RNA sequencing (scRNA-seq) in an experimental TBI model, we mapped transcriptional changes across vari

Deletion Testing of the DEGS1 Gene Should Be Part of the Diagnostic Pipeline for Hypomyelinating Leukodystrophy (HLD18).

Hypomyelinating leukodystrophies are a heterogeneous group of disorders characterized by abnormal myelin formation in the central nervous system. Thanks to the increased use of NGS, a growing number of pathogenic single nucleotide variants in DEGS1 have recently been reported to be responsible for hypomyelinating leukodystrophy 18 (HLD18), a rare and severe autosomal recessive form. DEGS1 is a small gene (4 exons and 17 kb) encoding Δ4-dihydroceramide desaturase, which catalyzes the final step in ceramide biosynthesis. Here, we present two patients from unrelated families affected by severe an

Oral administration of the probiotic strain Lactobacillus helveticus BGRA43 reduces high-fat diet-induced hepatic steatosis in mice and restores gut microbiota composition.

INTRODUCTION: The prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD) is rapidly increasing. Modulation of the gut microbiota through the use of probiotics has been recognized as an important option for the treatment of hepatic steatosis. Previous studies suggested that the bacterial strain Lactobacillus helveticus BGRA43 (LHBGRA43) can reduce inflammation and improve the bacterial balance in the gut. The aim of this study was to investigate whether oral administration of LHBGRA43 in mice fed a high-fat diet contributes to the reduction of hepatic steatosis through i
