We report a case of cryptococcal sinusitis, a rare presentation of Cryptococcus neoformans infection in a patient with multiple myeloma. The objective of this case report is to highlight the utility of structural and functional imaging modalities in the differential diagnosis of sinonasal soft tissue masses in the immunocompromised patient population. PET-CT was the first imaging modality in this patient, who presented for routine follow-up staging of multiple myeloma, and was asymptomatic at the time of his presentation. PET-CT findings prompted further evaluation with MRI, to aid in the diff

Cyanogenic glycosides (CNglcs) are bioactive plant products derived from amino acids. Structurally, these specialized plant compounds are characterized as α-hydroxynitriles (cyanohydrins) that are stabilized by glucosylation. In recent years, improved tools within analytical chemistry have greatly increased the number of known CNglcs by enabling the discovery of less abundant CNglcs formed by additional hydroxylation, glycosylation, and acylation reactions. Cyanogenesis--the release of toxic hydrogen cyanide from endogenous CNglcs--is an effective defense against generalist herbivores but less

The dimorphic fungus Paracoccidioides spp. is responsible for paracoccidioidomycosis, the most prevalent systemic mycosis in Latin America, causing serious public health problems. Adequate treatment of mycotic infections is difficult, since fungi are eukaryotic organisms with a structure and metabolism similar to those of eukaryotic hosts. In this way, specific fungus targets have become important to search of new antifungal compound. The role of the glyoxylate cycle and its enzymes in microbial virulence has been reported in many fungal pathogens, including Paracoccidioides spp. Here, we show

Pdr5 is a major ATP-binding cassette (ABC) multidrug transporter regarded as the founding member of a fungal subfamily of clinically significant efflux pumps. When these proteins are overexpressed, they confer broad-spectrum ultraresistance. To better understand the evolution of these proteins under selective pressure, we exposed a Saccharomyces cerevisiae yeast strain already overexpressing Pdr5 to a lethal concentration of cycloheximide. This approach gave mutations that confer greater resistance to a subset of transport substrates. One of these mutations, V656L, is located in intracellular

Signal transduction systems comprising histidine kinases are suggested as new molecular targets of antibiotics. The important human fungal pathogen Candida albicans possesses three histidine kinases, one of which is the type III histidine kinase CaNik1, which activates the MAP kinase Hog1. We established a screening system for inhibitors of this class of histidine kinases by functional expression of the CaNIK1 gene in S. cerevisiae. This transformant was susceptible to fungicides to which the wild type strain was resistant, such as fludioxonil and ambruticin. Growth inhibition correlated with

BACKGROUND: Actinomyces is a gram-positive rod, some of which organisms are anaerobic and others which are facultative anaerobes. Most species of Actinomvces cluster together, forming a structure resembling a fungal colony. The organism is an opportunistic pathogen most commonly seen in the oral cavity but may also infect the gastrointestinal tract, lungs and uterus. Actinomyces is also a part of the normal vaginal flora and can result in infection independent of intrauterine device (IUD) use. Abscesses and swelling are common at the site of the infection. The organisms are grossly apparent by

Mucormycosis is a rare disorder with a potentially lethal course caused by an opportunistic fungus of the Phycomycetes family. Rhinocerebral mucormycosis (RCM) is an acute fulminant form of invasive fungal sinusitis occurring principally in individuals who are immunologically or metabolically compromised. This form originates from one of the paranasal sinuses and may spread to the orbit or the intracranial space either by direct extension or using the unique capability of spreading along vascular walls, leaving the bony structure intact. Carotid artery occlusion is a complication of RCM. Becau

We report that the fission yeast nucleoporin Nup124p is required for the nuclear import of both, retrotransposon Tf1-Gag as well as the retroviral HIV-1 Vpr. Failure to import Tf1-Gag into the nucleus in a nup124 null mutant resulted in complete loss of Tf1 transposition. Similarly, nuclear import of HIV-1 Vpr was impaired in nup124 null mutant strains and cells became resistant to Vpr's cell-killing activity. On the basis of protein domain similarity, the human nucleoporin Nup153 was identified as a putative homolog of Nup124p. We demonstrate that in vitro-translated Nup124p and Nup153 coimmu

UNLABELLED: Distiller's grains (DG) are a potential source of animal feeds, and many studies have indicated positive regulatory roles of feeding DG diets in animal breeding. However, there is currently a dearth of research on the actions and underlying mechanisms of probiotics-fermented distiller's grains (FDG)-based diets in cattle breeding. This study aimed to assess the impact of integrating FDG into the diet of finishing cattle on their fecal microbial community and metabolites. Thirty Simmental crossbred cattle (local yellow cattle × Simmental cattle, 8.5 months old, 420.38 ± 68.11 kg) we

Lignin is the third most abundant organic resource in nature. The utilization of white-rot fungi for wood degradation effectively circumvents environmental pollution associated with chemical treatments, facilitating the benign decomposition of lignin. Trametes gibbosa is a typical white-rot fungus with rapid growth and strong wood decomposition ability. The lignin content decreased from 23.62 mg/mL to 17.05 mg/mL, which decreased by 27 % in 30 days. The activity of manganese peroxidase increased steadily by 9.44 times. The activities of laccase and lignin peroxidase had the same trend of chang

The study investigates the antifungal potential of citrinin, the bioactive compound isolated from Aspergillus terreus (OP480427), against aflatoxin-producing fungi isolated from wheat rhizospheric soil and its ability to interfere with aflatoxin biosynthesis. Citrinin, purified using TLC, identified through polyphasic characterisation, effectively inhibited the growth of Aspergillus flavus (OP600575) and reduced aflatoxin production, a major food safety concern. Structural characterisation was performed using FTIR, HRMS and NMR. The reduction of aflatoxin levels was confirmed by HPLC analysis.

Biosurfactants are amphiphilic metabolites with antimicrobial properties, with their synthesis contingent upon the media utilized. This study investigated the antifungal activity of the biosurfactant generated by Bacillus licheniformis SCV1 grown in a mineral salt medium with various carbon sources. The research demonstrated that biosurfactants derived from media with several carbon sources, including glucose, glycerol, sucrose, starch, and mannitol, exhibited different antifungal efficacy against fungal pathogens, namely Trichophyton mentagrophytes and Trichophyton rubrum. The biosurfactant d

The soil-borne fungal pathogen Verticillium dahliae (V. dahliae) is the causal agent of Verticillium wilt (VW), a vascular disease that severely threatens global cotton production. Although cell wall lignification represents a cornerstone of plant immunity, the precise regulatory circuits that bridge this structural reinforcement with Verticillium dahliae resistance in cotton have yet to be fully elucidated. Here, we demonstrate that the NAC transcription factor GhNAC043 is a key positive regulator of this defense. GhNAC043 expression was rapidly induced upon V. dahliae infection. Silencing Gh

Fungi have a potential to produce a variety of natural products for their survival by utilizing dozens of biosynthetic gene clusters (BGCs). Fungal BGCs are typically conserved in species but recent studies showed that many BGCs are maintained in limited number of strains. However, little is reported for the products of strain specific BGCs. In this study, we analyzed publicly available genomic data of human pathogenic fungus Aspergillus lentulus and identified seven strain-specific BGCs in addition to (-)- and (+)-auranthine BGCs. One of these BGCs, leq cluster, was found in nine of thirteen

Dihydroxynaphthalene (DHN) derivatives are precursors of biologically important molecules, including melanin and perylenequinones (PQs). Biosynthesis of DHN-derived metabolites requires precise spatial control of radical reactions to form specific C─C or C─O─C bonds. Investigating fungal PQ biosynthesis in Alternaria alternata, we identified a cytochrome P450 enzyme that catalyzes a highly regio- and stereospecific C─C radical coupling reaction. Conversely, a DHN-converting P450 from Berkleasmium sp. utilizes a diradical mechanism yielding a C─O─C bridge. Comparing these enzymes revealed five

Phytopathogenic fungi pose a constant threat to worldwide agricultural production. Given the widespread development of fungicide resistance and increasing environmental and regulatory constraints, precision disease-control strategies are urgently needed to enhance selectivity, durability, and sustainability. This review critically examines recent advances in targeted fungicide discovery against phytopathogenic fungi. We categorize these strategies into three complementary groups: (1) targeting fungal biological processes that are essential or infection-associated; (2) disarming pathogen virule
