Nondirected Living Kidney Donation and Recipient Outcomes in the United States: A 20-Year Review.

INTRODUCTION: Nondirected donation (NDD) of the kidneys is a growing practice where donors who do not have any genetic or emotional relationship are selected to donate to a wide variety of recipients with a range of selection criteria and decisions which are left up to individual transplant centers. METHODS: We review all adult living kidney donor-recipient (DR) pairs and outcomes from NDDs who were recorded in United Network for Organ Sharing (UNOS) database as code 10 (anonymous) from October 1997 to September 2017 for demographics and outcomes. RESULTS: A total of 2174 DR pairs were ident

Virome in the Lungs: The Role of Anelloviruses in Childhood Respiratory Diseases.

More recently, increasing attention has been directed to exploring the function of the global virome in health and disease. Currently, by new molecular techniques, such as metagenomic DNA sequencing, the virome has been better unveiled. By investigating the human lung virome, we could provide novel insights into respiratory diseases. The virome, as a part of the microbiome, is characterized by a constant change in composition related to the type of diet, environment, and our genetic code, and other incalculable factors. The virome plays a substantial role in modulating human immune defenses an…

Genetic epidemiological characteristics of a Hungarian subpopulation of patients with Huntington's disease.

BACKGROUND: Recent advances in therapeutic options may prevent deterioration related to Huntington's disease (HD), even at the pre-symptomatic stage. Be that as it may, a well-characterized patient population is essential for screening and monitoring outcome. Accordingly, the aim of this study was to describe the characteristics of a Hungarian subpopulation of HD patients and mutation carriers diagnosed at the University of Szeged. METHODS: We conducted a search for International Classification of Diseases (ICD) code G10H0 in the local medical database for the period of 1 January 1998 to 31 D

Mycoplasma pneumoniae Genome Editing Based on Oligo Recombineering and Cas9-Mediated Counterselection.

Mycoplasma species share a set of features, such as lack of a cell wall, streamlined genomes, simplified metabolism, and the use of a deviant genetic code, that make them attractive approximations of what a chassis strain should ideally be. Among them, Mycoplasma pneumoniae arises as a candidate for synthetic biology projects, as it is one of the most deeply characterized bacteria. However, the historical paucity of tools for editing Mycoplasma genomes has precluded the establishment of M. pneumoniae as a suitable chassis strain. Here, we developed an oligonucleotide recombineering method for …

[Profiles and Risk of Antibiotic Resistance Genes in Domestic Wells in the Maozhou River Basin].

Intensive use of antibiotics promotes the occurrence and development of antibiotic resistance. Antibiotic resistance genes (ARGs) enter water environments from human and animal sources. Groundwater serves as an important water supply, while the profiles and risk of ARGs in groundwater remain unknown. The abundance and profiles of ARGs in 11 domestic wells in the Maozhou River basin of Shenzhen City were analyzed by high-throughput qPCR. The results showed that a total of 141 ARGs and 8 mobile genetic elements (MGEs) were detected, of which the genes corresponding resistance to sulfonamides, mu

Compensatory expression of multidrug-resistance genes encoding ABC transporters in dermatophytes.

Genomic sequencing of several dermatophyte species has revealed that they show small differences in genetic content and genome organization, although each fungus has adapted to specific niches. Thus, it seemed relevant to compare gene expression between species. Here, we examined the transcription modulation of three ATP-binding cassette (ABC) transporter genes (pdr1, mdr2 and mdr4), which code for membrane transporter proteins in four species of Trichophyton ; T. interdigitale, T. rubrum, T. tonsurans and T. equinum . These fungal species were challenged with sub-lethal doses of griseofulvin,

Mutational History of a Human Cell Lineage from Somatic to Induced Pluripotent Stem Cells.

The accuracy of replicating the genetic code is fundamental. DNA repair mechanisms protect the fidelity of the genome ensuring a low error rate between generations. This sustains the similarity of individuals whilst providing a repertoire of variants for evolution. The mutation rate in the human genome has recently been measured to be 50-70 de novo single nucleotide variants (SNVs) between generations. During development mutations accumulate in somatic cells so that an organism is a mosaic. However, variation within a tissue and between tissues has not been analysed. By reprogramming somatic c…

Parenting quality and children's mental health: biological mechanisms and psychological interventions.

PURPOSE OF REVIEW: The quality of parenting that children receive can have a profound influence on their development and mental health. This article reviews articles published from late 2010 onwards that address the effects of parenting on the child's physiological and genetic systems, and how interventions can improve children's security of attachments, antisocial behaviour and other outcomes across a range of settings. RECENT FINDINGS: Biological indices of stress, such as C-reactive protein, show that prenatal anxiety is a significant determinant of later outcomes for children, and abusive

[Mitochondrial encephalopathy due to complex I deficiency. Brain tissue biopsy findings and clinical course following pharmacological].

INTRODUCTION: Mitochondrial encephalomyopathies belong to a heterogeneous group of diseases with a range of neurological symptoms caused by a dysfunction somewhere in the nervous system. They may arise from mutations of the mitochondrial DNA or nuclear DNA in the genes that code for the subunits of the respiratory chain. The results obtained from using different drugs to treat these diseases vary widely. CASE REPORT: A 33-year-old female with a history of migraine with aura, who was admitted to hospital because of epileptic seizures. Neuroimaging tests showed left-side occipital insult and a

[Chronic progressive external ophthalmoplegia: clinical and electromyographic manifestations in a series of cases].

INTRODUCTION: Chronic progressive external ophthalmoplegia (CPEO) is a common mitochondrial disease. The different conditions in this group of diseases overlap clinically, enzymatically and genetically. There is no effective treatment. Ptosis improves with corrective surgery involving tarsorrhaphy as a palliative measure. CASE REPORTS: Code numbers were examined in a retrospective study conducted in order to search for patients with ptosis or ophthalmoplegia who had either visited or been admitted to the neurology department over the last 10 years. Data concerning these patients' clinical fea

The Italian Twin Project: from the personal identification number to a national twin registry.

The unique opportunity given by the "fiscal code", an alphanumeric identification with demographic information on any single person residing in Italy, introduced in 1976 by the Ministry of Finance, allowed a database of all potential Italian twins to be created. This database contains up to now name, surname, date and place of birth and home address of about 1,300,000 "possible twins". Even though we estimated an excess of 40% of pseudo-twins, this still is the world's largest twin population ever collected. The database of possible twins is currently used in population-based studies on multip

Cutaneous malignant melanoma: Tabuk experience.

Cutaneous melanoma (CM) has a rising morbidity and mortality in the western world but is rare in certain geographical areas including the Middle East. The aim of this study is to define the pattern of CM in this environment over a period of about two decades. A review of associated histological reports, dermatology, plastic general surgical admissions and outpatient census statistic of the North West Military Hospital (N.W.A.F.H.) were carried out from January 1978 to June 1996. The clinico-therapeutic information from both the review case and newly discovered CM was then studied. The result s

Non-Coding Negative Regulatory Features in Livestock Genomes: Functional Annotation and Causal Validation.

Non-coding negative regulation is an important but incompletely resolved component of gene-expression control in livestock. We conducted a narrative review using PubMed, Web of Science Core Collection, and Scopus, prioritizing primary studies in pigs, cattle, sheep, goats, and poultry and using model-organism studies only for directly relevant mechanistic or methodological evidence. We distinguish sequence-defined silencers, context-dependent enhancer-silencer elements, functional microRNA-binding sites, long non-coding RNA-associated regulation, and candidate repressive chromatin regions, and

An accurate cellular assay to determine pathogenicity of coding and noncoding variants in Lynch syndrome genes.

Lynch syndrome (LS) is a genetic predisposition to mainly colorectal and endometrial cancer due to heterozygous disruptive germline mutations in the DNA mismatch-repair (MMR) genes MSH2, MSH6, MLH1, or PMS2. Beyond clearly pathogenic mutations, germline sequencing often reveals variants of uncertain significance (VUS), predominantly single base-pair alterations in coding or noncoding regions. These uncertain variants obstruct LS diagnosis, hampering personalized surveillance. To address this challenge, we developed a highly accurate functional assay that interrogates VUS pathogenicity in human

Autophagy at the Crossroads of Protein and RNA Toxicity in Repeat Expansion Cerebellar Ataxias.

Repeat expansion cerebellar ataxias comprise a genetically and mechanistically heterogeneous group of neurodegenerative disorders unified by the pathological expansion of short tandem repeats (STRs) beyond a disease-causing threshold. Depending on their genomic localization, these expansions can lead to toxic protein gain-of-function, as in polyglutamine (polyQ) cerebellar ataxias, or to RNA-mediated toxicity and repeat-associated non-AUG (RAN) translation, for which recent evidence supports a major pathogenic role in non-coding spinocerebellar ataxias (SCAs). Despite these distinct upstream m

Genome-wide association study of cocaine self-administration behavior in Heterogeneous Stock rats.

Cocaine use disorder (CUD) is a major public health crisis. The specific genes mediating CUD remain largely unknown. We conducted a genome-wide association study (GWAS) using outbred N/NIH Heterogeneous Stock (HS; n = 836, female = 415, male = 421) rats. We examined CUD-related phenotypes including acquisition of self-administration, escalation of intake, and compulsive-like responding. These traits were phenotypically correlated and exhibited modest SNP heritability (h2 = 0.07 - 0.16). We identified six genome-wide significant associations (>-log10(p)=5.58; α = 0.05 by permutation). One locus

INTRODUCTION: Rapid diagnosis of bacterial pneumonia is crucial for clinical diagnosis and treatment, but traditional methods are time-consuming. The wide application of machine learning techniques in medical diagnosis provides an effective way to solve this problem. However, the complexity of medical datasets and the problem of class imbalance poses serious challenges to classical machine learning algorithms. METHODS: Aiming at the multiclass imbalanced problem in complete blood count (CBC) datasets, this study proposes a novel ensemble learning algorithm, Forest of Evolutionary Multi-Classi
