Chronic arsenic exposure-provoked biotoxicity involved in liver-microbiota-gut axis disruption in chickens based on multi-omics technologies.

INTRODUCTION: Arsenic has been ranked as the most hazardous substance by the U.S. Agency for Toxic Substances and Disease Registry. Environmental arsenic exposure-evoked health risks have become a vital public health concern worldwide owing to the widespread existence of arsenic. Multi-omics is a revolutionary technique to data analysis providing an integrated view of bioinformation for comprehensively and systematically understanding the elaborate mechanism of diseases. OBJECTIVES: This study aimed at uncovering the potential contribution of liver-microbiota-gut axis in chronic inorganic ars

Genetic insights into therapeutic targets for gout: evidence from a multi-omics mendelian randomization study.

BACKGROUND: Considering that the treatment of gout is poor, we performed a Mendelian randomization (MR) study to identify candidate biomarkers and therapeutic targets for gout. METHODS: A drug-targeted MR study was performed for gout by integrating the gout genome-wide association studies (GWAS) summary data and cis expression quantitative trait loci of 2,633 druggable genes from multiple cohorts. Summary data-based Mendelian randomization (SMR) analyses based on transcript and protein levels were further implemented to validate the reliability of the identified potential therapeutic targets

Multi-omics investigation into long-distance road transportation effects on respiratory health and immunometabolic responses in calves.

BACKGROUND: Long-distance road transportation is a common practice in the beef industry, frequently resulting in bovine respiratory disease (BRD) and compromised growth performance. However, a comprehensive investigation integrating clinical performance, physiological conditions, and nasopharyngeal microflora remains lacking. METHODS: This study aimed to evaluate the respiratory health and immunometabolic status of 54 beef calves subjected to a 3000-km journey. The respiratory health of calves was monitored over 60 days post-arrival using a modified clinical scoring system. Nasopharyngeal mic

Associations of four biological age markers with child development: A multi-omic analysis in the European HELIX cohort.

BACKGROUND: While biological age in adults is often understood as representing general health and resilience, the conceptual interpretation of accelerated biological age in children and its relationship to development remains unclear. We aimed to clarify the relationship of accelerated biological age, assessed through two established biological age indicators, telomere length and DNA methylation age, and two novel candidate biological age indicators, to child developmental outcomes, including growth and adiposity, cognition, behavior, lung function and the onset of puberty, among European scho

Multi-omics reveals 2-bromo-4,6-dinitroaniline (BDNA)-induced hepatotoxicity and the role of the gut-liver axis in rats.

2-Bromo-4, 6-dinitroaniline (BDNA) is a widespread azo-dye-related hazardous pollutant. However, its reported adverse effects are limited to mutagenicity, genotoxicity, endocrine disruption, and reproductive toxicity. We systematically assessed the hepatotoxicity of BDNA exposure via pathological and biochemical examinations and explored the underlying mechanisms via integrative multi-omics analyses of the transcriptome, metabolome, and microbiome in rats. After 28 days of oral administration, compared with the control group, 100 mg/kg BDNA significantly triggered hepatotoxicity, upregulated t

ASCL2 drives colorectal cancer metastasis through transcriptional activation of TDGF1.

PURPOSE: Metastasis underlies most colorectal cancer (CRC)-related deaths, yet its molecular drivers remain incompletely characterized. This study aims to provide novel insights into the mechanisms underlying CRC metastasis through integrated multiomics and experimental validation. METHODS: Malignant epithelial cell subpopulations and key regulators of metastatic transition were identified through copy number variation inference and pseudotime trajectory analysis from CRC scRNA-seq data. Dual-luciferase reporter, chromatin immunoprecipitation, and functional studies were used for mechanistic v…

Integrated Single-Cell and Bulk Transcriptomic Analysis Reveals an Endothelial Gene Signature Shaping EndMT and Prognosis in Hepatocellular Carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most prevalent malignant tumors of the liver, with increasing incidence and mortality rates globally. This study investigates the role of endothelial cells (ECs) in the progression of HCC, particularly focusing on the process of endothelial-to-mesenchymal transition (EndMT) and its underlying molecular mechanisms. METHODS: We utilized single-cell RNA sequencing (scRNA-seq) and multiomics analyses to explore the involvement of ECs in HCC and their transformation mechanisms. The study emphasized the potential application of EC-related gen…

Juvenile exposure to imazalil disrupts epigenetic and transcriptomic regulation of hepatic energy metabolism in Xenopus tropicalis.

The extensive use of agricultural pesticides has led to widespread environmental contamination, raising concerns about their adverse biological effects. The fungicide imazalil is known to cause endocrine-disrupting and hepatotoxic effects in vertebrates, but potential underlying epigenetic mechanisms remain poorly understood. In this multiomics study, we investigated hepatic epigenetic and transcriptomic responses in juvenile Xenopus tropicalis (6.5 weeks post-metamorphosis) following two weeks exposure to an environmentally relevant concentration of imazalil (12.3 μg/L). Thousands of differen…

The GPCR Connection: Linking Alzheimer's Disease and Glioblastoma.

Alzheimer's disease (AD) and glioblastoma multiforme (GBM) are biologically distinct age-related brain disorders with opposing clinical phen … on or neuronal dysfunction, depending on the biological environment. Recent advances in integrative multiomics, computational modelling, artificial intelligence, and organoid systems have revealed GPCR-centred regulatory nodes and accelerated the identification of druggable targets. Collectively, these findings suggest that AD and GBM, although pathologicall …

Context-Dependent Signaling Network of Ginsenoside Rg2: A Molecular Basis for Its Bidirectional Pharmacological Effects.

Ginsenoside Rg2 (G-Rg2) is a key protopanaxatriol-type saponin from Panax ginseng. Metabolomics and multiomics studies in animal models suggest that G-Rg2 regulates metabolic networks and multiple signaling pathways simultaneously in a complex disease environment, suggesting that G-Rg2 is most likely a network-level regulatory mode rather than a linear singl …
