Eugenol in combination with lactic acid bacteria attenuates Listeria monocytogenes virulence in vitro and in invertebrate model Galleria mellonella.

Listeria monocytogenes is a human enteric pathogen that causes severe foodborne illness in high-risk populations. Crossing the intestinal barrier is the first critical step for Listeria monocytogenes infection. Therefore, reducing L. monocytogenes colonization and invasion of intestinal epithelium and production of virulence factors could potentially control listeriosis in humans. This study investigated the efficacy of sub-inhibitory concentration (SIC) of the plant-derived antimicrobial eugenol, either alone, or in combination with five lactic acid bacteria (LAB), namely Bifidobacterium bifi

Role of non-muscle myosin light chain kinase in neutrophil-mediated intestinal barrier dysfunction during thermal injury.

Neutrophils and non-muscle myosin light chain kinase (nmMLCK) have been implicated in intestinal microvascular leakage and mucosal hyperpermeability in inflammation and trauma. The aim of this study was to characterize the role of nmMLCK in neutrophil-dependent gut barrier dysfunction following thermal injury, a common form of trauma that typically induces inflammation in multiple organs. Histopathological examination of the small intestine in mice after a full-thickness burn revealed morphological evidence of mucosa inflammation characterized by neutrophil infiltration into the lamina propria

Treatment of atherosclerosis in apolipoprotein E-deficient mice with 4-(3-Bromobenzoyl)-6,7-dimethoxyquinazoline (WHI-P164), a potent inhibitor of triglyceride synthesis.

We identified a novel organic compound, 4-(3'-bromobenzoyl)-6,7-dimethoxyquinazoline (compound WHI-P164), as a potent inhibitor of triglyceride (TG) synthesis. In an in vitro model of lipid synthesis, WHI-P164 (but not any one of the three structurally similar control dimethoxyquinazoline compounds) inhibited the accumulation of TG-rich intracellular lipid droplets in Caco-2 human intestinal cells in a concentration-dependent fashion. WHI-P164 caused no acute toxicity associated with morbidity or mortality in mice when administered at dose levels ranging from 0.5 to 80 mg/kg. In pharmacokineti

Design and Synthesis of Novel Imidazole Derivatives Possessing Triazole Pharmacophore with Potent Anticancer Activity, and In Silico ADMET with GSK-3β Molecular Docking Investigations.

A library of novel imidazole-1,2,3-triazole hybrids were designed and synthesized based on the hybrid pharmacophore approach. Therefore, copper(I)catalyzed click reaction of thiopropargylated-imidazole 2 with several organoazides yielded two sets of imidazole-1,2,3-triazole hybrids carrying different un/functionalized alkyl/aryl side chains 4a-k and 6a-e. After full spectroscopic characterization using different spectral techniques (IR, 1H, 13C NMR) and elemental analyses, the resulted adducts were screened for their anticancer activity against four cancer cell lines (Caco-2, HCT-116, HeLa, an

Synthesis and evaluation of a novel β-cyclodextrin derivative for oral insulin delivery and absorption.

In this paper, we describe the synthesis and testing of a novel β-cyclodextrin derivative, carboxymethy-hydroxypropyl-β-cyclodextrin (CM-HP-β-CD). After synthesis, we verified that carboxymethyl and hydroxypropyl groups were successfully substituted onto β-cyclodextrin to form CM-HP-β-CD. Then, we performed in vitro experiments to investigate (1) the ability of CM-HP-β-CD to bind insulin, (2) the transportation of insulin-CM-HP-β-CD complexes across a Caco-2 cell monolayer, and (3) the cytotoxicity of CM-HP-β-CD. CM-HP-β-CD showed the strongest insulin-binding ability and the best transport pr

Plant-based beverages supplemented with Se-microparticles: a comprehensive study of absorption and gastrointestinal simulation using static and dynamic in vitro digestion models.

This study evaluated and compared the physicochemical properties, antioxidant capacity, and Selenium (Se) bioaccessibility and intestinal absorption (bioavailability estimate) of almond, rice, and soy beverages fortified with microencapsulated organic (Selenium-L-cystine) or inorganic (sodium selenate) Se. These were compared against a traditional homemade Brazil nut beverage, naturally rich in this mineral. Supplementation produced matrix-dependent effects, notably slight reductions in L* (lightness) and consistent increases in b* (yellowness). After in vitro digestion, antioxidant activity a

The conserved human astrovirus-receptor interface reveals a targetable vulnerability for antiviral development.

Human astrovirus (HAstV) is a major cause of viral enteritis in children and the older adults worldwide. The eight serotypes of classical HAstVs (HAstV1-8), employ the neonatal Fc receptor (FcRn) as an entry receptor. To elucidate the receptor binding mechanism of HAstVs, we determined the crystal structure of the HAstV8 spike protein complexed with human FcRn. The structure reveals that the HAstV8 spike protein engaged FcRn via a conserved surface depression, which is also present across the other seven classical HAstVs serotypes. The binding interface of HAstV8 spike protein on FcRn largely

Crude proline modulates the microbiota-gut-brain axis by attenuating curli-associated α-synuclein accumulation and regulating Wnt/β-catenin signaling in Parkinson's disease.

This study investigated whether crude proline (CP), a metabolite fraction derived from heat-killed Enterococcus faecium (E. faecium) FBL1 isolated from makgeolli, is associated with the modulation of microbiota-host interactions within the microbiota-gut-brain axis and attenuation of PD-related pathology. CP was purified using fast protein liquid chromatography and liquid chromatography-mass spectrometry. Its biological effects were evaluated in SH-SY5Y and Caco-2 cell models and in an MPTP-induced mouse model of PD, followed by behavioral, microbiota, and molecular analyses. CP treatment sign

Assessing the effects of tofacitinib on the gut microbiome in inflammatory bowel disease.

Gut microbiota dysbiosis and impaired epithelial barrier function play a key role in inflammatory bowel disease (IBD). Tofacitinib citrate, a Janus kinase (JAK) inhibitor approved for IBD, modulates immune responses via the JAK-STAT (Janus kinase-signal transducer and activator of transcription) pathway, yet its effects on the gut microbiome remain unclear. Here, we employed the short-term colon model (ProDigest, BE) containing human microbiota from three Crohn's Disease donors to assess fermentative and metabolic activities and microbial composition following 48 h of tofacitinib treatment. Pickering double emulsions stabilized by acylated cellulose nanocrystals for oral co-delivery of macromolecules and permeation enhancers.

Double emulsions are potential oral delivery systems for the simultaneous administration of hydrophilic drugs and hydrophobic permeation enhancers to enable effective intestinal absorption of macromolecular drugs. Emulsions stabilized by solid particles, i.e., Pickering stabilizers, have shown potential to form gastric-stable emulsions that can protect their cargo from release under gastric conditions. Here, we use acylated cellulose nanocrystals to facilitate the formation of stable double emulsions for intestinal drug delivery. Water-in-oil-in-water double emulsions were obtained by a two-st

Exploring protein stabilized multiple emulsion with permeation enhancer for oral delivery of insulin.

In present study, we have developed W/O/W microemulsion (ME) containing piperine (PiP) as a permeation enhancer and albumin (Alb) serving as a stabilizer for oral delivery of insulin (INS). The resultant formulation, ME(INS)-PiP-Alb exhibited droplet size of 3.35 ± 0.25 μm along with polydispersity index (PDI) of 0.30 ± 0.10. The formulation process employed for developing ME(INS)-PiP-Alb showed no effect on INS's chemical and conformational stability. Further, ME(INS)-PiP-Alb was able to maintain desired attributes (size & PDI) along with INS stability in simulated gastrointestinal fluids. Al

The stimulation of GLP-1 secretion and delivery of GLP-1 agonists via nanostructured lipid carriers.

Nanoparticulate based drug delivery systems have been extensively studied to efficiently encapsulate and deliver peptides orally. However, most of the existing data mainly focus on the nanoparticles as a drug carrier, but the ability of nanoparticles having a biological effect has not been exploited. Herein, we hypothesize that nanostructured lipid carriers (NLCs) could activate the endogenous glucagon-like peptide-1 (GLP-1) secretion and also act as oral delivery systems for GLP-1 analogs (exenatide and liraglutide). NLCs effectively encapsulated the peptides, the majority of which were only

Beneficial Effects of the Peroxisome Proliferator-Activated Receptor α/γ Agonist Aleglitazar on Progressive Hepatic and Splanchnic Abnormalities in Cirrhotic Rats with Portal Hypertension.

Recent studies have reported that peroxisome proliferator-activated receptor α (PPARα) agonist decreases intrahepatic resistance, whereas PPARγ agonist reduces portosystemic shunts (PSSs) and splanchnic angiogenesis in cirrhotic rats. The present study investigated the effects of a 21-day treatment with the dual PPARα/γ agonist aleglitazar (Ale) on progressive abnormalities in bile-duct-ligated and thioacetamide-induced cirrhotic rats with portal hypertension (PH). In vivo and in vitro effects were evaluated. Chronic Ale treatment significantly up-regulated PPARα/PPARγ receptors and down-regul

Polymeric microcontainers improve oral bioavailability of furosemide.

Microcontainers with an inner diameter of 223 μm are fabricated using the polymer SU-8, and evaluated in vitro, in situ and in vivo for their application as an advanced oral drug delivery system for the poorly water soluble drug furosemide. An amorphous sodium salt of furosemide (ASSF) is filled into the microcontainers followed by applying a lid using Eudragit L100. It is possible to control the drug release in vitro, and in vitro absorption studies show that the microcontainers are not a hindrance for absorption of ASSF. In situ perfusion studies in rats are performed with ASSF-filled microc

In Vitro Functional Analyses of Infrequent Nucleotide Variants in the Lactase Enhancer Reveal Different Molecular Routes to Increased Lactase Promoter Activity and Lactase Persistence.

The genetic trait that allows intestinal lactase to persist into adulthood in some 35% of humans worldwide operates at the level of transcription, the effect being caused by cis-acting nucleotide changes upstream of the lactase gene (LCT). A single nucleotide substitution, -13910 C>T, the first causal variant to be identified, accounts for lactase persistence over most of Europe. Located in a region shown to have enhancer function in vitro, it causes increased activity of the LCT promoter in Caco-2 cells, and altered transcription factor binding. Three other variants in close proximity, -13907…
