Rapid Generation of High-Affinity Rabbit Anti-Mouse IgG Monoclonal Antibodies by High-Throughput Single-B-Cell Sorting and Recombinant Expression.

Rabbit monoclonal antibodies (RabMAbs) are valuable for biomedical research and diagnostic applications because of their high affinity, specificity, and broad epitope recognition; here, we established an integrated phenotype-linked workflow for rapid RabMAb discovery using serum-derived polyclonal mouse IgG as a proof-of-concept model antigen. Three Big-Eared White rabbits were immunized in parallel, and the rabbit exhibiting the highest serum endpoint titer was selected for the complete downstream single-B-cell discovery workflow. Approximately 5 × 105 activated B cells were subjected to poly

Emerging non-invasive tools for hair follicle assessment: a narrative review on hyperspectral and terahertz imaging.

Hair loss, though often dismissed as cosmetic, reflects complex vascular, metabolic, structural, and inflammatory changes within the scalp microenvironment. Conventional diagnostic tools such as visual inspection, dermoscopy, trichoscopy, and biopsy remain limited in their ability to determine these evolving processes non-invasively. Recent advances in optical physics and biomedical engineering have introduced light- and wave-based imaging approaches that may expand scalp diagnostics beyond surface visualization. Hyperspectral imaging (HSI) and terahertz (THz) imaging have demonstrated compara

Neoantigen identification strategies enable personalized immunotherapy in refractory solid tumors.

BACKGROUND: Recent genomic and bioinformatic technological advances have made it possible to dissect the immune response to personalized neoantigens encoded by tumor-specific mutations. However, timely and efficient identification of neoantigens is still one of the major obstacles to using personalized neoantigen-based cancer immunotherapy. METHODS: Two different pipelines of neoantigens identification were established in this study: (1) Clinical grade targeted sequencing was performed in patients with refractory solid tumor, and mutant peptides with high variant allele frequency and predicte

Neuroprotection for ischaemic stroke: translation from the bench to the bedside.

Neuroprotection seeks to restrict injury to the brain parenchyma following an ischaemic insult by preventing salvageable neurons from dying. The concept of neuroprotection has shown promise in experimental studies, but has failed to translate into clinical success. Many reasons exist for this including the heterogeneity of human stroke and the lack of methodological agreement between preclinical and clinical studies. Even with the proposed Stroke Therapy Academic Industry Roundtable criteria for preclinical development of neuroprotective agents for stroke, we have still seen limited success in

CML mouse model in translational research.

Chronic myeloid leukemia (CML) is a myeloproliferative disorder characterized by increased proliferation of granulocytic cells without the loss of their capability to differentiate. CML is derived from the hematopoietic stem cells (1) with the Philadelphia chromosome resulting from of a reciprocal translocation between the chromosomes 9 and 22 t(9;22)-(q34;q11). This translocation produces a fusion gene known as BCR-ABL which acquires uncontrolled tyrosine kinase activity, constantly turning on its downstream signaling molecules/pathways, and promoting proliferation of leukemia cell through an

Clinical value of plasma pTau181 to predict Alzheimer's disease pathology in a large real-world cohort of a memory clinic.

BACKGROUND: The identification of patients with an elevated risk of developing Alzheimer's disease (AD) dementia and eligible for the disease-modifying treatments (DMTs) in the earliest stages is one of the greatest challenges in the clinical practice. Plasma biomarkers has the potential to predict these issues, but further research is still needed to translate them to clinical practice. Here we evaluated the clinical applicability of plasma pTau181 as a predictive marker of AD pathology in a large real-world cohort of a memory clinic. METHODS: Three independent cohorts (modelling [n = 991, 5

FlyDEGdb knowledge base on differentially expressed genes of Drosophila melanogaster, a model object in biomedicine.

Since the work of Nobel Prize winner Thomas Morgan in 1909, the fruit fly Drosophila melanogaster has been one of the most popular model animals in genetics. Research using this fly was honored with the Nobel Prize many times: in 1946 (Muller, X-ray mutagenesis), in 1995 (Lewis, Nüsslein-Volhard, Wieschaus, genetic control of embryogenesis), in 2004 (Axel and Buck, the olfactory system), in 2011 (Steinman, dendritic cells in adaptive immunity; Beutler and Hoffman, activation of innate immunity), and in 2017 (Hall, Rosbash and Young, the molecular mechanism of the circadian rhythm). A Targeted Computational Screen of the SWEETLEAD Database Reveals FDA-Approved Compounds with Anti-Dengue Viral Activity.

Affordable and effective antiviral therapies are needed worldwide, especially against agents such as dengue virus that are endemic in underserved regions. Many antiviral compounds have been studied in cultured cells but are unsuitable for clinical applications due to pharmacokinetic profiles, side effects, or inconsistent efficacy across dengue serotypes. Such tool compounds can, however, aid in identifying clinically useful treatments. Here, computational screening (Rapid Overlay of Chemical Structures) was used to identify entries in an in silico database of safe-in-human compounds (SWEETLEA

Crowd-funded micro-grants for genomics and "big data": an actionable idea connecting small (artisan) science, infrastructure science, and citizen philanthropy.

Biomedical science in the 21(st) century is embedded in, and draws from, a digital commons and "Big Data" created by high-throughput Omics technologies such as genomics. Classic Edisonian metaphors of science and scientists (i.e., "the lone genius" or other narrow definitions of expertise) are ill equipped to harness the vast promises of the 21(st) century digital commons. Moreover, in medicine and life sciences, experts often under-appreciate the important contributions made by citizen scholars and lead users of innovations to design innovative products and co-create new knowledge. We believe

From phthalates to microplastics: Emerging and unresolved biocompatibility risks in extracorporeal circulation.

BackgroundExtracorporeal circulation (ECC) is increasingly supported by biomaterials engineered to minimize chemical leaching, and many biomedical manufacturers have now removed or substantially reduced phthalate plasticizers particularly di(2-ethylhexyl) phthalate (DEHP) from tubing and circuit components. Nevertheless, emerging evidence highlights a parallel and underexplored concern: the generation of microplastic particles, especially during manual cutting and customization of circuit segments in the operating room. While screen and depth filters used in CPB and ECMO circuits efficiently r
