Presynaptic metabotropic glutamate receptors in adult and developing neurons: autoexcitation in the olfactory bulb.

The mitral cell of the olfactory bulb is the primary relay neuron that transmits information from the olfactory receptors to the rest of the brain. This excitatory neuron releases glutamate from presynaptic dendrites and axon terminals. All rat mitral cells studied showed strong, selective, and widespread metabotropic glutamate receptor mGluR1 alpha immunoreactivity on the presynaptic membrane of dendrites, often at the synaptic vesicle release site, when examined with light and electron microscopy. The finding of glutamate receptors on mitral cell secondary dendrites supports the conclusion t

Ion channel and receptor-mediated regulation of axonal conduction reliability in sympathetic preganglionic neurons.

UNLABELLED: Sympathetic preganglionic neurons (SPNs) provide the sole spinal output to the peripheral sympathetic nervous system. Although sympathetic control is traditionally attributed to synaptic integration within the spinal cord and ganglia, the reliability of spike propagation along SPN axons themselves has received little attention. Here, and in companion papers, we show that axonal conduction in adult mouse thoracic SPNs is highly modifiable and constitutes a critical site of sympathetic gain control. Using an ex vivo preparation preserving intact paravertebral and splanchnic pathways

Therapeutic potential of enzymes, neurosteroids, and synthetic steroids in neurodegenerative disorders: A critical review.

Neurodegenerative disorders present a significant challenge to healthcare systems, mainly due to the limited availability of effective treatment options to halt or reverse disease progression. Endogenous steroids synthesized in the central nervous system, such as pregnenolone (PREG), dehydroepiandrosterone (DHEA), progesterone (PROG), and allopregnanolone (ALLO), have been identified as potential therapeutic agents for neurodegenerative diseases. Neurosteroids such as ALLO, DHEA, and PROG, as well as their synthetic analogs like Ganaxolene, Fluasterone, and Olexoxime, offer promising effects f

Targeting Oxidative Stress and Neuroinflammation: Epigallocatechin-3-gallate-Selenium Nanoparticles Mitigate Sleep Deprivation-Induced Cortical Impairment.

Sleep deprivation (SD) has been revealed to provoke anxiety-like behavior. Phytochemicals and nanotechnology-based interventions have emerged as promising alternatives due to their pleiotropic activity and enhanced bioavailability. Here we investigated the effect of sodium selenite, Epigallocatechin-3-gallate (EGCG), and EGCG-Selenium nanoparticles (SeNPs) on SD-provoked cortical impairment and tried to recognize the possible underlying mechanisms in addition to in silico analysis of EGCG. SD was provoked in rats utilizing a modified multiple platform model. We performed an in silico analysis

In vivo characterization of the effects of human hemokinin-1 and human hemokinin-1(4-11), mammalian tachykinin peptides, on the modulation of pain in mice.

Human hemokinin-1 (h HK-1) and its truncated form h HK-1(4-11) are mammalian tachykinin peptides encoded by the recently identified TAC4 gene in human, and the biological functions of these peptides have not been well investigated. In the present study, an attempt has been made to investigate the effects and mechanisms of action of h HK-1 and h HK-1(4-11) in pain modulation at the supraspinal level in mice using the tail immersion test. Intracerebroventricular (i.c.v.) administration of h HK-1 (0.3, 1, 3 and 6 nmol/mouse) produced a dose- and time-related antinociceptive effect. This effect wa

Interaction of 2,4,4'-trichloro-2'-hydroxydiphenyl ether with microsomal cytochrome P450-dependent monooxygenases in rat liver.

We studied the effects of 2,4,4'-trichloro-2'-hydroxydiphenyl ether (Irgasan DP300) on the kinetics of the cytochrome P450 (P450)-dependent monooxygenases in rat liver microsomes. The activities of 7-ethoxyresorufin O-deethylase (EROD) and 7-pentoxyresorufin O-depentylase (PROD) in rat liver microsomes exposed to 3-methylcholanthrene (MC) and phenobarbital (PB) respectively, were substantially inhibited by Irgasan DP300. The inhibition profile of EROD was competitive, whereas that of PROD was noncompetitive; the Ki values from Hanes plots were 0.24 and 1.48 microM for EROD and PROD, respective

The protective effects and mechanisms of Polygonatum sibiricum polysaccharides in chronic stress-induced neural damage.

ETHNOPHARMACOLOGICAL RELEVANCE: Polygonatum sibiricum (PS) has a long-standing history of application in traditional Chinese medicine and various ethnic pharmacopeias to treat fatigue, aging-related weakness, memory decline, and metabolic disorders. It is believed to "tonify Qi and Yin," "nourish the kidney and spleen," and enhance vitality. Recent pharmacological studies have confirmed its antioxidant, neuroprotective, and anti-aging properties, suggesting its potential in improving cognitive impairment and neural damage, particularly under chronic stress or degenerative conditions. However,

Gut microbiota and neurotransmitter metabolic profiling in a rat model simulating hypoactive sexual desire disorder.

BACKGROUND: Hypoactive Sexual Desire Disorder (HSDD) is a highly prevalent, clinically impactful female sexual health condition whose pathogenesis is closely associated with disrupted excitation-inhibition homeostasis in the central nervous system, while the mechanistic role of gut microbiota in this pathological process via the gut-brain axis remains largely unelucidated. AIM: To explore gut microbiota-neurotransmitter interactions in an HSDD-simulating rat model and verify their causal role via fecal microbiota transplantation (FMT). METHODS: Female Sprague-Dawley rats were stratified into

Tirzepatide attenuates mesolimbic cocaine-evoked dopamine levels and reduces cocaine taking, motivation and seeking behaviours in male rodents.

BACKGROUND: Cocaine use disorder (CUD) remains among the most treatment-resistant addictions, characterised by high relapse rates even following extended abstinence periods. Dopamine signalling in mesocorticolimbic circuits contributes to cocaine's reinforcing effects and remains an important target for therapeutic intervention. Growing evidence suggests appetite-regulating peptides may influence central dopamine transmission. Whether recently approved incretin polyagonists can modulate cocaine-related behaviours through their capacity to simultaneously engage multiple appetite-regulating pept

Impaired neuronal macroautophagy in the prelimbic cortex contributes to comorbid anxiety-like behaviors in rats with chronic neuropathic pain.

A large proportion of patients with chronic pain experience co-morbid anxiety. The medial prefrontal cortex (mPFC) is proposed to underlie this comorbidity, but the molecular and neuronal mechanisms are not fully understood. Here, we reported that impaired neuronal macroautophagy in the prelimbic cortical (PrL) subregion of the mPFC paralleled the occurrence of anxiety-like behaviors in rats with chronic spared nerve injury (SNI). Intriguingly, such macroautophagy impairment was mainly observed in a FOS/c-Fos+ neuronal subpopulation in the PrL. Chemogenetic inactivation of this comorbid anxiet

Changes in resting-state functional connectivity after stroke in a mouse brain lacking extracellular matrix components.

In the brain, focal ischemia results in a local region of cell death and disruption of both local and remote functional neuronal networks. Tissue reorganization following stroke can be limited by factors such as extracellular matrix (ECM) molecules that prevent neuronal growth and synaptic plasticity. The brain's ECM plays a crucial role in network formation, development, and regeneration of the central nervous system. Further, the ECM is essential for proper white matter tract development and for the formation of structures called perineuronal nets (PNNs). PNNs mainly surround parvalbumin/GAB

Enhanced Neurophysiological Benefits of Magnesium-Acetyl-Taurate Over Magnesium-L-Threonate: A Comparative pre-clinical Study on Bioavailability, Synaptic Plasticity and Cognitive Functions.

The study examines how magnesium compounds, especially Magnesium-L-Threonate (MLT) and Magnesium-Acetyl-Taurate (MAT), affect neurophysiological functions in adult Wistar rats. Magnesium is essential for many cellular processes in the brain & peripheral system, such as neurotransmitter regulation, muscle function, and energy metabolism. Tissue Mg²⁺ levels were defined as the primary endpoint of the study, as they directly reflect the central objective of evaluating magnesium-based interventions. All other outcomes, including behavioral, biochemical, and molecular parameters, were considered se

SETX (senataxin), the helicase mutated in AOA2 and ALS4, functions in autophagy regulation.

SETX (senataxin) is an RNA/DNA helicase that has been implicated in transcriptional regulation and the DNA damage response through resolution of R-loop structures. Mutations in SETX result in either of two distinct neurodegenerative disorders. SETX dominant mutations result in a juvenile form of amyotrophic lateral sclerosis (ALS) called ALS4, whereas recessive mutations are responsible for ataxia called ataxia with oculomotor apraxia type 2 (AOA2). How mutations in the same protein can lead to different phenotypes is still unclear. To elucidate AOA2 disease mechanisms, we first examined gene

A thioacrylamide-based compound directly counteracts hepatic fibrosis with profound anti-obesity action.

BACKGROUND & AIMS: Obesity-associated metabolic dysfunction-associated steatotic liver disease (MASLD) and its progression to steatohepatitis (MASH) with advanced fibrosis pose a major global health challenge. Targeting γ-aminobutyric acid (GABA)ergic system has shown promise in mitigating liver injury. Therefore, we investigated HK3, a positive allosteric modulator of the GABAA receptor, as a potential therapy for MASH, with a particular focus on liver fibrosis and obesity. METHODS: Human-derived 3D MASH spheroids (n = 4-27) and hepatic stellate cells (LX2, n = 3-5) were treated with increas

Comparative Analysis of Pain Perception, Corticomotor Excitability, Disability, and Cognitive Function in Chronic Low Back Pain Patients and Age-Matched Healthy Controls.

Background and introduction Chronic low back pain (CLBP) is a common, disabling condition with a multifactorial aetiology that extends beyond nociceptive dysfunction. Among the Indian population, low backache is a significant contributor to years lived with disability (YLD). Recent evidence suggests that CLBP can cause significant alterations in central nervous system functioning and neuroplasticity, via deficits in intracortical modulation involving glutamatergic and gamma-aminobutyric acid (GABA)-mediated mechanisms. Understanding the broader impact of CLBP and its effects on the nervous sys
