Genomic characterization of the first mpox and varicella-zoster co-infection in Pakistan (2023) through next-generation sequencing.

The global mpox outbreak spanning 2022-2023 has affected numerous countries worldwide. In this study, we present the first report on the detection, whole-genome sequence, and coinfection of the mpox virus and varicella zoster virus (VZV) from Pakistan. During April-May 2023, samples from 20 suspected cases of mpox were tested at the National Institutes of Health, Islamabad among which 4 tested positive. All four cases had a travel history of Saudi Arabia. All the suspected samples were processed by using a Zymo research kit for DNA extraction, followed by qRT-PCR amplification by using a DaAn

Genomic variability and immunological aspects involved in response to MPXV infection.

Mpox, caused by the monkeypox virus (MPXV), is a zoonotic disease that has gained global relevance after the 2022-2024 outbreak. MPXV exists in two distinct clades: clade I, associated with higher virulence and mortality, and clade II, which demonstrated increased human-to-human transmission and adaptation. Clinically, Mpox presents with rash, fever, and lymphadenopathy, with severe complications in immunocompromised individuals. Genomic surveillance has revealed rapid evolution, partly driven by APOBEC3-mediated mutagenesis, especially within immune-modulating regions. Although smallpox vacci

Multi-Body Biomarker Entrapment System: An All-Encompassing Tool for Ultrasensitive Disease Diagnosis and Epidemic Screening.

Ultrasensitive identification of biomarkers in biofluids is essential for the precise diagnosis of diseases. For the gold standard approaches, polymerase chain reaction and enzyme-linked immunosorbent assay, cumbersome operational steps hinder their point-of-care applications. Here, a bionic biomarker entrapment system (BioES) is implemented, which employs a multi-body Y-shaped tetrahedral DNA probe immobilized on carbon nanotube transistors. Clinical identification of endometriosis is successfully realized by detecting an estrogen receptor, ERβ, from the lesion tissue of endometriosis patient

Correlates of Mpox Vaccination among Sexual Minority Men in the United States: Sexual Behavior, Substance Use, and Main Partner Relationships.

Sexual minority men (SMM) have accounted for the majority of infections during the 2022 outbreak of the orthopox virus known as mpox (previously "monkeypox") in the US. This study examined correlates of mpox vaccination. Between July 28 and September 22, 2022, adult cisgender SMM (n = 2,620) not previously diagnosed with mpox responded to recruitment advertisements on social networking applications and completed an online survey. Of these, 730 (27.9%) received at least one vaccine dose. Logistic regression indicated sex with a casual partner was positively associated with vaccination. Stimulan

Monkeypox outbreak predominantly affecting men who have sex with men, Madrid, Spain, 26 April to 16 June 2022.

Up to 22 June 2022, 508 confirmed cases of monkeypox (MPX) have been reported in the Madrid region of Spain, 99% are men (n = 503) with a median age of 35 years (range: 18-67). In this ongoing outbreak, 427 cases (84.1%) reported condomless sex or sex with multiple partners within the 21 days before onset of symptoms, who were predominantly men who have sex with men (MSM) (n = 397; 93%). Both the location of the rash, mainly in the anogenital and perineal area, as well as the presence of inguinal lymphadenopathy suggest that close physical contact during sexual activity played a key role in tr

Evaluation of the efficacy of modified vaccinia Ankara (MVA)/IMVAMUNE against aerosolized rabbitpox virus in a rabbit model.

Infection of rabbits with aerosolized rabbitpox virus (RPXV) produces a disease similar to monkeypox and smallpox in humans and provides a valuable, informative model system to test medical countermeasures against orthopoxviruses. Due to the eradication of smallpox, the evaluation of the efficacy of new-generation smallpox vaccines depends on relevant well-developed animal studies for vaccine licensure. In this study, we tested the efficacy of IMVAMUNE [modified vaccinia Ankara-Bavarian Nordic (MVA-BN)] for protecting rabbits against aerosolized RPXV. Rabbits were vaccinated with either phosph

Mpox and sexually transmitted infections: syndemic drivers of transmission and disease outcomes-A Narrative Review.

The worldwide outbreak of mpox (formerly monkeypox) in 2022 emerged as a public health emergency posing greater threat to persons with sexually transmitted infections (STIs). This outbreak featured an increase in overlap between mpox and STIs reflecting evolving transmission dynamics in sexual networks. Epidemiological data reveal that STIs such as human immunodeficiency virus (HIV), Chlamydia trachomatis, herpes simplex virus, syphilis have been frequently associated with mpox infection highlighting the role of sexual contact in its transmission. A narrative review was conducted using PubMed

Multiomics profiling of plasma reveals lipid-immune dysregulation and exosome remodeling in mpox and mpox-HIV co-infection.

BACKGROUND: Monkeypox virus (MPXV) infects diverse human cell types, and human immunodeficiency virus (HIV) co-infection is common. The immunometabolic consequences of MPXV infection, and how it may be altered by HIV, remain poorly defined. METHODS: We performed quantitative plasma lipidomics and precise metabolomics in a discovery cohort (n = 81) comprising MPXV-monoinfected (MPLWOH), MPXV-HIV-coinfected (MPLWH), and HIV-monoinfected (PLWH) patients and healthy controls, integrating exosome proteomics, cytokine profiling, and transcriptomics of exosome-treated HepG2 and A549 cells for functi

Nonhuman primates as valuable models for mpox drug and vaccine discovery.

INTRODUCTION: In recent months, monkeypox (mpox) virus (MPXV) infections has grown to be a major worldwide concern. Cynomolgus monkeys, rhesus macaques, marmosets, and baboons are the nonhuman primate (NHP) models that provide the much needed means for developing new therapies against MPXV due to their genetic proximity to humans. AREA COVERED: In this review, the authors discuss epidemiology, transmission, clinical presentation, and the use of NHP in studying the treatment of MPXV over the past two decades on Google Scholar. NHP models have been widely used to evaluate the efficacy of antivi

Comprehensive update on the monkeypox outbreak.

Monkeypox (MPX) was first reported in 1970 in humans and outbreaks were restricted and highly localised to endemic regions of western and central Africa. However, after the first reported case in the UK in early May, 2022, the pattern of epidemic spreading in the geographical regions was much larger compared to past, posing a risk MPX might become entrenched beyond endemic areas. This virus is less transmissible than SARS-CoV-2, as it transmitted mainly through personal, close, often skin-to-skin contact with infectious MPX rash, body fluids, or scabs from an individual with MPX. Infections us

Salinomycin inhibits orthopoxvirus infection in vitro and in vivo by blocking endosomal acidification.

The recent global spread of monkeypox virus (MPXV) highlights the urgent need for effective antiviral therapies. Through high-throughput screening of an FDA-approved small molecule drug library, we identified salinomycin as a potent inhibitor of orthopoxvirus infection. Salinomycin showed nanomolar anti-MPXV activity in human keratinocytes (HaCaT) and lung epithelial cells (A549), with selectivity indices >100. Importantly, its efficacy was validated in primary human fibroblasts, reducing infectious viral titers by ∼2.6 log units (440-fold) versus controls. Mechanistic studies revealed salinom
