In utero and postnatal VX-770 administration rescues multiorgan disease in a ferret model of cystic fibrosis.

Cystic fibrosis (CF) is a multiorgan disease caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR). In patients with CF, abnormalities initiate in several organs before birth. However, the long-term impact of these in utero pathologies on disease pathophysiology is unclear. To address this issue, we generated ferrets harboring a VX-770 (ivacaftor)-responsive CFTR G551D mutation. In utero VX-770 administration provided partial protection from developmental pathologies in the pancreas, intestine, and male reproductive tract. Homozygous CFTR G551D/G551D animals sho

Duchenne Muscular Dystrophy Mice and Men: Can Understanding a Genetic Cardiomyopathy Inform Treatment of Other Myocardial Diseases?

Duchenne muscular dystrophy (DMD) mouse models have a predictable and reproducible time-course of cardiomyopathy progression with discrete pathogenic steps, which closely parallel what we know occurs in the cardiomyopathy of patients with DMD. The slow progression of early pathogenic steps common to many cardiomyopathies may make DMD models useful for identifying novel treatment targets and testing the therapeutic value of new treatment paradigms for an at-risk patient population far beyond those with muscular dystrophies.

Searching for the synergistic effect between aclidinium and formoterol: From bench to bedside.

Aim of our study was to understand if the interaction between aclidinium and formoterol administered at therapeutic doses leads to a synergistic rather than additive broncholytic effect. We tested the type of effect ex vivo on isolated human bronchi and then in vivo in COPD patients. The analysis of the interaction between aclidinium and formoterol in vitro was measured by applying the Unified Theory, whereas that in COPD patients was measured by applying the Bliss Independence criterion. Aclidinium and formoterol administered alone completely relaxed human isolated bronchial tissues sub-maxim

Stem cells for brain repair and recovery after stroke.

INTRODUCTION: Stroke is a major worldwide cause of death and disability. Currently, intravenous thrombolysis and reperfusion therapies, but not the so-called neuroprotectant drugs, have been shown to be effective for acute ischemic stroke. Thus, new strategies to promote brain plasticity are necessary. Stem cell administration is an attractive future therapeutic approach. AREAS COVERED: Brain protection and repair mechanisms are activated after stroke. This article is focused on the capacity of stem cell-based therapy to enhance this postinfarct brain plasticity and recovery. Future therapeut

Allelic-dependent expression of an activating Fc receptor on B cells enhances humoral immune responses.

B cells are pivotal regulators of acquired immune responses, and recent work in both experimental murine models and humans has demonstrated that subtle changes in the regulation of B cell function can substantially alter immunological responses. The balance of negative and positive signals in maintaining an appropriate B cell activation threshold is critical in B lymphocyte immune tolerance and autoreactivity. FcγRIIb (CD32B), the only recognized Fcγ receptor on B cells, provides immunoglobulin G (IgG)-mediated negative modulation through a tyrosine-based inhibition motif, which down-regulates

Immunohistochemistry of thymic epithelial tumors as a tool in translational research.

Due to their heterogeneity and infrequency, thymic epithelial cell tumors (TET) represent a diagnostic as well as a therapeutic problem. In the early stage of disease TET are usually cured by performing radical resections, whereas in advanced stages of disease they are usually radically unresectable from the beginning, and often show multiple relapses and/or intra- or extrathoracic metastases. Trained pathologists are required in TET diagnostics; awareness of the complexity of the mediastinum and of the differential diagnostic possibilities is mandatory. Immunohistochemical (IHC) studies play

Cellular localization of NKCC2 and its possible role in the Cl- absorption in the rat and human distal colonic epithelia.

Recently, we demonstrated the expression of NKCC2, an absorptive isoform of NKCC specifically expressed in the kidney, in the rat gastrointestinal tract including the distal colonic mucosa. This study aims to investigate its localization in colonic epithelia and possible role in the colonic ion transport. Reverse transcription polymerase chain reaction (RT-PCR), Western blotting, and immunohistochemistry were used to investigate the expression and localization of NKCC2. The role of NKCC2 on the colonic ion transport was examined by mean of short-circuit current (I(SC)) monitoring. The results

Early trajectory of clinical global impression as a transdiagnostic predictor of psychiatric hospitalisation: a retrospective cohort study.

BACKGROUND: Identifying patients most at risk of psychiatric hospitalisation is crucial to improving service provision and patient outcomes. Existing predictors focus on specific clinical scenarios and are not validated with real-world data, limiting their translational potential. This study aimed to determine whether early trajectories of Clinical Global Impression Severity are predictors of 6 month risk of hospitalisation. METHODS: This retrospective cohort study used data from the NeuroBlu database, an electronic health records network from 25 US mental health-care providers. Patients with

Characterization in the rat of the individual tendency to rely on alcohol to cope with distress and the ensuing vulnerability to drink compulsively.

Only some vulnerable individuals who recreationally drink alcohol eventually develop the compulsive drinking pattern that characterizes alcohol use disorder. A new frontier in biomedical research lies in understanding the neurobehavioural mechanisms of this individual vulnerability, a necessary step towards developing novel effective therapeutic strategies. Translational research has been hindered by the lack of valid, reliable and robust approaches that enable the study of the influence of the reliance on alcohol to cope with stress or self-medicate negative emotional states on the subsequent

A large-scale genome-wide association study meta-analysis of cannabis use disorder.

BACKGROUND: Variation in liability to cannabis use disorder has a strong genetic component (estimated twin and family heritability about 50-70%) and is associated with negative outcomes, including increased risk of psychopathology. The aim of the study was to conduct a large genome-wide association study (GWAS) to identify novel genetic variants associated with cannabis use disorder. METHODS: To conduct this GWAS meta-analysis of cannabis use disorder and identify associations with genetic loci, we used samples from the Psychiatric Genomics Consortium Substance Use Disorders working group, iP

Plasma proteomic signature predicts who will get persistent symptoms following SARS-CoV-2 infection.

BACKGROUND: The majority of those infected by ancestral Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) during the UK first wave (starting March 2020) did not require hospitalisation. Most had a short-lived mild or asymptomatic infection, while others had symptoms that persisted for weeks or months. We hypothesized that the plasma proteome at the time of first infection would reflect differences in the inflammatory response that linked to symptom severity and duration. METHODS: We performed a nested longitudinal case-control study and targeted analysis of the plasma proteome of 1

Innate Immunity of Framework Nucleic Acids.

ConspectusNucleic acid nanotechnology has fundamentally transcended the classic paradigm of DNA and RNA as passive carriers of genetic blueprints, which enables the rational design and construction of precise nanostructures with defined shapes, dynamics, and functions. This programmability has revolutionized approaches in biomedicine, facilitating breakthroughs in high-resolution molecular diagnostics, spatially and temporally controlled drug delivery, and the creation of synthetic cellular machinery. However, a central challenge for clinical translation is the inherent immunogenicity of nucle

Multifunctional electrospun nanofibers for cancer therapy and regenerative reconstruction: current progress and future prospects.

Breast cancer continues to be the most commonly diagnosed cancer among women globally, and despite significant progress in standard treatment approaches, it is still associated with substantial recurrence rates and postoperative complications. Systemic toxicity, drug resistance, poor tumor targeting, and compromised quality of life limit current treatment strategies, such as chemotherapy, radiotherapy, and surgery. In this context, multifunctional electrospun nanofibers have emerged as a promising platform for localized cancer therapy combined with regenerative reconstruction. This review comp

[Optimization and scale-up of recombinant elastomeric protein expression and fermentation process].

Recombinant elastomeric protein exhibits excellent biocompatibility, tunable mechanical properties, and biomimetic microenvironment characteristics, offering broad application prospects in the biomedical and food sectors. However, its low production efficiency has hindered further research and industrial translation. To address this challenge, this study established a stable and efficient large-scale preparation process through a multi-stage optimization strategy. First, codon optimization of ure(80) increased the yield to 0.42 g/L. Targeted truncation analysis identified amino acid residues 5

Nanoantibodies: A Review of Diagnostic and Therapeutic Applications in Respiratory Diseases.

BACKGROUND: Conventional monoclonal antibodies generated by hybridoma technology have long been applied in disease diagnosis and treatment, yet they are constrained by inherent limitations, including epitope loss caused by chemical antigen processing and insufficient tissue penetration. Nanobodies, as single-domain antibody fragments derived from camelids and chondrichthyans, possess unique superior physicochemical characteristics such as low molecular weight, low immunogenicity, easy modification, strong tissue permeability, and precise epitope targeting. Notably, camelid-derived nanobodies e

Implementation of a computational medical assemblage heuristic to improve rational design of phytomedicines for safety, efficacy and regulatory approval.

Phytomedicines are key to global healthcare and underutilized in Western medical systems. The inherent complexity of multi-species polypharmaceutical formulations presents barriers (standardization, reproducibility and regulatory approval) that can relegate potentially safe, effective and lower cost medication to nutraceutical settings in countries such as the US. The identification of Minimal Essential Effective Formulations (MEEFs) is a strategy to rationally simplify phytomedicines while preserving efficacy, but it requires computational innovation to de-risk and prioritize compounds due to

Newborn care practices in rural Bangladesh: Implications for the adaptation of kangaroo mother care for community-based interventions.

Bangladesh has one of the world's highest rates of low birth weight along with prevalent traditional care practices that leave newborns highly vulnerable to hypothermia, infection, and early death. We conducted formative research to explore existing newborn care practices in rural Bangladesh with an emphasis on thermal protection, and to identify potential facilitators, barriers, and recommendations for the community level delivery of kangaroo mother care (CKMC). Forty in-depth interviews and 14 focus group discussions were conducted between September and December 2012. Participants included p
