Distinct transcriptomic and metabolomic profiles characterize NSAID-induced urticaria/angioedema patients undergoing aspirin desensitization.

BACKGROUND: There is limited data on the mechanisms of aspirin desensitization in patients with nonsteroidal anti-inflammatory drug (NSAID)-induced urticaria/angioedema (NIUA). OBJECTIVES: We sought to characterize the transcriptomic and metabolomic profiles of patients with NIUA undergoing aspirin desensitization. METHODS: PBMCs and plasma were separated from the blood of patients with NIUA undergoing aspirin desensitization for coronary artery disease and NSAID-tolerant controls. RNA was isolated from PBMCs and subjected to messenger RNA (mRNA)- and long noncoding RNA (lncRNA)-sequencing.

Harnessing Artificial Intelligence-driven Approaches in the Diagnosis and Management of Diabetes and Diabetes-Associated Complications.

Diabetes mellitus is a global health issue with escalating metabolic dysfunction, clinical heterogeneity, and rising microvascular and macrovascular consequences. Traditional diabetes diagnosis, risk assessment, treatment selection, and complication surveillance rely on periodic clinical measures and population-level risk models, which may not fully capture dynamic, individualized disease pathways. Precision diabetes treatment is being transformed by Artificial intelligence (AI), including machine learning (ML), deep learning (DL), and multimodal data analytics. This review critically examines

AutoPCOS: a stepwise multimodal intelligent framework for polycystic ovary syndrome risk stratification and diagnostic support.

INTRODUCTION: Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder among women of reproductive age, typically diagnosed through a combination of clinical evaluation, laboratory testing, and ultrasonography. However, this multimodal diagnostic pathway is often time-consuming, costly, and dependent on resource availability, thereby limiting its accessibility in real-world clinical settings. METHODS: In this study, we propose AutoPCOS, a stepwise multimodal intelligent framework for flexible PCOS risk stratification and diagnostic support. Using a publicly available Kaggle PCOS dat

Examining selection dynamics and limitations in multi-round protein selection of high diversity libraries.

Proteins and peptides underpin essential biological functions and technological applications, from targeting disease-relevant interactions to providing broad enzymatic activities. However, engineering molecules with desired properties remains difficult, owing to complex sequence-structure-function relationships and the lack of data on specific systems. Experimental selection strategies, including directed evolution, phage display, and mRNA display, address this challenge by leveraging high diversity libraries and iterative enrichment under defined selection pressures. This allows for the ident

Development and Clinical Application of Digital Biomarkers for Sarcopenia.

Sarcopenia, the age-related decline in skeletal muscle mass and function, has become a recognized disease entity with serious health implications. However, traditional diagnostic methods relying on handgrip strength, walking speed, and dual energy X-ray absorptiometry-based muscle mass assessment are limited by subjective components, irregular measurement intervals, and inability to capture muscle quality. This paper comprehensively reviews how digital biomarker technologies including smart watch, smart insoles, surface electromyography, and markerless motion analysis via OpenPose enable objec

Brain cancer in silico modelling - the roadmap ahead.

Brain cancer comprises a diverse group of aggressive malignancies, often associated with a poor prognosis and a pressing need for more effective therapeutic strategies. The complexity of brain tumours arises from their highly dynamic and heterogeneous nature, involving intricate biological processes and interactions across multiple scales. Addressing these complexities requires an integrative approach that incorporates various levels of biological organisation to develop more accurate and predictive models of brain cancer. Cutting-edge techniques, including advanced imaging and omics technolog

Detecting Patterns of Intimate Partner Violence Using Qualitative Analyses and Machine Learning Algorithms.

Intimate partner violence (IPV) survivors increasingly use social media platforms to share their experiences and to seek help and support for their IPV-related concerns. IPV evidence extracted from social media platforms can provide valuable information and complement data obtained from conventional data sources (e.g., self-reports and interviews) thereby enhancing our understanding of IPV victimization. This study addressed three research questions: (1) What range of IPV behaviors emerge through qualitative coding? (2) To what extent do machine learning (ML) based text classifications yield r

Exposome factors predict the association between sleep health and depression.

BACKGROUND: Sleep health and depression are multidimensional constructs, yet their shared determinants remain obscure. We aim to identify the key exposome factors in predicting sleep-related depression (SRD). METHODS: We integrated regularized canonical correlation analysis with machine-learning models within a nested cross-validation to identify a shared phenotype (i.e., sleep-related depression (SRD)) and to predict SRD from exposome factors in the UK Biobank (n = 64,781). The best-performing model was validated in an independent subsample at baseline and follow-up (n = 8,139) and in a clin

Toddlers' Active Gaze Behavior Supports Self-Supervised Object Learning.

Toddlers learn to recognize objects from different viewpoints with almost no supervision. During this learning, they execute frequent eye and head movements that shape their visual experience. It is presently unclear if and how these behaviors contribute to toddlers' emerging object recognition abilities. To answer this question, we here combine head-mounted eye tracking during dyadic play with unsupervised machine learning. We approximate toddlers' central visual field experience by cropping image regions from a head-mounted camera centered on the current gaze location estimated via eye track

A Multimodal Fusion Model of Radiomics and Deep Learning Integrating the Tumor Microenvironment Accurately Predicts Pathological Complete Response in Breast Cancer.

BACKGROUND: Pathological complete response (pCR) to neoadjuvant chemotherapy (NAC) is a critical prognostic marker in breast cancer, yet its prediction remains challenging due to tumor heterogeneity and limitations of conventional imaging. While radiomics and deep learning (DL) have shown promise, prior studies often neglect the peritumoral microenvironment, a key determinant of therapeutic response. METHODS: We developed a multimodal model integrating intratumoral radiomics, peritumoral features (9-mm expansion), and DL-derived patterns from pre-NAC MRI. The model was trained and internally

Drivers and regional heterogeneity of cropland net ecosystem productivity across China's three major plains.

Net ecosystem productivity (NEP) in croplands is a core indicator of carbon exchange between agroecosystems and the atmosphere, directly reflecting net carbon budgets and sequestration capacity. To resolve its spatiotemporal patterns and dominant controls, we combined remote-sensing, land-cover, and meteorological datasets with the Boreal Ecosystem Productivity Simulator (BEPS) coupled to a Geostatistical Model of Soil Respiration (GSMSR) to simulate cropland NEP across China's three major plains-the Northeast China Plain (NCP), Huang-Huai-Hai Plain (HHHP), and Middle-Lower Yangtze Plain (MLYP

Beyond saponins: An integrated mass spectrometry strategy for profiling, spatial mapping, and rapid authentication of non-saponin constituents in Panax species.

Non-saponin constituents of Panax species, including amino acids, sugars, and nucleosides, have attracted increasing attention due to their nutritional relevance and potential health benefits in food-medicine homologous materials. However, their high polarity and chemical diversity pose significant challenges for comprehensive characterization, limiting their application in food quality evaluation and authenticity assessment. In this study, an integrated mass spectrometry-based workflow was developed to systematically profile water-soluble non-saponins in Panax ginseng, Panax notoginseng, and

Enhancing prostate cancer diagnosis: a machine learning-based biomarker approach.

BACKGROUND: The lack of reliable screening biomarkers for prostate cancer (PC) diagnosis makes tissue biopsy the gold-standard strategy. However, its frequent inconclusive results often lead to repeated procedures, increasing patient burden and healthcare costs. In this context, machine learning (ML) presents a novel approach for identifying gene signatures associated with tumor presence, offering a promising avenue for improved PC detection. Therefore, the present study explored the diagnostic potential of an ML-based gene signature and its applicability in tissue and plasma samples. METHODS:

"Rethinking High-KDPI Kidneys: A Multidomain Approach to Predicting Success".

BACKGROUND: The allocation and acceptance of deceased-donor kidneys in the United States is influenced by theKidney Donor Profile Index (KDPI). We conducted a national analysis of high-KDPI kidney transplants performed from 2014 to 2021 to identify key predictors of post-transplant outcomes beyond those incorporated in KDPI. METHODS: This retrospective cohort study used data extracted from the Scientific Registry of Transplant Recipients (SRTR). Adult, first-time recipients of kidney-only deceased-donor transplants with KDPI greater than 85% were included. Regression models were used to ident

Multi-horizon machine learning for population-level hypertension risk stratification in the UK Biobank.

AIMS: Hypertension is a major contributor to cardiovascular morbidity and mortality, yet identifying individuals at risk before clinical diagnosis remains challenging. Here, we present a multi-horizon machine learning framework designed to model incident hypertension risk across multiple clinically meaningful time windows using data from 246 286 participants in the UK Biobank. The framework systematically compares predictive performance across five horizons under severe class imbalance, enabling analysis of how discrimination, precision, and risk drivers evolve as outcome prevalence changes ov

Topological Data Analysis-Driven fNIRS Signal Processing for Alzheimer's Disease Stage Identification.

This paper proposes a novel Topological Data Analysis (TDA) pipeline to extract robust structural features from functional near-infrared spectroscopy (fNIRS) signals for the classification of Alzheimer's Disease (AD) stages. Alzheimer's disease is increasingly understood as a disconnection syndrome, where the disruption of functional brain networks precedes gross anatomical atrophy. However, traditional graph-theoretic approaches rely on arbitrary connectivity thresholds, which can obscure critical multi-scale topological information, and are sensitive to noise. To address this, our framework

Network analysis of serotonin CNVs shows biological convergence from genetic heterogeneity and discriminates between autism and developmental delay.

Differentiating autism spectrum disorder (ASD) from developmental delay (DD) is critical for guiding early intervention, but overlapping features and shared biological mechanisms pose challenges. This study investigates whether copy number variations (CNVs) affecting serotonergic genes carry sufficient information to distinguish between these neurodevelopmental disorders (NDDs). Using network mapping and machine learning, we applied gene ontology (GO) terms related to serotonergic systems to filter CNVs and construct networks modeling genetic and biological variation in ASD and DD. We identifi
