King Kong Pox: A Case of Severe Mpox and Bartonella Co-infection in a Patient Living With Human Immunodeficiency Virus (HIV).

Mpox, formerly monkeypox (MPXV), is a zoonotic virus that has emerged globally in recent times, disproportionately affecting immunocompromised individuals, particularly people living with human immunodeficiency virus (HIV) (PWH). PWH with advanced HIV and low CD4 counts are at higher risk for severe, prolonged courses and sometimes fatal Mpox infections, which may often be complicated by bacterial superinfections and opportunistic co-infections. We present a case of Mpox in a PWH with an initial CD4 count of 2 cells/μL and a high HIV viral load, who had worsening disease despite treatment with

An overview on mRNA-based vaccines to prevent monkeypox infection.

Источник пометил публикацию как отозванную

The human monkeypox virus (Mpox) is classified as a member of the Poxviridae family and belongs to the Orthopoxvirus genus. Mpox possesses double-stranded DNA, and there are two known genetic clades: those originating in West Africa and the Congo Basin, commonly known as Central African clades. Mpox may be treated with either the vaccinia vaccination or the therapeutics. Modifying the smallpox vaccine for treating and preventing Mpox has shown to be beneficial because of the strong link between smallpox and Mpox viruses and their categorization in the same family. Cross-protection against Mpox…

Brincidofovir in the Era of Mpox.

INTRODUCTION: Mpox (formerly monkeypox), a systemic infection caused by the mpox virus (MPXV), has become a global problem of increasing concern. There are currently no antiviral treatments that have been shown to be safe and effective for the treatment of mpox. AREAS COVERED: Brincidofovir is a lipid-modified acyclic nucleotide with in vitro and in vivo activity against multiple DNA viruses, including MPXV. It is licensed in the US and Canada for the treatment of human smallpox disease in adults and children, including neonates. It has been used under FDA-authorized emergency use for patient

Monkeypox Presenting as a Hand Consult in the Emergency Department: Two Case Reports.

The ongoing outbreak of the monkeypox virus (now referred to as "mpox") was deemed a public health emergency by the World Health Organization in 2022. The United States now reports the highest number of mpox cases, with 29 980 cases and 21 deaths as of January 11, 2023. The most common presenting symptom is a pruritic, vesicular rash that commonly involves the hands. While covering hand call, our division has encountered 2 cases of mpox in the emergency department for which the chief complaint was a hand lesion. Because hand surgeons will be called upon to make an initial diagnosis, the purpos…

Plasma-Like Culture Medium for the Study of Viruses.

Viral infections attract more and more attention, especially after the emergence of novel zoonotic coronaviruses and the monkeypox virus over the last 2 decades. Research on viruses is based to a great extent on mammalian cell lines that are permissive to the respective viruses. These cell lines are usually cultivated according to the protocols established in the 1950s to 1970s, although it is clear that classical media have a significant imprint on cell growth, phenotype, and especially metabolism. So, recently in the field of biochemistry and metabolomics novel culture media have been develo…

Use of JYNNEOS (Smallpox and Mpox Vaccine, Live, Nonreplicating) for Persons Aged ≥18 Years at Risk for Mpox During an Mpox Outbreak: Recommendations of the Advisory Committee on Immunization Practices - United States, 2023.

Since the worldwide eradication of smallpox in 1980, orthopoxvirus vaccines had been used nearly exclusively by persons at risk for occupational exposure to orthopoxviruses, including Monkeypox virus, the virus that causes mpox. However, during recent years, the epidemiology of mpox has been changing in countries where the animal reservoirs are believed to live and where endemic transmission has been known to occur for decades. CDC issues outbreak-specific vaccination recommendations based on the epidemiology at the time specific cases or clusters are identified; however, because of the increa…

BoHV-4-Based Vector Single Heterologous Antigen Delivery Protects STAT1(-/-) Mice from Monkeypoxvirus Lethal Challenge.

Monkeypox virus (MPXV) is the etiological agent of human (MPX). It is an emerging orthopoxvirus zoonosis in the tropical rain forest of Africa and is endemic in the Congo-basin and sporadic in West Africa; it remains a tropical neglected disease of persons in impoverished rural areas. Interaction of the human population with wildlife increases human infection with MPX virus (MPXV), and infection from human to human is possible. Smallpox vaccination provides good cross-protection against MPX; however, the vaccination campaign ended in Africa in 1980, meaning that a large proportion of the popul…

PoxiPred: An Artificial-Intelligence-Based Method for the Prediction of Potential Antigens and Epitopes to Accelerate Vaccine Development Efforts against Poxviruses.

Poxviridae is a family of large, complex, enveloped, and double-stranded DNA viruses. The members of this family are ubiquitous and well known to cause contagious diseases in humans and other types of animals as well. Taxonomically, the poxviridae family is classified into two subfamilies, namely Chordopoxvirinae (affecting vertebrates) and Entomopoxvirinae (affecting insects). The members of the Chordopoxvirinae subfamily are further divided into 18 genera based on the genome architecture and evolutionary relationship. Of these 18 genera, four genera, namely Molluscipoxvirus, Orthopoxvirus, P

The study of human monkeypox disease in 2022 using the epidemic models: herd immunity and the basic reproduction number case.

As of May 2022, a new outbreak of the human monkeypox (MPOX) disease appeared in multiple countries, where the 2022 human MPOX disease spread to more than 109 cases, excluding the suspected cases up to the end of 2022. The deaths of the 2022 human MPOX exceeded 200 cases up to the same date. The human MPOX is not a new disease, this disease was once endemic in some countries on the African continent. Despite this, this disease began to spread in a number of countries around the world in 2022. The first case of the 2022 human MPOX was recorded in the United Kingdom in May. After that date, this

Epitope mapping by random peptide phage display reveals essential residues for vaccinia extracellular enveloped virion spread.

BACKGROUND: A33 is a type II integral membrane protein expressed on the extracellular enveloped form of vaccinia virus (VACV). Passive transfer of A33-directed monoclonal antibodies or vaccination with an A33 subunit vaccine confers protection against lethal poxvirus challenge in animal models. Homologs of A33 are highly conserved among members of the Orthopoxvirus genus and are potential candidates for inclusion in vaccines or assays targeting extracellular enveloped virus activity. One monoclonal antibody directed against VACV A33, MAb-1G10, has been shown to target a conformation-dependent

Mpox virus D8L protein binds to STAT1 and inhibits its phosphorylation to antagonize IFN-induced signaling.

In August 2024, mpox was declared a public health emergency. However, little is known about how the proteins of the mpox virus (MPXV) interact with host innate immunity. This study identified that the D8L protein of MPXV inhibited the interferon (IFN)-mediated antiviral innate immune response and suppressed the mRNA transcription and protein expression of several IFN-stimulated genes, including Cig5, MX2, ISG56, IFITM1, OAS1, and ISG15. Mechanistically, D8L prevented antiviral immune responses by interacting with signal transducer and activator of transcription 1 (STAT1). Furthermore, D8L inte

Tiantan vaccinia virus-based vaccine with promising safety provides sustained protection against mpox in non-human primates.

The World Health Organization (WHO) has declared the mpox outbreak a public health emergency of international concern (PHEIC). Safe and efficient vaccines against the mpox virus (MPXV) are urgently needed to impede the surge in cases. Here, we report the results of a preclinical study employing different dosing strategies on a vaccine candidate named NTV, obtained via targeted gene deletion in the Tiantan strain vaccinia virus, resulting in a replication-deficient variant. Following optimisation of the NTV immunization dose and confirmation of its protective efficacy against MPXV in a mouse mo

[First appearance of monkey pox in human beings in Gabon].

The authors report on the first coming out in Gabon of human monkey-pox. Four children in the same family were simultaneously attacked. In one of the two fatal cases, diagnosis of monkey-pox was confirmed by the isolation of the virus. In the three other cases, diagnosis was based on clinical and epidemiological findings. The coming out of monkey-pox in Gabon squares with sporadic coming out of this zoonosis observed in the humid tropical forests of west and Central Africa. Two fatal cases presented some symptoms of hemorrhagic fever and cutaneous symptoms did not dominate the clinical feature
