Preclinical Safety Assessment of the HB-ATV-8 Intranasal Vaccine Designed to Control Atherosclerosis and Metabolic-Associated Fatty Liver Disease.

BACKGROUND AND AIM: The intranasal vaccine HB-ATV-8 contains the Seq-1 peptide (CHLLVDFLQSLS) as the active component within a nanoparticle formulation. It targets the cholesterol ester transfer protein (CETP) by inducing the formation of specific autoantibodies and a cellular antifibrotic response. It has demonstrated efficacy in preventing atherogenesis, liver steatosis, and fibrosis in preclinical models. Given that previously synthesized CETP inhibitors have been associated with increased cardiovascular risk, a preclinical safety assessment of HB-ATV-8 was conducted. RESULTS: Histopatholo

Investigation of liposomal self-adjuvanting peptide epitopes derived from conserved blood-stage Plasmodium antigens.

Malaria is a vector born parasitic disease causing millions of deaths every year. Despite the high mortality rate, an effective vaccine against this mosquito-borne infectious disease is yet to be developed. Up to date, RTS,S/AS01 is the only vaccine available for malaria prevention; however, its efficacy is low. Among a variety of malaria antigens, merozoite surface protein-1(MSP-1) and ring-infected erythrocyte surface antigen (RESA) have been proposed as promising candidates for malaria vaccine development. We developed peptide-based Plasmodium falciparum vaccine candidates that incorporated

Immune response of C57BL/6J mice to herpes zoster subunit vaccines formulated with nanoemulsion-based and liposome-based adjuvants.

Herpes zoster (HZ) is a recurrent nerve tissue infection caused by the reactivation of varicella-zoster virus (VZV). At present, two vaccines, the live attenuated vaccine Zostavax™ and AS01B-adjuvanted recombinant subunit vaccine Shingrix™, are commercially available for HZ. The latter is superior to the former in terms of efficacy and duration of immunity in the elderly. In this study, we used glycoprotein E (gE) as an antigen, and investigated the effects of various adjuvants (MF59, MF59/CpG 2006, and MF59/QS-21) on the immune response of C57BL/6J mice to find an alternative adjuvant to AS01

Primary evaluation of immune responses in mice after a single mRNA immunization with Echinococcus granulosus Eg95 and Coenurus cerebralis TM16.

Echinococcus granulosus and Taenia multiceps cause zoonotic cystic diseases. Studies have shown that Eg95 from E. granulosus is highly conserved, exhibits excellent immunogenicity, and serves as a critical candidate antigen. In recent years, the TM16 recombinant protein antigen has been one of the most studied antigens for preventing C. cerebralis and demonstrates optimal experimental immunogenicity. This study developed both recombinant protein and mRNA-LNP vaccines targeting the Eg95 and TM16 antigens. Immunization of BALB/c mice showed that both vaccines significantly increased specific ant

Proteome analysis of Neisseria meningitidis serogroup A.

Neisseria meningitidis is an encapsulated Gram-negative bacterium responsible for significant morbidity and mortality worldwide. Meningococci are opportunistic pathogens, carried in the nasopharynx of approximately 10% of asymptomatic adults. Occasionally they enter the bloodstream to cause septicaemia and meningitis. Meningococci are classified into serogroups on the basis of polysaccharide capsule diversity, and serogroup A strains have caused major epidemics mainly in the developing world. Here we describe a two-dimensional gel electrophoresis protein map of the serogroup A strain Z4970, a

AlphaFold reveals how pathogenic Leptospira use cross-kingdom thiol-disulfide exchange to evade the complement membrane attack complex.

UNLABELLED: Leptospirosis is a globally significant severe infectious disease that affects a range of mammals. To understand its complex aetiopathogenesis, Leptospira spp. are classified as pathogenic or saprophytic, with serum resistance a key differentiator of virulence. LIC13259 is a leptospiral surface lipoprotein that binds C8 of the host complement system; however, the molecular basis of this interaction has remained unclear. AlphaFold2 structural predictions, together with protein engineering, biochemical, and immunological validations, revealed that LIC13259 binds specifically to the C

Clinical Immunogenicity Assessment of HIV/AIDS Vaccines: Current Progress and Future Perspectives.

Developing a safe and effective HIV-1 vaccine remains the cornerstone of global efforts to terminate the AIDS pandemic. However, the virus's immense genetic diversity and the persistent absence of a universal Correlate of Protection (CoP) present formidable barriers. Concomitant with the iterative evolution of vaccine design-spanning from protein subunits to mRNA and germline-targeting platforms-immunogenicity evaluation frameworks have undergone a critical paradigm shift. The field has transitioned from relying on conventional serological assays to embracing comprehensive, high-dimensional im

Truncated Diphtheria Toxin DT390 Enhances the Humoral Immunogenicity of Porcine Circovirus Type 2 Capsid Antigen in Mice.

Porcine circovirus type 2 (PCV2) is the causative agent of PCV-associated disease, which harms the swine industry worldwide. Open reading frame 2 of PCV2 encodes the principal immunogenic capsid (Cap) protein, which induces neutralizing antibodies and protective immunity. Cap has been developed as a subunit vaccine against PCV2 infection, although its use is hindered by low immunogenicity. Here, we hypothesized that the truncated diphtheria toxin DT390 might enhance the immunogenicity of Cap. To verify this hypothesis, we fused Cap with DT390, which was expressed using the unique diphtheria to

Survey of Omp19 immunogenicity against Brucella abortus and Brucella melitensis: influence of nanoparticulation versus traditional immunization.

Brucellosis is the most common zoonotic bacterial disease. Prevention of human brucellosis is achieved through pasteurization of dairy products, appropriate sanitation and vaccination of domestic animals against the Brucella species. B. abortus unlipidated 19 kDa outer membrane protein (U-Omp19) is a promising candidate for a subunit vaccine against brucellosis. This study investigates immunogenicity of Omp19 alone and with Freund's adjuvant (Omp19-IFA) and N-trimethyl chitosan (TMC/Omp19) nanoparticles, as well as the effect of Omp19 administration route on immunological responses and protect

Enhanced Transdermal Immunization via Solid-in-Oil Nanodispersions Incorporating Dendritic Cell-Targeting Peptide.

Transdermal immunization represents a promising needle-free alternative to conventional vaccination. However, efficient antigen delivery and robust immune activation remain major challenges. In this study, a solid-in-oil (S/O) nanodispersion system comprising a dendritic cell-targeting peptide (HR8), ovalbumin (OVA), and an adjuvant was developed for transdermal immunization. The HR8 peptide along with OVA was successfully incorporated into an S/O nanodispersion with an optimal hydrodynamic diameter of particles (<200 nm) and exhibited stable physical properties for up to 90 days. In vitro and

Development of a Heat-Killed fbp1 Mutant Strain as a Therapeutic Agent To Treat Invasive Cryptococcus Infection.

In previous studies, we determined that the F-box protein Fbp1, a subunit of the SCF(Fbp1) E3 ligase in Cryptococcus neoformans, is essential for fungal pathogenesis. Heat-killed fbp1Δ cells (HK-fbp1) can confer vaccine-induced immunity against lethal challenge with clinically important invasive fungal pathogens, e.g., C. neoformans, C. gattii, and Aspergillus fumigatus. In this study, we found that either CD4+ T cells or CD8+ T cells were sufficient to confer protection against lethal challenge by C. neoformans in HK-fbp1-induced immunity. Given the potent effect of HK-fbp1 as a preventative

Safety and immunogenicity of the chlamydia vaccine candidate CTH522 adjuvanted with CAF01 liposomes or aluminium hydroxide: a first-in-human, randomised, double-blind, placebo-controlled, phase 1 trial.

BACKGROUND: Chlamydia is the most common sexually transmitted bacterial infection worldwide. National screening programmes and antibiotic treatment have failed to decrease incidence, and to date no vaccines against genital chlamydia have been tested in clinical trials. We aimed to assess the safety and immunogenicity, in humans, of a novel chlamydia vaccine based on a recombinant protein subunit (CTH522) in a prime-boost immunisation schedule. METHODS: This phase 1, first-in-human, double-blind, parallel, randomised, placebo-controlled trial was done at Hammersmith Hospital in London, UK, in

T cell-associated immunity induced by heterologous recombinant BCG and purified protein vaccination confers cross-variant protection against SARS-CoV-2.

Previously, we have developed a recombinant BCG-based vaccine (rBCG) expressing a chimeric protein with spike and nucleocapsid epitopes (rChimera) to combine BCG's innate immune training capacity with SARS-CoV-2-specific adaptive immune responses. The heterologous prime-boost rBCG/rChimera+Alum regimen induced strong humoral and cellular immunity, conferring protection against the ancestral Wuhan strain in K18-hACE2 mice. Here, we used knockout mice lacking functional B cells, CD4+, or CD8+ T lymphocytes expressing hACE2 to uncover the mechanisms of anti-viral immune protection, while also eva

A VSV-vector vaccine simultaneously targeting H5N1 hemagglutinin and matrix protein 2 induces robust neutralizing and ADCC antibody responses and provides full protection against lethal H5N1 infection in a mouse model.

UNLABELLED: Avian influenza A viruses, especially highly pathogenic avian influenza (HPAI) viruses, pose a significant public health threat, and a multivalent vaccine is the primary prophylactic measure to control these viruses. To establish such a vaccine, we generated two multivalent vesicular stomatitis virus (VSV)-based vaccine candidates that simultaneously target H5N1 hemagglutinin (H505 or H522) and the ectodomain of matrix protein 2 (M2e) and characterized their ability to induce protective immune responses. Our results revealed that vaccine immunization induced high humoral immune res

Autophagy induction by the pathogen receptor NECTIN4 and sustained autophagy contribute to peste des petits ruminants virus infectivity.

Macroautophagy/autophagy is an essential cellular response in the fight against intracellular pathogens. Although some viruses can escape from or utilize autophagy to ensure their own replication, the responses of autophagy pathways to viral invasion remain poorly documented. Here, we show that peste des petits ruminants virus (PPRV) infection induces successive autophagic signalling in host cells via distinct and uncoupled molecular pathways. Immediately upon invasion, PPRV induced a first transient wave of autophagy via a mechanism involving the cellular pathogen receptor NECTIN4 and an AKT-

Sequence analysis and characterization of pyruvate kinase from Clonorchis sinensis, a 53.1-kDa homopentamer, implicated immune protective efficacy against clonorchiasis.

BACKGROUND: Clonorchis sinensis, the causative agent of clonorchiasis, is classified as one of the most neglected tropical diseases and affects more than 15 million people globally. This hepatobiliary disease is highly associated with cholangiocarcinoma. As key molecules in the infectivity and subsistence of trematodes, glycolytic enzymes have been targets for drug and vaccine development. Clonorchis sinensis pyruvate kinase (CsPK), a crucial glycolytic enzyme, was characterized in this research. RESULTS: Differences were observed in the sequences and spatial structures of CsPK and PKs from h
